Abstract

본 발명은 세균 프럭토실트랜스퍼라제 유전자를 선택적으로 발형함으로써 유전자 전환 식물의 종자, 괴경 또는 잎에서 프럭토스 중합체를 합성 및 축척시키는 방법에 관한 것이다. 선택적 발형은 시기, 조직 특이적 발현 및 특히 세포내 위치의 조절을 포함한다. 성공적인 형질전환제를 확립된 작물의 세포중 액포내에, 식물의 성숙, 수확 또는 저장시의 분해에 기인한 공동 생성물의 손실 또는 수율 손실의 염려없이, 슈크로스를 이용하여 프럭탄을 축적시킨다. 프럭턴 생산 증가는 프럭토스 감마제 산업에 유익할 것이며 사료에 사용되는 낟알의 가치를 상승시킬 것이다.

Claims (27)

  1. 옥수수, 감자 및 담배로 구성된 군으로부터 선택된 식물 세포를 형질전환시켜 상기 식물 세포의 액포에서 프럭탄을 생산할 수 있도록, 레반슈크라제 유전자의 코딩 서열에 작동가능하게 연결된 액포 표적 서열에 작동 가능하게 연결된 조직 특이적 프로모터로 이루어진 재조합 DNA 구조물.
  2. 식물의 세포내 액포에 프럭탄을 생산하여 축적하도록 제1항의 구조물로 형질전환된, 옥수수, 감자 및 담배로 구성된 군으로부터 선택된 식물.
  3. 제2항의 식물을 재배하고 수확한 다음 수확된 식물로부터 프럭탄을 추출하는 것으로 이루어진, 프럭토스의 생산 방법.
  4. 옥수수, 감자 및 담배로 구성된 군으로부터 선택된 식물 세포를 형질전환시켜 상기 식물 세포의 액포에서 덱스트란을 생산할 수 있도록, 덱스트란슈크라제 유전자의 코딩 서열에 작동가능하게 연결된 액포 표적 서열에 작동가능하게 연결된 조직 특이적 프로모터로 이루어진 재조합 DNA 구조물.
  5. 식물의 세포내 액포에 덱스트란을 생산하여 축적하도록 제4항의 구조물로 형질전환된, 옥수수, 감자 및 담배로 구성된 군으로부터 선택된 식물.
  6. 제5항의 식물을 재배하고 수확한 다음 수확된 식물로부터 프럭탄을 추출하는 것으로 이루어진, 덱스트란의 생산 방법.
  7. 옥수수, 감자 및 담배로 구성된 군으로부터 선택된 식물 세포를 형질전시켜 상기 식물 세포의 액포에서 알터난을 생산할 수 있도록, 알터난슈크라제 유전자의 코딩 서열에 작동가능하게 연결된 액포 표적 서열에 작동가능하게 연결된 조직 특이적 프로모터로 이루어진 재조합 DNA 구조물.
  8. 식물의 세포내 액포에 알터난을 생산하여 축적하도록 제 7항의 구조물로 형질전환된, 옥수수, 감자 및 담배로 구성된 군으로부터 선택된 식물.
  9. 제8항의 식물을 재배하고 수확한 다음 수확된 식물로부터 알터난을 추출하는 것으로 이루어진, 알터난의 생산 방법.
  10. 제1항에 있어서, 조직 특이적 프로모터가 종자 특이적인 재조합 DNA 구조물.
  11. 제1항에 있어서, 조직 특이적 프로모터가 괴경 특이적인 재조합 DNA 구조물.
  12. 제4항에 있어서, 조직 특이적 프로모터가 종자 특이적인 재조합 DNA 구조물.
  13. 제4항에 있어서, 조직 특이적 프로모터가 괴경 특이적인 재조합 DNA 구조물.
  14. 제7항에 있어서, 조직 특이적 프로모터가 종자 특이적인 재조합 DNA 구조물.
  15. 제7항에 있어서, 조직 특이적 프로모터가 괴경 특이적인 재조합 DNA 구조물.
  16. 종자중에 축적되는 프럭탄을 생산하는, 제10항의 구조물로 형질전환된 식물.
  17. 종자중에 축적되는 덱스트란을 생산하는, 제12항의 구조물로 형질전환된 식물.
  18. 종자중에 축적되는 알터난을 생산하는, 제14항의 구조물로 형질전환된 식물.
  19. 괴경중에 축적되는 프럭탄을 생산하는, 제11항의 구조물로 형질전환된 식물.
  20. 괴경중에 축적되는 덱스트란을 생산하는, 제13항의 구조물로 형질전환된 식물.
  21. 괴경중에 축적되는 알터난을 생산하는, 제15항의 구조물로 형질전환된 식물.
  22. 천연 슈크로스 농도보다 높은 슈크로스 농도를 갖는 식물 세포내로, 식물 세포를 형질환시켜 형질전환된 식물이 프럭탄을 생산 및 축적할 수 있도록, 레반슈크라제 유전자의 코딩 서열에 작동가능하게 연결된 액포 표적 서열에 작동가능하게 연결된 조직 특이적 프로모터로 이루어진 재조합 DNA 구조물을 삽입하는 것을 포함하는, 식물중 프럭탄 농도를 증가시키는 방법.
  23. 제22항의 구조물로 형질전환된 식물.
  24. 제23항의 형질전환된 식물로부터 생산된 종자.
  25. 제22항에 있어서, 식물이 옥수수이고, 생산된 종자가 변화된 프럭탄 농도를 갖는 옥수수 종자인 방법.
  26. 식물 세포를 형질전환시켜 식물 세포의 세포질에서 프럭탄을 축적할 수 있도록, 레반슈크라제 유전자의 변이된 코딩 서열에 작동가능하게 연결된 액포 표적 서열에 작동가능하게 연결된 조직 특이적 프로모터로 이루어진 재조합 DNA 구조물.
