背景技术
随着社会的发展,人们的生活压力越来越大,各类妇科疾病发病率居高不下,生殖系统各类问题早已成为影响女性健康和生存质量的重大问题。
女性朋友由于外生殖器构造复杂,外阴部与尿道、肛门邻近,皮肤、黏膜皱褶较多,既有皮脂腺、汗腺,同时又有前庭大腺和宫颈、阴道的分泌物,还有月经来潮,易受排泄物、分泌物的刺激而引起感染。妇科疾病比如阴道炎等是一些常见的多发病,通常且久治不愈,给正常的生活、工作带来极大的不便。西医认为女性私处的环境经常受到宿主的代谢产物、细菌本身的产物及外源性因素而不稳定。菌群非常复杂,除原虫、真菌外,尚包括很多需氧菌及厌氧菌,随着人们对身体健康的日益重视,尤其一些女性朋友,开始对私处的护理日渐重视,由于女性朋友容易受到私处等多种问题的困扰,因此,日常的杀菌、护理尤显重要。
女性的阴道粘膜对刺激或炎症极为敏感。易感染的女性在例如接触游泳池或其他水体、或性交后,有可能会引发阴道粘膜区的炎症反应。激素变化,例如服用避孕药、妊娠期、哺乳期、或者在绝经期,也可能引起阴道粘膜的刺激反应。阴道粘膜刺激不仅疼痛,而且可能发展成进一步的炎症,可影响女性其他器官的正常功能。实际上,每年有几十亿女性被这种刺激感染,并导致女性的幸福感下降。初步研究,阴道粘膜刺激或炎症是由于阴道菌群紊乱引起的。因此,女性日常中应加强阴道的护理,维持阴道内环境的健康平衡。
不少女性清洗外阴会使用普通沐浴露或者香皂等清洁用品,甚至于部分女性还会使用高锰酸钾或者盐水。如长期使用会造成外阴酸性环境被破坏,改变了阴道的pH值,使菌群失调,局部抵抗力下降,诱发妇科疾病。
目前治疗妇科炎症的药物主要分为西药,中药以及中西药结合。从临床治疗效果来看,西药医尚无理想的治疗效果,中药对妇科炎症的治疗拥有较长历史,积累了较多的经验,目前,市场上的妇科药琳琅满目,但都存在抑菌效果差,并且不能有效治疗妇科疾病,容易复发的缺点。

进一步的,在本发明中,所述水解燕麦,采用下述方法制备而成:将燕麦粉碎过20-40目筛,得到燕麦粉;将燕麦粉加入水中配制成燕麦粉质量分数为10-20%的原液,将原液用0.3-0.8mol/L的醋酸调节pH至1-3,再加入壳聚糖混合均匀得到浆液,所述壳聚糖在浆液中的质量分数为0.1-0.5%,将浆液以转速为150-350r/min进行搅拌处理40-60min,过滤,得到水解液;将水解液在绝对压强为0.04-0.08MPa、温度为50-65℃下进行减压浓缩,浓缩时间为4-8小时,得到浓缩液;将浓缩液进行真空冷冻干燥,得到水解燕麦。

在本发明中,所述水解大豆,采用下述方法制备而成:8-12重量份大豆粉、0.1-0.4重量份菠萝蛋白酶加入95-105重量份水混合均匀,水解1-3小时,灭酶,离心分离,得到水解液;在水解液中加入水解液重量0.03-0.09%的蒜糖醇混合均匀,减压浓缩,真空冷冻干燥。
进一步的,在本发明中,所述水解大豆,采用下述方法制备而成:将大豆粉碎过10-20目筛,取8-12重量份粉碎后的大豆、0.1-0.4重量份菠萝蛋白酶加入95-105重量份水中混合均匀,在温度为30-50℃以转速为100-300r/min搅拌下水解1-3小时,然后在温度为85-95℃灭酶活4-10分钟,离心分离,得到水解液;在水解液中加入水解液重量0.03-0.09%的蒜糖醇混合均匀,然后在真空度为0.04-0.08MPa、温度为50-65℃下进行减压浓缩,浓缩时间为4-8小时,得到浓缩液;将浓缩液进行真空冷冻干燥,得到水解大豆。
所述离心分离的转速为5000-8000r/min,时间为15-25min。
所述真空冷冻干燥的条件为:控制物料高度为4-7mm,预冻温度为-25~-18℃,当温度降到设定温度后保持1-2.5小时,升华温度设定为8-13℃,解析温度设定为22-28℃,绝对压强为15-25Pa,干燥时间为17-27小时。
本发明的抑菌护理液,可以采用行业通用方法进行制备,例如将各原料混合均匀即得。

本发明抑菌护理液,无耐药性,不影响阴道有益菌群,能够有效维护阴道内正常微生态环境,用于日常阴道部位抑菌、护理、清洁,能有效抑菌,洁净及滋润私密部位,保护私处粘膜和皮肤不受损伤,此外还可以缓解阴道干涩、性交疼痛和性欲低下等症状。使用方法:清洁后,取本品喷雾私密部位,无需清洗,使用方便。
具体实施方式
下面通过具体实施方式对本发明进一步说明。
实施例中茶叶提取物,茶氨酸HPLC含量98%,CAS号:3081-61-6,陕西金润生物科技有限公司提供。
实施例中莲叶提取物,CAS号:475-83-2,HPLC含量10%,陕西慈缘生物技术有限公司提供。
实施例中菠萝蛋白酶采用南宁东恒华道生物科技有限责任公司提供的型号为A003的菠萝蛋白酶,酶活力为60万u/g。
实施例中燕麦,拉丁学名:Avena sativa L.,为禾本科植物。本实施例中燕麦为内蒙古沃禾泥农业科技有限公司提供的全胚芽燕麦仁,等级为1,品种为裸燕麦,产地为内蒙古。
实施例中壳聚糖为浙江一梦生物科技有限公司提供的食品级壳聚糖。
实施例中大豆由哈尔滨市兴利米制品有限公司提供,等级为1,产地为哈尔滨。
实施例中蒜糖醇,CAS号:488-44-8。
抗自由基的测定,采用DPPH法:
准确称取DPPH,配制成0.2mmol/LDPPH的溶液,于棕色瓶避光低温保存。用去离子水将抑菌护理液样品稀释到500mg/mL,得到样品液。取2mL样品液于试管内,加入2mL、0.2mmol/LDPPH溶液混合均匀,室温下避光静置30min后,用紫外分光光度计UV-2000(尤尼柯(上海)有限仪器公司)测定517nm处吸光度值,计为Ai,同时测定2mL、1.0mg/mL样品液加2mL无水乙醇,室温下避光静置30min后,用紫外分光光度计UV-2000(尤尼柯(上海)有限仪器公司)测定在517nm处吸光度值,计为Aj,以及2mL、0.2mmol/LDPPH溶液加2ml蒸馏水静置,用紫外分光光度计UV-2000(尤尼柯(上海)有限仪器公司)测定在517nm处吸光度值,计为Ac。样品对DPPH自由基的清除率按下式计算:DPPH自由基清除率=[1-(Ai-Aj)/Ac]×100%。
抗菌性能测定:
(1)药敏片的制备:取直径3cm无菌干燥的滤纸片,每片滴加抑菌护理液25μL,然后将滤纸片平放于清洁的无菌平皿内,干燥后备用;
(2)阴性对照样片的制备:取直径3cm无菌干燥滤纸片,每片滴加无菌蒸馏水25μL,干燥后备用;
(3)K-B纸片扩散法药敏试验:用无菌棉签蘸取浓度为5×106cfu/ml试验菌悬液,在管壁上挤去多余菌液,涂布整个营养琼脂培养基平板表面,反复3次;每涂抹1次,平板转动60度,涂布均匀,最后将棉签绕平板边缘涂抹一周;盖好平皿,置室温干燥5min;贴上待测药敏片和阴性对照样片,37℃培养30h后观察抑菌效果,测试菌种白色念珠菌ATCC 10231(Candida albicans),测量抑菌环直径。
实施例1
抑菌护理液,由以下质量百分比的原料制备而成:莲叶提取物0.4%、余量为水。将水加热至45℃,加入莲叶提取物,搅拌混合均匀,冷却至至室温即可制得本发明的抑菌护理液。
实施例2
抑菌护理液,由以下质量百分比的原料制备而成:茶叶提取物0.4%、莲叶提取物0.4%、余量为水。将水加热至45℃,加入莲叶提取物和茶叶提取物,搅拌混合均匀,冷却至至室温即可制得本发明的抑菌护理液。
实施例3
抑菌护理液,由以下质量百分比的原料制备而成:茶叶提取物0.4%、莲叶提取物0.4%、水解燕麦0.3%、余量为水。将水加热至45℃,加入水解燕麦、莲叶提取物和茶叶提取物,搅拌混合均匀,冷却至至室温即可制得本发明的抑菌护理液。
所述水解燕麦的制备方法为:将燕麦粉碎过30目筛,得到燕麦粉;将燕麦粉加入去离子水中配制成燕麦粉质量分数为15%的原液,将原液用0.5mol/L的醋酸调节pH至1.5,再加入壳聚糖混合均匀得到浆液,所述壳聚糖在浆液中的质量分数为0.2%,将浆液以转速为200r/min进行搅拌处理50min,采用300目滤布过滤,得到水解液;将水解液在绝对压强为0.06MPa、温度为55℃下进行减压浓缩,浓缩时间为6小时,得到浓缩液;将浓缩液进行真空冷冻干燥,所述真空冷冻干燥的条件为:控制浓缩液高度为6mm,预冻温度为-22℃,当温度降到设定温度后保持1.5小时,升华温度设定为10℃,解析温度设定为24℃,绝对压强为20Pa,干燥时间为22小时,得到水解燕麦。
实施例4
抑菌护理液,由以下质量百分比的原料制备而成:茶叶提取物0.4%、莲叶提取物0.4%、水解大豆0.3%、余量为水。将水加热至45℃,加入水解大豆、莲叶提取物和茶叶提取物,搅拌混合均匀,冷却至至室温即可制得本发明的抑菌护理液。
所述水解大豆的制备方法为:将大豆粉碎过16目筛,取10重量份粉碎后的大豆、0.2重量份菠萝蛋白酶加入100重量份去离子水中混合均匀,在温度为45℃以转速为150r/min搅拌下水解2小时,然后在温度为90℃灭酶活6分钟,以转速为6000r/min离心分离20min,弃沉淀,取上清液,得到水解液;在水解液中加入水解液重量0.05%的蒜糖醇混合均匀,然后在绝对压强为0.06MPa、温度为55℃下进行减压浓缩,浓缩时间为6小时,得到浓缩液;将浓缩液进行真空冷冻干燥,所述真空冷冻干燥的条件为:控制浓缩液高度为6mm,预冻温度为-22℃,当温度降到设定温度后保持1.5小时,升华温度设定为10℃,解析温度设定为24℃,绝对压强为20Pa,干燥时间为24小时,得到水解大豆。
实施例5
抑菌护理液,由以下质量百分比的原料制备而成:茶叶提取物0.4%、莲叶提取物0.4%、水解燕麦0.15%、水解大豆0.15%、余量为水。将水加热至45℃,加入水解大豆、水解燕麦、莲叶提取物和茶叶提取物,搅拌混合均匀,冷却至至室温即可制得本发明的抑菌护理液。
所述水解燕麦的制备方法,同实施例3;所述水解大豆的制备方法,同实施例4。
实施例6
抑菌护理液,由以下质量百分比的原料制备而成:茶叶提取物0.4%、莲叶提取物0.4%、水解燕麦0.15%、水解大豆0.15%、余量为水。将水加热至45℃,加入水解大豆、水解燕麦、莲叶提取物和茶叶提取物,搅拌混合均匀,冷却至至室温即可制得本发明的抑菌护理液。
所述水解大豆的制备方法,同实施例5。
所述水解燕麦的制备方法为:将燕麦粉碎过30目筛,得到燕麦粉;将燕麦粉加入去离子水中配制成燕麦粉质量分数为15%的原液,将原液用0.5mol/L的醋酸调节pH至1.5,再加入壳聚糖混合均匀得到浆液,所述壳聚糖在浆液中的质量分数为0.2%,将浆液进行超声处理50min,所述超声处理的超声功率为250W、超声频率为35kHz,采用300目滤布过滤,得到水解液;将水解液在绝对压强为0.06MPa、温度为55℃下进行减压浓缩,浓缩时间为6小时,得到浓缩液;将浓缩液进行真空冷冻干燥,所述真空冷冻干燥的条件为:控制浓缩液高度为6mm,预冻温度为-22℃,当温度降到设定温度后保持1.5小时,升华温度设定为10℃,解析温度设定为24℃,绝对压强为20Pa,干燥时间为22小时,得到水解燕麦。

对实施例1-6的抑菌护理液的性能进行测试,具体测试结果见表1。

Claims (2)
1.一种抑菌护理液,包括下述质量百分比的原料:茶叶提取物0.1-0.3%、莲叶提取物0.1-0.3%、水解燕麦0.1-0.5%、水解大豆0.1-0.5%、余量为水;
所述水解燕麦,采用下述方法制备而成:将燕麦粉碎过20-40目筛,得到燕麦粉;将燕麦粉加入水中配制成燕麦粉质量分数为10-20%的原液,将原液用0.3-0.8mol/L的醋酸调节pH至1-3,再加入壳聚糖混合均匀得到浆液,所述壳聚糖在浆液中的质量分数为0.1-0.5%,将浆液进行超声处理40-60min,过滤,得到水解液;将水解液在真空度为0.04-0.08MPa、温度为50-65℃下进行减压浓缩,浓缩时间为4-8小时,得到浓缩液;将浓缩液进行真空冷冻干燥,得到水解燕麦;

2.一种抑菌护理液,其特征在于,由下述质量百分比的原料制备而成:茶叶提取物0.4%、莲叶提取物0.4%、水解燕麦0.15%、水解大豆0.15%、余量为水;
所述水解燕麦的制备方法为:将燕麦粉碎过30目筛,得到燕麦粉;将燕麦粉加入去离子水中配制成燕麦粉质量分数为15%的原液,将原液用0.5mol/L的醋酸调节pH至1.5,再加入壳聚糖混合均匀得到浆液,所述壳聚糖在浆液中的质量分数为0.2%,将浆液进行超声处理50min,所述超声处理的超声功率为250W、超声频率为35kHz,采用300目滤布过滤,得到水解液;将水解液在绝对压强为0.06MPa、温度为55℃下进行减压浓缩,浓缩时间为6小时,得到浓缩液;将浓缩液进行真空冷冻干燥,所述真空冷冻干燥的条件为:控制浓缩液高度为6mm,预冻温度为-22℃,当温度降到设定温度后保持1.5小时,升华温度设定为10℃,解析温度设定为24℃,绝对压强为20Pa,干燥时间为22小时,得到水解燕麦;
所述水解大豆的制备方法为:将大豆粉碎过16目筛,取10重量份粉碎后的大豆、0.2重量份菠萝蛋白酶加入100重量份去离子水中混合均匀,在温度为45℃以转速为150r/min搅拌下水解2小时,然后在温度为90℃灭酶活6分钟,以转速为6000r/min离心分离20min,弃沉淀,取上清液,得到水解液;在水解液中加入水解液重量0.05%的蒜糖醇混合均匀,然后在绝对压强为0.06MPa、温度为55℃下进行减压浓缩,浓缩时间为6小时,得到浓缩液;将浓缩液进行真空冷冻干燥,所述真空冷冻干燥的条件为:控制浓缩液高度为6mm,预冻温度为-22℃,当温度降到设定温度后保持1.5小时,升华温度设定为10℃,解析温度设定为24℃,绝对压强为20Pa,干燥时间为24小时,得到水解大豆。
