Abstract
Description
도 2는 아토피 피부염 질환 모델의 구축 및 실험 물질의 적용 일정을 개괄적으로 보여준다.
도 3은 아토피 피부염 질환 유발 NC/Nga 마우스의 피부염 증상 점수를 보여준다.
도 4는 아토피 피부염 질환 유발 28일 후에 NC/Nga 마우스의 피부 긁음(scratch) 횟수를 측정한 결과를 보여준다.
도 5는 아토피 피부염 질환 유발 28일 후에 촬영한 NC/Nga 마우스 피부의 사진이다.
도 6은 아토피 피부염을 유발한 NC/Nga 마우스 피부에서 표피의 두께와 염증 세포 침투 모습을 보여주는 사진이다. 패널 A는 아토피 피부염 비유도(non-induction) 대조군, 패널 B는 아토피 피부염 유도 대조군, 패널 C는 아토피 피부염 유도 마우스에 노근 추출물 함유 연고를 적용한 실험군이다.
도 7은 아토피 피부염을 유발시킨 NC/Nga 마우스 피부에서 비만세포(mast cell)의 세포수이다. 패널 A는 아토피 피부염의 유도 대조군이고, 패널 B는 아토피 피부염 유도 마우스에 노근 추출물 함유 연고를 적용한 실험군이다.
도 8은 아토피 피부염을 유발시킨 NC/Nga 마우스 피부에서 신경 섬유의 개수를 보여준다. 패널 A는 아토피 피부염 비유도(non-induction) 대조군, 패널 B는 아토피 피부염 유도 마우스에 노근 추출물 함유 연고를 적용한 실험군이다.
도 9는 아토피 피부염을 유발시킨 NC/Nga 마우스 혈청에서 IgE 항체의 농도 변화를 보여준다. "C"는 아토피 피부염의 유도 대조군이고, "N"은 아토피 피부염 유도 마우스에 노근 추출물 함유 연고를 적용한 실험군이다.
도 10은 아토피 피부염을 유발시킨 NC/Nga 마우스에서 추출한 액와림프절(axillary lymph node, ALN)의 크기를 비교한 사진이다. 왼쪽 사진은 아토피 피부염 비유도(non-induction) 대조군 사진이며, 가운데 사진은 아토피 피부염 유도군 사진이고, 오른쪽 사진은 아토피 피부염 유발시킨 마우스에 노근 추출물 연고를 적용한 실험군의 사진이다.
도 11은 Dfb로 자극하여 배양한 액와림프절(axillary lymph node, ALN) 세포와 비장세포에서의 IFN-γ의 생성을 측정한 결과이다. "C"는 아토피 피부염의 유도 대조군이고, "N"은 아토피 피부염 유도 마우스에 노근 추출물 함유 연고를 적용한 실험군이다.

도 14는 NC/Nga 마우스에 진드기 추출물(mite extract)을 도포하여 피부염을 유발하고, 양성 대조군 약물로서 덱사메타손(dexamethasone, Dex)을 5mg/kg 체중의 용량으로, 실험군으로서 노근 추출물(Nk)을 250 mg/kg 체중의 용량으로, 음성 대조군으로서는 약물 전달체(vehicle)을 각각 경구투여한 후, 피부염의 완화 효과를 피부염 유발지수 스코어링(scoring)을 통해 평가한 결과이다. *: p<0.05 vs. vehicle (D19: 아토피 유발 종료일, D27: 투여종료일).
도 15는 NC/Nga 마우스에 진드기 추출물(mite extract)을 도포하여 피부염을 유발하고, 양성 대조군 약물로서 덱사메타손(dexamethasone,Dex)을 5mg/kg 체중의 용량으로, 실험군으로서 노근 추출물(Nk)을 250 mg/kg 체중의 용량으로, 음성 대조군으로서는 약물전달체(vehicle)을 각각 경구투여한 후, 피부염의 완화 효과를 긁음(scratching) 횟수 측정을 통해 평가한 결과이다. *: p<0.05 vs. vehicle (D19: 아토피 유발 종료일, D27: 투여종료일).
도 16은 C57BL/6 생쥐를 0.5% DNFB(2,4-dinitro-1-fluorobenzene) 25μL로 감작시킨 후 5일 후, 0.2% DNFB를 다시 귀의 피하에 주사하여 접촉성 피부염을 유발하고, 노근 수추출물을 투여하여 귀의 부종(염증)을 억제하는 정도를 평가한 결과이다.
도 17은 NC/Nga 마우스에 진드기 추출물(mite extract)를 도포하여 피부염을 유발하고, 노근 수추출물을 경구 투여하여 경피수분손실(transepidermal water loss)을 억제하는 효과를 평가한 결과이다.
도 18은 본 발명의 노근 수추출물의 성분을 TLC(Thin Layer chromatography)로 분석한 결과이다. 노근의 수추출물을 노근의 알코올 추출물의 성분과 비교 분석하였다. 레인 1은 수추출물이며, 레인 2는 알코올 추출물이다. 조건 1은 chloroform : methanol : water = 70 : 30 : 4의 전개용매, 조건 2는 ethyl formate : formic acid : water = 70 : 10 : 10의 전개용매, 조건 3은 butanol : isopropanol : acetic acid : water = 3 : 1 : 1 : 1의 전개용매를 사용하여 약 10cm 전개한 다음 UV 254nm (패널 A), UV 365nm (패널 B), 10% 황산(sulfuric acid)발색(패널 C)과, 닌히드린(ninhydrin) 발색(패널 D) 후 반점들의 Rf값을 확인하였다.

Claims (8)
- 수용성 당류(sugar)가 포함된 노근(Phragmitis Rhizoma)의 열수 추출물을 유효성분으로 포함하는 피부염 또는 아토피 피부염의 치료 또는 예방용 약제학적 조성물.

- 제 7 항에 있어서, 상기 화장품학적 조성물의 제형은 용액, 현탁액, 유탁액, 페이스트, 겔, 크림, 로션, 파우더, 비누, 계면활성제-함유 클렌징, 오일, 분말 파운데이션, 유탁액 파운데이션, 왁스 파운데이션 및 스프레이로 이루어지는 군에서 선택되는 제형인 것을 특징으로 하는 조성물.
