Abstract
Patente de Invenção: <B>''POLIMERASES DE DNA TERMOESTáVEL ALTERADA PARA SEQuENCIAMENTO''<D>. São providas polimerases de DNA termoestáveis modificadas possuindo eficiência melhorada para incorporar nucleotídeos não-convencionais, tais como aqueles marcados com corantes da família da fluoresceína. Tais polimerases de DNA termoestável modificadas podem ser preparadas por processos da tecnologia de DNA recombinante. Polimerases de DNA termoestável recombinantes preparadas de acordo com a presente invenção são vantajosas em muitas aplicações de síntese de DNA <MU>in vitro<MV>, tais como, seq³enciamento de DNA, síntese de DNA marcado e produção de produtos de extensão de primer marcados. As ditas enzimas de polimerase são especificamente úteis nos processos de seq³enciamento de ácido nucléico de terminação em cadeia. São também providos os ácidos nucléicos codificando as polimerases de DNA termoestáveis, recombinantes da presente invenção, bem como vetores e células hospedeiras compreendendo os mesmos. Também são fornecidos cojuntos compreendendo as polimerases de DNA termoestáveis recombinantes da presente invenção. As polimerases e processos da presente invenção fornecem custo significativo e vantagens eficiente para seq³enciamento de DNA.
