Abstract
酸素感受性の非経口投与薬剤の貯蔵性を向上させる柔軟な透明容器を提供する。
【解決手段】
容器は、外被を密封した後に残留した本質的に全ての酸素を消費する能力があり、さらに十分な期間、前記外被の中に侵入した酸素を消費する能力がある酸素吸収剤とともに実質的に酸素不透過性の外被の中に、封入された一次内容器を含む。この内容器は、ポリプロピレンを含み、親脂性薬剤との間に適合性を有し、永久シールおよび剥離可能シールの両方を形成することができる柔軟なポリマー材料から作られており、一方、外被は、実質的に不透水性で、酸素障壁形成能力を有する外側の第一のポリマーフィルム、およびこれに組み付けられた補助的酸素障壁形成能力を有する内側の第二のポリマーフィルムを含む実質的に不透水性の柔軟な多層ポリマー材料から作られている。この容器は、蒸気滅菌または放射線滅菌を実施した後もその特性を本質的に維持する。

Description
a)ポリエチレン/ポリプロピレンコポリマー(FINA Dypro Z 9450)とShell社のKraton(登録商標)G1652(スチレン/エチレン/ブタジエン/スチレン(SEBS)コポリマー)の混合物から成り、医療用流体と接する内側の密封材層、
b)純粋なKraton(登録商標)G1652の中間結合層、および
c)熱可塑性の脂環式コポリエステル(コポリ(エステルエーテル)シクロヘキサンジメタノールの1,4−ジメチル−シクロヘキサンジカルボキシレートのトランス異性体とヒドロキシル基末端のポリテトラメチレングリコールとの縮合産物)であるEastman Chemical社のEcdel(登録商標)9965(または9566ないし9967)の外部剥離層である。
この例には、商標名Oxnil(Pharmacia & Upjohn AB)が与えられたPET−酸化アルミニウム/接着剤/PP/EVOH/PPの層からできた周囲の外被の中に酸素吸収剤(Mitsubishi Gas社のAgeless FX100)とともに封入された500mlのExcel(登録商標)ポリマー内容器中のIntralipid(登録商標)20%の安定性を示す。500mlガラスびんに入れたIntralipid(登録商標)20%を参照として使用した。
充填した内容器の外被として選択した材料の酸素障壁形成能力を調べた。
この例の目的は、本発明に基づく容器に、+5℃および+25℃で12カ月間貯蔵した1バッチの脂質エマルジョンを、安全に投与できるTPN溶液に混合できるか否かを調べることにある。
Excel(登録商標)製の三チャンバ内容器に充填し、蒸気滅菌したIntralipid(登録商標)20%(Pharmacia社の20%大豆油脂質エマルジョン)の混合特性を、ガラスびん中で加熱滅菌し貯蔵したIntralipid(登録商標)20%のそれと比較した。それぞれの三チャンバ容器は、充填直前にろ過した窒素で二回パージし、無菌ではないIntralipid(登録商標)500mlを、ガラスびんから中央の区画に移した。その他の区画には、注射用の水614mlおよび1193mlをそれぞれ充填した。充填、密封した容器は、サドル形ポートシステムの出口ポートと入口ポートの間に置いた酸素吸収剤とともに、前記例で述べたPET−金属酸化物/接着剤/PP/EVOH/PP製の外被の中に置いた。外被を密封する前に、Multivac中で脱気し窒素を外被内に注入して滅菌に適当なガス量とし、その後、密封した。その後この容器を、121.1℃で17〜20分間オートクレーブ処理した。参照用のガラスびんを、通常の製造プロセスに従って121.1℃で12分間オートクレーブ処理した。混合は、無菌条件下で、三チャンバ容器で実施されるのと同じ順番で実施した。17.2%グルコース溶液を窒素保護の下で混合容器に移し、その後、前述のとおりに処理した脂質エマルジョン(Intralipid(登録商標)20%)を追加し、静かに振とうした後、アミノ酸溶液(電解質を含むVamin(登録商標)18)を混合し、撹拌した。この混合物を、窒素保護の下で無菌注入びんに入れた。びんを密封した後、これらに、周囲温度(約25℃)で2日間の貯蔵、または5℃で6日間、次いで25℃で2日間の貯蔵を行った。
Claims (48)
- 酸素感受性の非経口投与薬剤の貯蔵性を向上させる柔軟な透明容器であって、外被を密封した後に残留した本質的に全ての酸素を消費する能力があり、さらに十分な期間、前記外被の中に侵入した酸素を消費する能力がある酸素吸収剤とともに実質的に酸素不透過性の外被の中に封入された一次内容器を含む容器において、
(i)内容器が、ポリプロピレンを含み、親脂性薬剤との間に適合性を有し、永久シールおよび剥離可能シールの両方を形成することができる柔軟なポリマー材料から作られており、
(ii)外被が、
a)実質的に不透水性で、酸素障壁形成能力を有する外側の第一のポリマーフィルムと、
b)これに組み付けられた、補助的酸素障壁形成能力を有する内側の第二のポリマーフィルムと
を含む実質的に不透水性の柔軟な多層ポリマー材料から作られており、
容器が、蒸気滅菌または放射線滅菌を実施した後もその特性を本質的に維持することを特徴とする容器。 - 一次内容器が、チャンバの内容を混合し、前記内容器の底部の流体出口を通してこれを患者に投与したいときに、外側から手で破断させることができる一つまたは複数の非漏出性剥離可能シールによって二つ以上のチャンバに分割されていることを特徴とする請求項1に記載の容器。
- 一次内容器が、その外面に指定されたより高い融点を有する領域、およびその密封内面に指定されたより低い融点を有する領域を有する柔軟なフィルムから作られていることを特徴とする請求項1または請求項2に記載の容器。
- より低い融点を有する前記内側の領域が、異なる溶着条件を与えたときに、永久シールおよび剥離可能シールの両方を形成する能力を有することを特徴とする請求項3に記載の容器。
- 一次内容器が、少なくとも二つの層を有するフィルムから作られており、少なくとも内側の密封材層が、ポリプロピレンを含み、高温で溶着したときに永久シールを、低温で溶着したときに剥離可能シールを形成する能力を有することを特徴とする請求項3または請求項4に記載の容器。

- 充填手順中に密封接続を提供するために充填装置の充填ノズルと形状を一致させた少なくとも一つの充填オリフィスが、内容器の側面シームに形成される請求項40に記載の方法。
- 一時的な充填ポートまたは充填オリフィスを有する内容器の側面を充填後に切り離し、続いてバッグの側面を永久シームで再溶着する請求項43から請求項45に記載の方法。
- 永久溶着よりも低い温度および/または圧力で少なくとも一つの剥離可能密封シームを溶着することによって、少なくとも二つのチャンバを形成することを特徴とする請求項39から請求項46のいずれか一項に記載の方法。
- 蒸気で容器を滅菌することを特徴とする請求項39から請求項47のいずれか一項に記載の方法。
