即ち、本発明は、(a)葛花処理物、(b)甘藷茎葉末もしくは抽出物、(c)ケール末もしくは抽出物、(d)大麦葉粉砕物もしくは抽出物、及び(e)ジャガイモ抽出物から選ばれる少なくとも一種を主成分として含有する抗糖化用組成物を提供する。
本発明の抗糖化用組成物は、(a)葛花処理物、(b)甘藷茎葉末もしくは抽出物、(c)ケール末もしくは抽出物、(d)大麦葉末もしくは抽出物、及び(e)ジャガイモ抽出物から選ばれる少なくとも一種を含有する。
本発明において葛花処理物とは、葛の花の処理物をいう。葛の種類としては、特に制限はないが、プエラリア・トムソニイ(Pueraria thomsonii)、プエラリア・ロバータ(Pueraria lobata)、プエラリア・スンバーギアナ(Pueraria thunbergiana)等を例示でき、プエラリア・トムソニイ(Pueraria thomsonii)を用いることが好ましい。
本発明において甘藷茎葉末は、甘藷茎葉(即ち、甘藷の茎及び/又は葉)を乾燥し粉砕したもの、又は甘藷茎葉の搾汁液を乾燥し粉末化したものであり、甘藷茎葉抽出物は、甘藷茎葉又はその乾燥物から得られる抽出物である。
本発明においてケール末は、アブラナ科植物であるケールの葉及び/又は茎を乾燥し粉砕したもの、又はケールのスラリー又は搾汁液を乾燥し粉末化したものであり、ケール抽出物は、ケール又はその乾燥物から得られた抽出物である。
また、本発明においては、ケールの葉部および茎部のいずれの部分も適用することができる。
大麦葉末は、イネ科植物である大麦(Hordeum vulgare L.)の葉を乾燥し粉砕したもの、又は大麦の葉の搾汁液を乾燥し、粉末化したものであり、大麦葉抽出物は、大麦の葉又はその乾燥物から得られた抽出物である。
また、大麦葉としては、分べつ開始期から出穂開始期(背丈が20〜40cm程度)に収穫した若葉を使用することが好ましい。
ジャガイモ抽出物は、ジャガイモの塊茎から得られる抽出物である。ジャガイモの品種としては、例えば男爵薯、メークイン、キタアカリ、とうや、トヨシロ、インカのめざめ、デジマ、十勝こがねなどを用いることができる。
乾燥方法としては賦型剤を添加しスプレードライなどにより乾燥する方法であってもよい。
溶媒抽出は、一般に70〜90℃にて10〜60分、好ましくは75〜85℃にて10〜20分で行われる。
実施例1の被験物質として、葛花処理物(東洋新薬社製「葛の花エキス」、イソフラボン含量20%)を用意し、また、比較例1の被験物質として大豆抽出物(イソフラボン含量40%)を用意し、参考例の被験物質(ポジジティブコントロール)として、代表的な抗糖化剤であるアミノグアニジン(アミノグアニジン塩酸塩、Cayman Chemical Co.)を用意した。

結果を表2に示す。
実施例2の被験物質としてジャガイモ抽出物(東洋新薬社製「ポテイン」)を用意し、参考例の被験物質として実施例1と同様にアミノグアニジンを用意した。
実施例1と同様にして、被験物質が表3に示す濃度の被験物質溶液を調製し、それを用いて、被験物質が同表の濃度となる試験溶液を調製し、それをインキュベートし、AGEs生成阻害率(%)を算出した。結果を表3に示す。
表3から、ジャガイモ抽出物が抗糖化作用を有することがわかる。
実施例3、4、5の被験物質として、甘藷茎葉末(東洋新薬社製すいおう(緑色粉末))、大麦葉末(東洋新薬社製大麦若葉(緑色粉末))、ケール末(東洋新薬社製ケール(緑色粉末))を用意し、参考例の被験物質として実施例1と同様にアミノグアニジンを用意した。
結果を表4〜表6に示す。
実施例3、5の被験物質を1:1で混合したものを使用し、実施例1と同様にして表6に示す濃度の被験物質溶液を調製し、それを用いて、被験物質が同表の濃度となる試験溶液を調製し、それをインキュベートし、AGEs生成阻害率(%)を算出した。結果を表7に示す。
表7から、甘藷茎葉末とケール末をそれぞれ単独で使用するよりも、組み合わせて使用すると抗糖化作用が強まることがわかる。
実施例3、4の被験物質を1:1で混合したものを使用し、実施例1と同様にして表8に示す濃度の被験物質溶液を調製し、それを用いて、被験物質が同表の濃度となる試験溶液を調製し、それをインキュベートし、AGEs生成阻害率(%)を算出した。結果を表8に示す。
表8から、甘藷茎葉末と大麦葉末をそれぞれ単独で使用するよりも、組み合わせて使用すると、抗糖化作用が強まることがわかる。
実施例4、5の被験物質を1:1で混合したものを使用し、実施例1と同様にして表8に示す濃度の被験物質溶液を調製し、それを用いて、被験物質が同表の濃度となる試験溶液を調製し、それをインキュベートし、AGEs生成阻害率(%)を算出した。結果を表9に示す。
表9から、ケール末と大麦葉末をそれぞれ単独で使用するよりも、組み合わせて使用すると、抗糖化作用が強まることがわかる。

- 甘藷茎葉末もしくは甘藷茎葉抽出物のいずれか1種、及び大麦葉末もしくは大麦葉抽出物のいずれか1種を含有することを特徴とする、抗糖化用組成物。
