Description

【発明の詳細な説明】 ［産業上の利用分野］ 本発明は、新規にして、かつ安全なるテストステロン−
５α−レダクターゼ阻害剤に関するものである。
［従来の技術］ 男性型の禿頭、粗毛症、尋常性座癒、脂漏などの生理学
上の徴候は、代謝系に男性ホルモンが過剰に蓄積される
ことに起因する男性ホルモン刺激の増大に基づいている
ことが知られている。また最近、毛根、皮脂腺をはじめ
いくつかの器官において男性ホルモン活性の本体は、５
α−ジヒドロテストステロン（以下、５α−ＤＨＴと略
す。）であり、これは標的器官においてテストステロン
がテストステロン−５α−レダクターゼ（以下、レダク
ターゼと略す。）により還元されることによって生成す
ることが周知になっている。
そのため、男性ホルモン刺激の増大に起因する男性型の
禿頭、粗毛症、尋常性座癒、脂漏などの徴候は、レダク
ターゼを阻害することにより低減あるいは防止すること
ができると考えられ、レダクターゼを特異的に阻害する
いくつかの阻害剤が見出され、また合成されてきた。し
かし、今までに新規に合成された阻害剤は、ステロイド
構造あるいはステロイド類似構造を持っており、好まし
くないホルモン様作用を多少なりとも有しているという
欠点があった。
［発明が解決しようとする問題点］ 本発明者らは、上記事情に鑑み、ホルモン様作用を全く
持たず、安全性にも優れた天然のレダクターゼ阻害剤に
ついて鋭意研究を重ねた結果、工ビカテキン等のタンニ
ンがレダクターゼを阻害することを見出し本発明を完成
するに至った。
し問題点を解決するための手段］ すなわち、本発明は、エピカテキン、エピガロカテキン
、エピカテキンガレートおよびエピガロカテキンガレー
トの一種又は二種以上からなるテストステロン−５α−
レダクターゼ阻害剤である。
以下、本発明の構成について詳述する。
本発明に用いられるエピカテキン、エピガロカテキン、
エピカテキンガレートおよびエピガロカテキンガレート
の構造は下記に示すとおりである。
（以下余白） エピカテキン　　　　　　　ＨＨ これらは合成又は天然物から抽出して得られる。天然物
から抽出して得る一方法は、特に限定されないが以下の
ような方法が好ましい。
茶菓を熱湯もしくは４０〜７５％メタノール水溶液、４
０〜７５％エタノール水溶液および３０〜８０％アセト
ン水溶液から選ばれた１種の溶剤で抽出し、抽出成分を
含む溶液をクロロホルムで洗浄し、次いで該抽出成分を
酢酸エチル、ｎ−ブタノール、メチルイソブチルケトン
、アセトン等の有機溶媒に転溶したのち、該有機溶媒を
留去し、しかる後乾燥して粉末状またはフレーク状のタ
ンニン混合物が得られる。得られたものは、エピカテキ
ン、エピガロカテキン、エピカテキンガレートおよびエ
ピガロカテキンガレートを含んでおり、このものを使用
しても良いし、ざらに逆相分配カラムを用いアセトン：
テトラヒドロフラン：水＝Ｏ〜２５：０〜３５　：　６
５〜８５（容量％）なる展開溶媒にて高速液体クロマト
グラフィーを行ってそれぞれを単離し使用することがで
きる。（特開昭６Ｏ−１３７８０）エピカテキン、エピ
ガロカテキン、エピカテキンガレート°およびエピガロ
カテキンガレートは、ホルモン様作用は一切持たず、ま
た、他の好ましくない副作用も有していない。従って、
本発明のエピカテキン、エピガロカテキン、エピカテキ
ンガレートおよびエピガロカテキンガレートをレダクタ
ーゼ阻害剤として使用しても、ホルモン様作用は全く持
たず、しかも長期にわたり継続的に外用しても、安全性
には問題がないという優れた利点を有している。
［実施例］ 次に、本発明のエピカテキン、エピガロカテキン、エピ
カテキンガレートおよびエピガロカテキンガレートのレ
ダクターゼ阻害作用を実証する実験例を以下に具体的に
示す。
高安らの方法（Ｓ　、　Ｔ　ａｋａｙａｓｕ、　Ｋ　、
　Ａｄａｃｈｉ、　Ｊ　。
ＣＩｉｎ、　Ｅ　ｎｄｏｃｒｉｎｏｌ、　Ｍｅｔａｂ、
　、　；Ｑ　、　１０９８−１１０１　（１９７２））
に従い、大毛根を用い、テストステロンが５α−ＤＨＴ
に還元される量を測定し、エピカテキン、エピガロカテ
キン、エピカテキンガレートおよびエピガロカテキンガ
レートによるレダクターゼ阻害作用を検討し、表１に示
した。

２１７　（１９８１）　）に基づいて、皮脂線のレダク
ターゼを阻害するエピカテキン、エピガロカテキン、エ
ピカテキンガレートおよびエピガロカテキンガレートの
効果を表２に示した。
表２　ハムスター皮脂腺テストステロンー５α−レダク
ターゼ阻害率（％） 前記表１および表２において、人毛根、ハムスター皮脂
線におけるエピカテキン、エピガロカテキン、エピカテ
キンガレートおよびエピガロカテキンガレートのレダク
ターゼ阻害作用が明らかに認められた。また、個々のカ
テキンを任意の割合で混合しても効果が損なわれること
はなかった。
［発明の効果］ このように、エピカテキン、エピガロカテキン、エピカ
テキンガレートおよびエピガロカテキンガレートはテス
トステロン−５α−レダクターゼ阻害作用を有する安全
性の高い物質であり、長期かつ継続的に外用できるとい
う極めて有用な特徴を有している。

    トおよびエピガロカテキンガレートの一種又は二種以上
