本明細書で使用される「抗原結合タンパク質」という用語は、1つ又は複数の標的抗原に特異的に結合するタンパク質を指す。抗原結合タンパク質には、抗体及びその機能性断片が含まれ得る。「機能性抗体断片」は、全長の重鎖及び/又は軽鎖に存在するアミノ酸の少なくともいくつかを欠くが、抗原に特異的に結合する能力を依然として有する、抗体の一部分を指す。機能性抗体断片には、限定はされないが、Fab断片、Fab’断片、F(ab’)2断片、Fv断片、Fd断片、及び相補性決定領域(CDR)断片が含まれ、機能性抗体断片は、ヒト、マウス、ラット、ウサギ、又はラクダ科の動物などの任意の哺乳類源に由来し得る。機能性抗体断片は、標的抗原への結合で未変化の抗体と競合し得るものであり、断片は、未変化の抗体の改変(例えば、酵素的切断もしくは化学的切断)によって産生させるか、又は組換えDNA技術もしくはペプチド合成を使用してデノボ合成してよい。

Claims (7)
- 二重特異性の4価抗原結合タンパク質であって、
a)第1の重鎖可変領域(VH1)及び第1のCH1ドメインを含む第1の抗体の第1の重鎖を含む第1のポリペプチドであって、前記第1の抗体が、第1の抗原に特異的に結合し、第2の抗体の第2の重鎖可変領域(VH2)を含むポリペプチドのN末端に前記第1の重鎖がそのC末端を介して融合され、第2のCH1ドメインのN末端に前記VH2がそのC末端を介して融合され、前記第2の抗体が第2の抗原に特異的に結合し、
i)前記VH1又は第1のCH1ドメインが、EUの番号付けを使用した位置39、位置44、及び位置183からなる群から選択される残基に正に荷電したアミノ酸を導入するためのアミノ酸置換を少なくとも1つ含み、かつ
ii)前記VH2又は第2のCH1ドメインが、EUの番号付けを使用した位置39、位置44、及び位置183に対応する残基からなる群から選択される残基に負に荷電したアミノ酸を導入するためのアミノ酸置換を少なくとも1つ含む、
前記第1のポリペプチドと、
b)a)に記載の第1の抗体の第1の軽鎖を含む第2のポリペプチドであって、前記第1の軽鎖が、第1の軽鎖可変領域(VL1)及び第1のCL領域を含み、前記VL1又は第1のCLドメインが、EUの番号付けを使用した位置38、位置100、及び位置176からなる群から選択される残基に負に荷電したアミノ酸を導入するためのアミノ酸置換を少なくとも1つ含む、
前記第2のポリペプチドと、
c)a)に記載の第2の抗体の第2の軽鎖を含む第3のポリペプチドであって、前記第2の軽鎖が、第2の軽鎖可変領域(VL2)及び第2のCL領域を含み、前記VL1又は第2のCLドメインが、EUの番号付けを使用した位置38、位置100、及び位置176からなる群から選択される残基に正に荷電したアミノ酸を導入するためのアミノ酸置換を少なくとも1つ含む、
前記第3のポリペプチドと、
を含む、抗原結合タンパク質。 - 前記第1の重鎖が、ペプチドリンカーを介して前記VH2に融合される、請求項1に記載の抗原結合タンパク質。
- 前記ペプチドリンカーが、(Gly3Ser)2、(Gly4Ser)2、(Gly3Ser)3、(Gly4Ser)3、(Gly3Ser)4、(Gly4Ser)4、(Gly3Ser)5、(Gly4Ser)5、(Gly3Ser)6、及び(Gly4Ser)6からなる群から選択される配列を含む、請求項2に記載の抗原結合タンパク質。
- a)前記VH1又は第1のCH1ドメインが、EUの番号付けを使用したQ39K、G44K、及びS183Kからなる群から選択される変異を含み、
b)前記VH2又は第2のCH1ドメインが、EUの番号付けを使用したQ39E、G44E、及びS183Eからなる群から選択される変異を含み、
c)前記VL1又は第1のCLドメインが、EUの番号付けを使用したQ38E、G100E、及びS176Eからなる群から選択される変異を含み、かつ
d)前記VL2又は第2のCLドメインが、EUの番号付けを使用したQ38K、G100K、及びS176Kからなる群から選択される変異を含む、
請求項1に記載の抗原結合タンパク質。 - a)前記第1のCH1ドメインが、EUの番号付けを使用したS183K変異を含み、
b)前記第2のCH1ドメインが、EUの番号付けを使用したS183E変異を含み、
c)前記第1のCLドメインが、EUの番号付けを使用したS176E変異を含み、かつ
d)前記第2のCLドメインが、EUの番号付けを使用したS176K変異を含む、
請求項4に記載の抗原結合タンパク質。 - a)前記VH1が、EUの番号付けを使用したQ39K変異を含み、前記第1のCH1ドメインが、EUの番号付けを使用したS183K変異を含み、
b)前記VH2が、EUの番号付けを使用したQ39E変異を含み、前記第2のCH1ドメインが、EUの番号付けを使用したS183E変異を含み、
c)前記VL1が、EUの番号付けを使用したQ38E変異を含み、前記第1のCLドメインが、EUの番号付けを使用したS176E変異を含み、かつ
d)前記VL2が、EUの番号付けを使用したQ38K変異を含み、前記第2のCLドメインが、EUの番号付けを使用したS176K変異を含む、
