Porphyromonas gingivalis(以下、P.gingivalis)を、アネロパック・ケンキ(三菱化学株式会社製)および角型ジャー(三菱化学株式会社製)を用いて嫌気環境にし、高圧蒸気滅菌した変法GAMブイヨン(日水製薬株式会社製)で37℃で培養した。
P.gingivalisをダルベッコリン酸緩衝生理食塩水(ナカライテスク株式会社製)で希釈し、濁度が0.5McFarlamdとなるように調製した。濁度の指標には、マクファーランドスタンダード(シスメックス・ビオメリュー株式会社製)を用いた。

Porphyromonas gingivalis (hereinafter referred to as P. gingivalis) was cultured at 37°C in modified GAM bouillon (manufactured by Nissui Pharmaceutical Co., Ltd.) that had been sterilized by high-pressure steam, in an anaerobic environment created using Anaeropack Kenki (manufactured by Mitsubishi Chemical Corporation) and a square jar (manufactured by Mitsubishi Chemical Corporation).
P. gingivalis was diluted with Dulbecco's phosphate buffered saline (Nacalai Tesque, Inc.) to adjust the turbidity to 0.5 McFarland standard (Sysmex bioMérieux, Inc.) was used as an index of turbidity.

[副素材]

菩提樹(Tilia cordata Mill.)は、市販されている花のエキス末を用いた。
葛花(Pueraria thomsonii)は、熱水抽出した花のエキス末(東洋新薬社)を用いた。
大麦若葉は、粉砕末(東洋新薬社)を含水エタノールで抽出したエキス末を用いた。
甘藷若葉は、粉砕末(東洋新薬社)を含水エタノールで抽出したエキス末を用いた。
[Secondary material]
For Coffee robusta, commercially available extract powder of coffee seeds (green coffee beans) was used.
For linden tree (Tilia cordata Mill.), a commercially available flower extract powder was used.
For kudzu flowers (Pueraria thomsonii), a powdered extract of the flowers extracted with hot water (Toyo Shinyaku Co., Ltd.) was used.
For the barley leaves, an extract powder was used, which was obtained by extracting crushed barley leaves (Toyo Shinyaku Co., Ltd.) with aqueous ethanol.
For the young sweet potato leaves, an extract powder was used, which was obtained by extracting crushed powder (Toyo Shinyaku Co., Ltd.) with aqueous ethanol.

図1~5に示すように、松樹皮抽出物と特定の植物素材を組み合わせることにより、P.gingivalisの増殖が効果的に抑制されることがわかる。特に、コーヒーノキ、菩提樹、大麦若葉及び甘藷若葉は単独では効果が弱い若しくはないにもかかわらず、松樹皮抽出物と組み合わせることで、P.gingivalisに対する増殖抑制作用が効果的に高まった。葛花はそのものにも増殖抑制作用が見られたが、松樹皮抽出物と組み合わせることでより効果が高まった。よって、本発明の経口組成物は、P.gingivalisの増殖を効果的に抑制することで、歯周病を予防及び/又は改善し、また、歯周病に特有な口臭を予防及び/又は改善する、優れたオーラルケア用経口組成物であることがわかる。 As shown in Figures 1 to 5, it can be seen that the growth of P. gingivalis is effectively inhibited by combining pine bark extract with specific plant materials. からなる混合物を打錠し、500mgのチュアブルタブレット2粒を製造した。
