Description
臨床慣例において、血液は分析される試料の最も重要な供給源である。全血は得られる最初の試料であるが、全血試料は通常、簡便な試料取り扱いまたは信頼性のある分析物検出を可能にするためにさらに処理されなければならない。
第一の態様において、本発明は、全血試料を回収および処理するためのサンプリングチューブであって、該サンプリングチューブは該全血試料の特異的溶血(differential hemolysis)のための試薬を含み、該特異的溶血のための試薬は特異的溶血のための化学物質および抗凝固剤を含み、すぐ使用できる(ready-to-use)および使い捨て(single-use)サンプリングチューブである、サンプリングチューブに関する。
好ましい態様において、本発明は、全血試料を回収および処理するためのサンプリングチューブであって、該サンプリングチューブは該全血試料の特異的溶血のための試薬を含み、該特異的溶血のための試薬は特異的溶血のための化学物質および抗凝固剤を含み、すぐ使用できるおよび使い捨てサンプリングチューブである、サンプリングチューブに関する。

から選択され、アニオンが好ましくはクロライド、テトラフルオロボレート、オクチルスルフェート、ヨージドおよびチオシアネートから選択される塩である。
1.正常相クロマトグラフィーは非極性(分散性)移動相と共に極性固定相の使用を必要とする。
2.反対の可能性である、逆相クロマトグラフィーは、非極性固定相および極性移動相(溶媒の水、メタノール、アセトニトリルおよびテトラヒドロフランの1つ以上から構成される)の使用を必要とする。
3.イオン交換クロマトグラフィーはイオン性相互作用を伴う。この場合において、移動相はイオン性溶質の溶解性を確実にするためにイオン化を支持しなければならない。固定相はまた、ある保持を促進するために部分的にイオン性でなければならない。結果的に、固定相を用いた相互作用は、強力であり、これは通常、長い分析時間および広いピークに反映される。
4.サイズ排除クロマトグラフィーは分子サイズのみに基づいた分離を伴い、理想的に溶質の固定相との強力な相互作用がないことを必要とする。
5.アフィニティークロマトグラフィーは、例えば抗原および対応する抗体またはレセプターおよび対応するリガンドのような、特異的結合対の一員の間の特定の相互作用に基づいている。例えば、結合対の第一パートナーは適切な固定相に結合され、結合対の第二のパートナーを捕捉するために使用される。第二のパートナーは、適切な手段によって放出でき、単離できる。
様々な候補溶血試薬の評価
実施例1.1 溶血の視覚評価
溶液A:新鮮EDTA安定化全血を、0.15モル塩化ナトリウム溶液で1:10の比に希釈する(50μL EDTA血液+450μL 塩化ナトリウム溶液)。
上述の表から明らかなように、視覚評価によって特異的溶血のための良好な候補試薬が視覚で同定することができる。
溶液A:新鮮EDTA安定化全血を、0.15モル塩化ナトリウム溶液で1:10の比に希釈する(50μL EDTA血液+450μL 塩化ナトリウム溶液)。
溶血物の混合後に、滴を顕微鏡スライドに塗り、室温で空気乾燥し、メイ-グルンワルド染色試薬(Merck Cat. No. 1.01424 May-Grunwald's Eosin Methylene Blue Solution)で染色する。メイ-グルンワルド染色後に、核は様々な彩度の紫色に染色し、細胞質は青色から淡いピンク色の色調で見られ、鮮やかな赤みがかった色からライラック色の顆粒がいくつかの細胞種類の細胞質に存在することがあり、好塩基球は細胞質で濃い青黒色顆粒を示し、好中球は、細胞質で明るいオレンジ色顆粒を示し、赤血球はピンク色からオレンジ色に染色される。
処理全血試料をトリパンブルー溶液(Merck cat. no 1.11732; Trypanblau C.I. 23850)と(1:1)混合し、顕微鏡検査のためのNeugebauer-chamberに分配する。顕微鏡検査は、油浸光学顕微鏡(倍率X630倍)によって実施される。
HPLCによる様々な候補溶血試薬の評価
溶解効率を評価するために、実施例1によって調製された溶血された全血試料をHPLC系に注入し、系の背圧をモニターする。
溶解効率は、50の10μLの処理全血試料を、床物質として5μm Symmetry C18粒子、2mmの内部カラム直径、20mmのカラム長および0.5μmの孔径を有するフリットを有するHPLCカラムに適用することによって評価される。溶出は、5分以内の0.1%蟻酸を有する水〜0.1%蟻酸を有するアセトニトリルの勾配、および0.2mL/分の流速である。50インジェクションに対する観察された背圧の増加は、20 bar未満である。
特異的溶血のための試薬を含むチューブにピペッティングして優しく混合物を振とうすることによるEDTA抗凝固全血の処理
3.1 1-ブチル-4-メチルピリジニウムテトラフルオロボレート
溶解試薬を、11.25mLチオシアネートカリウム溶液(0.1モル;即ち9.72g KSCNが1L蒸留水に溶解される)を12.5mL 1-ブチル-4-メチルピリジニウムテトラフルオロボレート(BMPBF4)および23.7mL塩化ナトリウム(水中0.15モル)と混合することによって調製する。従って、特異的溶血のためのこの試薬は約25%BMPBF4の濃度を有する。チオシアネートカリウムがかなり低いモル濃度で存在するので、観察される効果に有意にはほとんど寄与しないようである。
この溶解試薬を、23.7mLチオシアネートカリウム溶液(0.2モル;即ち9.72g KSCNが0.5L蒸留水に溶解される)を1000mg 1-メチル-1-オクチルピロリジニウムクロライド(Me-octPCI)および23.7mL塩化ナトリウム(水中0.15モル)と混合することによって調製する。従って、特異的溶血のためのこの試薬は、約2%Me-octPClの濃度を有する。
実施例3.2と比較してこの濃縮溶解試薬を、470μLチオシアネートカリウム溶液(1モル)を300μL塩化ナトリウム溶液(水中の0.15モル)および110mgの1-メチル-1-オクチルピロリジニウムクロライドと混合することによって調製する。特異的溶血のためのこの試薬は、約15%Me-octPCIの濃度を有する。
この溶解試薬を、80μL水と50μL塩化ナトリウム溶液(水中0.15モル)および20mg 3-カルバモイル-1-オクチルオキシメチルピリジニウムクロライドと混合することによって調製する。特異的溶血のためのこの試薬は、約10%COMPClの濃度を有する。
Claims (6)
- 全血試料を回収および処理するためのサンプリングチューブであって、該サンプリングチューブは該全血試料の特異的溶血のための試薬を含み、該特異的溶血のための試薬は塩および抗凝固剤を含み、該塩は、カチオン
(式中、mは0または1であり、nは4または6である)
の1つ以上、ならびにクロライド、テトラフルオロボレート、オクチルスルフェート、ヨージドおよび/またはチオシアネートから選択されるアニオンの1つ以上からなる塩から選択され、該サンプリングチューブは、すぐ使用でき、かつ使い捨てのサンプリングチューブであり、10μLの溶血物の50のアリコートを、2mmの直径および0.5μmの孔径を有するHPLCシステムのフィルターに適用しても該フィルターをふさがない場合に、該特異的溶血が起こっており、ここで、該溶血物は、生理学的食塩水中に1:10で希釈され、該特異的溶血のための試薬と1:1で混合された血液試料である、サンプリングチューブ。 - 該塩が、KBr、KIおよびKSCNから選択される塩をさらに含む、請求項1記載のサンプリングチューブ。
- 前記特異的溶血のための試薬が赤血球の細胞膜の溶解を生じ、同時に試料成分の沈殿を生じない、請求項1または2記載のサンプリングチューブ。
- 前記特異的溶血のための試薬がプロテアーゼインヒビターをさらに含む、請求項1〜3いずれかに記載のサンプリングチューブ。
- 液体クロマトグラフィーのための全血試料の処理における請求項1〜4いずれかに記載のサンプリングチューブの使用。
- 液体クロマトグラフィー系分析における、請求項1〜4いずれかに記載のサンプリングチューブ内での特異的溶血によって得られた処理血液試料の使用。
