Claims (3)
- 타겟 서열이 존재하는 데옥시리보 핵산(DNA)으로 부터 시작하여 리보핵산(RNA)를 합성하는 방법으로서, 하나 이상의 제한효소로 상기 DNA를 처리한후, 수득한 DNA단편물을 분리하고, 그에 의해 단일가닥 DNA가 형성되며, 그후 타겟 서열의 일부에 대응하는 뉴클레오티드 서열에 결합하는 프로모터 서열을 함유한 하나의 핵산 프라이머를 적당한 조건하에 대응하는 단일가닥 DNA에 하이브리드 시키며, 그후, 하이브리드 형태로 수득한 2개의 핵산 서열을 DNA폴리머라제에 의해 3'-말단으로 부터 연장시켜서 이중가닥 DNA를 생성하며, 이것을 RNA합성을 위해 RNA폴리머라제에 의해 매트릭스로 사용하는 것을 특징으로 하는 방법.
- 제1항에 있어서, 합성된 RNA를 변성 조건하에 겔 전기영동과 블롯팅으로 검출한후 라벨된 특이 올리고뉴클레오티드 서열과 하이브리드 반응을 시키는 것을 특징으로 하는 방법.
