Abstract
本发明提供含有原花色素作为有效成分、不改变总消耗热量、使脂质代谢亢进的新型脂肪燃烧促进剂。

技术领域
本发明涉及对维持健康有益的脂肪燃烧促进剂、体脂肪减少剂、脂肪蓄积防止剂以及用于促进脂肪燃烧、用于减少体脂肪或用于防止脂肪蓄积的饮食品。
背景技术
原花色素是植物中含有的多酚之一,已知其具有抗氧化作用等各种活性(专利文献1、非专利文献1以及非专利文献2)。关于原花色素的作用,已知有超氧自由基清除活性、与具有抗氧化作用的维生素C的协同作用(非专利文献1)。此外,作为临床作用,已知其对慢性胰腺炎有治疗效果(非专利文献1以及非专利文献2)。

发明内容
本发明的目的在于提供不改变总消耗热量、使脂质代谢亢进的新型脂肪燃烧促进剂、体脂肪减少剂、脂肪蓄积防止剂以及用于促进脂肪燃烧、用于减少体脂肪或用于防止脂肪蓄积的饮食品。
本发明者等为了实现上述目的,潜心研究,结果发现:将给与原花色素与不给与原花色素的情况进行比较,两者之间的总消耗热量并无差别,但给与原花色素时,脂质代谢亢进,即脂质被更高效地消耗,由此得知原花色素可作为有效的脂肪燃烧促进剂、体脂肪减少剂、脂肪蓄积防止剂。
本发明者等基于上述各种发现,进一步反复探讨后完成了本发明。

本发明的脂肪燃烧促进剂、体脂肪减少剂、脂肪蓄积防止剂以及用于促进脂肪燃烧、用于减少体脂肪或用于防止脂肪蓄积的饮食品,对维持健康非常有益,其不改变总消耗热量,达到使脂质代谢亢进的效果。

具体实施方式
本发明的脂肪燃烧促进剂、体脂肪减少剂、脂肪蓄积防止剂以及用于促进脂肪燃烧、用于减少体脂肪或用于防止脂肪蓄积的饮食品(下面,也将它们统称为“脂肪燃烧促进剂等”),其特征在于含有原花色素作为有效成分。
作为本发明的脂肪燃烧促进剂等的有效成分使用的原花色素指:由以黄烷-3-醇和/或黄烷-3,4-二醇为结构单位的聚合度2以上、优选2~10聚体、更优选2~4聚体的缩聚物构成的化合物组、衍生物以及它们的立体异构体。原花色素中,以黄烷-3-醇和/或黄烷-3,4-二醇为结构单位的聚合度是2~4的缩聚物称为OPC(oligomeric proanthocyanidin)。OPC是强力抗氧化物质(参照:日本特公平3-7232),在植物的叶、树皮、水果的皮或种子的部分含量丰富。具体而言,在葡萄、松树的树皮、花生的薄皮、银杏、洋槐的果实、越橘、南方越橘、草莓、鳄梨、大麦、小麦、大豆、黑豆、可可等中含有。还已知在西非的可乐果、秘鲁的Rathania的根中也含有OPC。OPC是不能在人体内生物合成的物质。
关于作为本发明的脂肪燃烧促进剂等的有效成分使用的原花色素,原料的来源或原料的利用部分、制造方法、精制方法不受任何限制,可采用上述树皮、果实或种子的粉碎物、或它们的提取物之类的食品原料。特别优选松树皮、更优选富含OPC的法国海岸松树皮的提取物。优选法国海岸松树皮作为原花色素的原料。
通过采用公知的方法[例如,专利文献1记载的方法或从松树的树皮提取的方法;R.W.Hemingway等,Phytochemistry,1983年,第22卷,p.275-281]或以其为基准的方法,可容易地从上述各种植物体中得到原花色素。
下面,以富含OPC的松树皮的提取物为例,说明原花色素的制备方法。
用水或有机溶剂提取松树皮,可得到松树皮提取物。用水提取时,采用温水或热水。作为提取时使用的有机溶剂,采用食品或药剂制造中可接受的有机溶剂,例如甲醇、乙醇、丙醇、丁醇、丁烷、丙酮、己烷、环己烷、丙二醇、含水乙醇、含水丙二醇、丁酮、甘油、乙酸甲酯、乙酸乙酯、乙醚、二氯甲烷、食用油脂、1,1,1,2-四氟乙烷、1,1,2-三氯乙烯等。上述水以及有机溶剂既可单独使用,亦可组合使用。特别优选热水、含水乙醇、含水丙二醇。
对从松树皮提取原花色素的方法无特殊限制,可采用加温提取法、超临界流体提取法等。
超临界流体提取法是用超过物质的气液临界点(临界温度、临界压力)的状态的流体即超临界流体进行提取的方法。作为超临界流体,可采用二氧化碳、乙烯、丙烷、一氧化二氮(笑气)等,特别优选二氧化碳。
超临界流体提取法包括用超临界流体提取目标成分的提取过程和将目标成分与超临界流体分离的分离过程。分离过程可实施利用压力变化的提取分离、利用温度变化的提取分离、或利用吸附剂、吸收剂的提取分离中的任一种。
另外,还可以实施采用提携剂添加法的超临界流体提取。该方法在超临界流体中添加2~20%左右的例如乙醇、丙醇、正己烷、丙酮、甲苯等,用得到的提取流体进行超临界流体提取,使OPC等被提取物相对于提取流体的溶解度显著上升或使分离的选择性增强,是高效制得松树皮提取物的方法。
除上述方法外,还可利用液体二氧化碳间歇法、液体二氧化碳回流法、超临界二氧化碳回流法等方法从松树皮中提取。
按上述方法得到的原花色素,呈液状或半固体状,通过减压馏去、喷雾干燥、冷冻干燥等公知方法除去其中的提取溶剂后,可直接作为含有原花色素的浓缩物或干燥物使用。进一步精制时,可采用柱色谱法、逆流分配法等公知的精制方法来实现目的。
本发明的脂肪燃烧促进剂等中含有的原花色素,水中溶解性好,生体的吸收性高。无论是酸性、中性还是碱性条件下均具有较高的稳定性,易配合于饮食品中并维持其功能。另外,在摄取开始后短期间内即可产生效果,少量摄取也能获得充分的效果,因此,作为在饮食品的摄取许可量和摄取形态上有限制的幼儿和老人等的食物原材料,利用价值高。因此,本发明的饮食品也可以是例如健康食品、保健功能食品(特定保健食品、营养功能食品)等。
本发明的脂肪燃烧促进剂、体脂肪减少剂、脂肪蓄积防止剂(下面,也统称为本法明的制剂),可通过公知的方法,适宜地制成片剂、丸剂、胶囊、颗粒、粉末、散剂、液体制剂等固体或溶液的形态。即,将原花色素与所需添加剂混合,采用以往已充分确立的公知的制剂制造方法,制造作为本发明的制剂有效的固体制剂或液体制剂。作为添加剂,可列举:赋形剂、pH调节剂、清凉剂、悬浮剂、稀释剂、消泡剂、增稠剂、助溶剂、崩解剂、粘合剂、润滑剂、抗氧化剂、包衣剂、着色剂、矫味矫臭剂、表面活性剂、增塑剂或香料等。
上述赋形剂可列举:D-山梨糖醇、D-甘露糖醇或木糖醇等糖醇、葡萄糖、白糖、乳糖或果糖等糖类、微晶纤维素、羧甲基纤维素纳、磷酸氢钙、小麦淀粉、大米淀粉、玉米淀粉、马铃薯淀粉、糊精、β-环糊精、轻质无水硅酸、氧化钛或硅酸铝镁等。
上述pH调节剂可列举:柠檬酸、苹果酸、磷酸氢钠或磷酸二钾等。
上述清凉剂可列举:1-薄荷醇或薄荷水等。
上述悬浮剂可列举:高岭土、羧甲基纤维素钠、黄原胶、甲基纤维素或西黄蓍胶等。
上述稀释剂可列举:纯化水、乙醇、植物油或乳化剂等。
上述消泡剂可列举:聚二甲基硅氧烷或硅消泡剂等。
上述增稠剂可列举:黄原胶、西黄蓍胶、甲基纤维素或糊精等。
上述助溶剂可列举:乙醇、蔗糖脂肪酸酯或聚乙二醇等。
上述崩解剂可列举:低取代羟丙基纤维素、羧甲基纤维素钙、交联羧甲基纤维素钠、羟丙基淀粉或部分α化淀粉等。
上述粘合剂可列举:甲基纤维素、羟丙基纤维素、羟丙基甲基纤维素、聚乙烯吡咯烷酮、明胶、阿拉伯胶、乙基纤维素、聚乙烯醇、支链淀粉、α化淀粉、琼脂、西黄蓍胶、海藻酸钠或海藻酸丙二醇酯等。
上述润滑剂可列举:硬脂酸、硬脂酸镁、硬脂酸钙、硬脂酸聚烃氧酯、鲸蜡醇、滑石粉、硬化油、蔗糖脂肪酸酯、聚二甲基硅氧烷、蜂蜡或白蜡等。
上述抗氧化剂可列举:抗坏血酸、二丁基羟基甲苯(BHT)、没食子酸丙酯、丁基羟基茴香醚(BHA)、生育酚或柠檬酸等。
上述包衣剂可列举:羟丙基甲基纤维素、羟丙基纤维素、甲基纤维素、乙基纤维素、羟丙甲纤维素酞酸酯、乙酸羟丙基甲基纤维素琥珀酸酯、羧甲基乙基纤维素、醋酸纤维素酞酸酯、聚乙烯基乙缩醛二乙胺乙酯、甲基丙烯酸氨基烷基酯共聚物、甲基丙烯酸共聚物、聚乙烯醇缩醛二乙胺基乙酸脂(polyvinylacetal diethylamino acetate)、或虫胶等。
上述着色剂可列举:姜黄提取液、核黄素、氧化钛或胡萝卜素溶液等。
上述矫味矫臭剂可列举:柠檬酸、己二酸、抗坏血酸、果糖、D-山梨糖醇、葡萄糖、糖精钠、单糖浆、白糖、蜂蜜、甜菊茶、甘草、橙皮油、橙皮酊、茴香油、薄荷或薄荷醇等。
上述表面活性剂可列举:聚氧乙烯氢化蓖麻油、单硬脂酸甘油酯、失水山梨糖醇单硬脂酸酯、失水山梨糖醇单月桂酸酯、聚氧乙烯聚氧丙烯、聚山梨酯类、月桂基硫酸钠、聚乙二醇类或蔗糖脂肪酸酯等。
上述增塑剂可列举:柠檬酸三乙酯、聚乙二醇、三乙酸甘油酯或鲸蜡醇等。
上述香料可列举:动物性香料或植物性香料等天然香料、或单离香料或纯合成香料等合成香料等。
本发明的制剂中的原花色素量,相对于制剂整体,一般约为1~80重量%,优选约为2~50重量%。
本发明制剂的给与方法,可以是经口给与,也可以是非经口给与。另外,本发明的有效成分原花色素的给与量,根据其种类、其剂型或患者的年龄、体重、适应症状等不同而不同,例如内服剂,成人1日给与1次~数次,1次的量约为1~500mg,优选3~300mg左右。
本发明的饮食品,可通过在饮食品制造时将原花色素或上述含有原花色素的制剂配合于饮食品材料中制得。可制成各种食品形态,例如:面包、口香糖、曲奇饼、巧克力、谷物食品等固体食品;果酱、冰淇淋、酸奶、果冻等果酱状、乳膏状或凝胶状食品;果汁、咖啡、可可饮料、绿茶、乌龙茶、红茶等饮料等。另外,还可以配合于调味料、食品添加物等。原花色素或上述含有原花色素的制剂在饮食品材料中的配合量不受特殊限制,一般约为0.0001~80重量%,优选约0.005~50重量%。尤其是饮料时,一般为1mg/L~20g/L,优选2mg/L~10g/L。
当本发明是饮食品时,由于无需担心副作用,原花色素摄取量可以与上述内服剂的给与量相同。
本发明的饮食品适用于燃烧体脂肪、减少体脂肪或防止体脂肪蓄积。
[实施例]
下面,以富含OPC的松树皮提取物为例,结合实施例,详细说明本发明,但本发明不限于此。
(实施例1)
将含有40重量%以上(作为OPC为20重量%以上)原花色素且含有5重量%以上儿茶素的法国海岸松树皮提取物8重量份与微晶纤维素22重量份、乳糖66重量份、蔗糖酯3重量份、二氧化硅1重量份配合,制造片剂(每片250mg,原花色素含量9.5mg)(样品片)。
(比较例1)
除不使用法国海岸松树皮提取物外,其余均与实施例1相同,制造片剂(每片250mg)(安慰片)。
(试验例1)
以9名成年男性(年龄22.1±1.1岁)为对象,采用双盲交叉法,摄取实施例1制造的样品片或比较例1制造的安慰片1周后,进行固定负荷运动。经1周洗脱(wash out)期后,摄取上一次未摄取的任一片剂1周,再次进行固定负荷运动。固定负荷运动3天前开始将饮食调整为蛋白质+脂质∶糖为1∶1。
原花色素1天给与1次,原花色素的每日给与量与受试者的体重无关,一律为2片/日(19mg/日)。关于固定负荷运动,先实施4分钟20瓦渐增的运动负荷试验,算出与血中乳酸浓度2mM相当的运动负荷,确定每个受试者的固定负荷运动强度。固定负荷运动为60分钟。
固定负荷运动开始30分钟前,使受试者处于安静状态,分别采集运动开始15分钟前、运动开始前(0分)、运动开始15分钟后、30分钟后、45分钟后、运动结束时(60分钟)、运动结束15分钟后(75分钟)的呼出气体。用质谱方式综合呼出气体分析装置(ARCO-1000 Series,Arco System公司制造),测定各时间点采集的呼出气体中氧摄取量(ml/分)(VO2)以及二氧化碳生成量(ml/分)(VCO2),算出各个时间点的VO2/VCO2,作为呼吸商(RQ)。
用得到的数值,通过以下方法,算出总消耗热量、糖消耗热量以及脂质消耗热量。
通过Zuntz Schmburg-Lusk的计算式(计算式1)算出单位时间的消耗热量。
将得到的单位时间消耗热量中运动开始15分钟后、30分钟后、45分钟后、运动结束时(60分钟)四点的平均值乘以60分钟,算出总消耗热量。
用下述计算式2得到糖消耗热量的比率(参照:日语原名「新エスカ21シリ一ズ16新エスカ21運動生理学」同文書院,P.26)后,乘以总消耗热量,算出糖消耗热量。
从总消耗热量中减去糖消耗热量,算出脂质消耗热量。
总消耗热量、糖消耗热量、脂质消耗热量的计算结果见图1和表1。

如图1和表1所示,运动前的消耗热量在样品片摄取时和安慰片摄取时并无差别。比较样品片摄取时与安慰片摄取时运动开始至运动结束为止的累积消耗热量,发现总消耗热量并无差别,但比较糖和脂质的消耗热量后,发现:与安慰片摄取时相比,样品片摄取时糖的消耗热量显著减少,脂质的消耗热量增加。由此可知,原花色素具有使脂质代谢亢进的作用。
本发明可提供使脂质代谢亢进的新型脂肪燃烧促进剂、体脂肪减少剂、脂肪蓄积防止剂、以及用于促进脂肪燃烧、用于减少体脂肪或用于防止脂肪蓄积的饮食品。
