Description
1)ムレスズメの根に抽出溶媒を加えて抽出するステップ;
2)ステップ1)の抽出物をろ過するステップ;
3)ステップ2)でろ過した抽出物を減圧濃縮した後、乾燥するステップ;及び
4)ステップ3)で乾燥した抽出物に有機溶媒を加えてムレスズメ根の分画物を製造するステップ。
(a)ムレスズメ根の粉砕物を水、C1〜C4の低級アルコール、又はこれらの混合溶媒で抽出するステップ;
(b)前記抽出物をヘキサン、クロロホルム、酢酸エチル、及びブタノールからなる群から選択されるいずれかの有機溶媒で分画するステップ;及び
(c)前記分画物からα−ビニフェリンを分離するステップ。
細切して陰干ししたムレスズメの根500gを粉砕して粉砕物を製造した。前記粉砕物に70%のエタノール5lを加えて5時間還流抽出して冷浸した。前記抽出物を400メッシュ(mesh)の濾布でろ過した後、濾紙No.2(Whatman)で再びろ過した。その後、減圧濃縮し、ムレスズメ根エタノール抽出物24gを得た。
前記実施例1で得られたムレスズメ根エタノール抽出物10gをヘキサンとクロロホルム、酢酸エチル、及びブタノールを用いて、通常の方法で溶媒分画を行った後、減圧濃縮して、実施例2のヘキサン分画物2.12g、実施例3のクロロホルム分画物1.32g、実施例4の酢酸エチル分画物1.24g、実施例5のブタノール分画物0.82gを得た。
前記実施例2〜5で得られたムレスズメ根抽出物の分画物0.1gをそれぞれメタノールに溶解し、10ml検液を得た。また、α−ビニフェリン(C42H30O9:678.68)標準品10mgをメタノールで100mlになるように溶解した後、このうち0.5mlを、メタノールを用いて100mlになるように再び溶解し、1/2、1/4に希釈して標準液を得た。前記検液及び標準液をそれぞれ20μlずつHPLC分析し、α−ビニフェリン含有量を測定した。
実験例2−1:ムレスズメ根抽出物及びその分画物のチロシナーゼ活性阻害効果の確認
チロシナーゼは、生体内でチロシン(tyrosine)という物質の酸化過程を促進し、メラニンを生成させる酵素である。そのため、前記実施例1〜5で製造されたムレスズメ根エタノール抽出物及びその分画物のチロシナーゼ活性阻害効果を確認することにした。
前記実施例1〜5で製造されたムレスズメ根エタノール抽出物及びその分画物の美白効果を調べるために、B16F1メラニン形成細胞に対するメラニン生成抑制効果を確認することにした。
前記実施例4のムレスズメ根の分画物から分離したα−ビニフェリンの美白効果を調べるために、B16F1メラニン形成細胞に対するメラニン生成抑制効果を確認することにした。
ムレスズメ根抽出物の分画物を有効成分として含有する化粧料組成物の安定性を調べるために、ブチレングリコールと精製水が50:50の割合で混合された溶媒に、前記実施例4のムレスズメ根抽出物の酢酸エチル分画物を溶解した後、下記[表4]の通り可溶化剤形を製造した。
(1)水相と可溶化相をそれぞれ均一に混合及び溶解した。
(2)水相に可溶化相を入れて混合及び可溶化した。
(3)添加Iは実施例4のムレスズメ根抽出物の酢酸エチル分画物を含有する相に可溶化した後、投入して混合し、添加IIを混合して完了した。
ムレスズメ根抽出物の分画物を有効成分として含有する化粧料組成物の安定性を調べるために、ブチレングリコールと精製水が50:50の割合で混合された溶媒に、前記実施例4のムレスズメ根抽出物の酢酸エチル分画物を溶解した後、下記[表5]の通りエッセンス剤形を製造した。
(1)水相と油相を加温して、それぞれ均一に混合及び溶解した。
(2)75℃で水相に油相を入れて混合し乳化した
(3)添加相Iは実施例4のムレスズメ根抽出物の酢酸エチル分画物を有効成分として含有する相で、50℃で投入して混合した後、添加相IIを混合した。
ムレスズメ根抽出物の分画物を有効成分として含有する化粧料組成物の安定性を調べるために、ブチレングリコールと精製水が50:50の割合で混合された溶媒に、前記実施例4のムレスズメ根抽出物の酢酸エチル分画物を溶解した後、下記[表6]の通りクリーム剤形を製造した。
(1)水相の油相を加温して、それぞれ均一に混合及び溶解した。
(2)75℃で水相に油相を入れて混合し乳化した。
(3)添加相Iは実施例4のムレスズメ根抽出物の酢酸エチル分画物を有効成分として含有する相で、50℃で投入して混合した後、添加相IIを混合した。
実験例3−1:ムレスズメ根抽出物の分画物を含有する化粧料組成物の原料安定性の確認
原料の安定性を確認するために、前記剤形例1〜3の可溶化剤形、エッセンス剤形、クリーム剤形を4℃、30℃、45℃、日光の条件下において12週間保管した後、前記実験例1と同様の方法でHPLC分析により、ムレスズメ根抽出物の酢酸エチル分画物に含有されているα−ビニフェリン成分の経時変化を観察し、その結果を[表7]に示した。
剤形安定性を確認するために、前記剤形例1〜3の可溶化剤形、エッセンス剤形、クリーム剤形を4℃、30℃、45℃、日光の条件下において12週間保管した後、肉眼的評価と官能評価により、色、匂い、剤形の変化を観察し、その結果を下記[表8]〜[表10]に示した。このとき、色、匂い、剤形の変化の程度は、下記評価基準に基づいて分類し評価した。

α−ビニフェリンの安定性を確認するために、前記実験例1のムレスズメ根エタノール抽出物の酢酸エチル分画物から分離したα−ビニフェリンを50℃で2週間保管した後、前記実験例1と同様の方法でHPLC分析により、含有量の変化を観察した。対照群としては、ムレスズメ根抽出物から分離したコボフェノールAを用い、α−ビニフェリンと同じ条件で保管した後、同様の方法で含有量の変化を観察した。その結果を下記[表11]に示した。
ムレスズメ根抽出物の分画物を含有する化粧料組成物の皮膚刺激性を調べるために、前記剤形例3のクリーム剤形の皮膚刺激性評価を行った。

ムレスズメ根抽出物の分画物を含有する化粧料組成物の有効性を評価するために、平均年齢44歳の女性23人に対して臨床実験を行った。試験製品として、ムレスズメ根抽出物の分画物が3.2%含有された化粧料組成物を用い、対照製品としては、試験製品でムレスズメ根抽出物の分画物の代わりに精製水を含有するプラセボクリームを用いた。前記試験製品と対照製品をそれぞれ被験者の顔の左側と右側にランダムに塗布し、1日2回のペースで塗布しながら8週間その変化を観察した。
試験製品及び対照製品の使用による皮膚の明るさの変化を調べるために、使用前、使用4週後、使用6週後、使用8週後の皮膚の明るさを皮膚色測定器であるChroma meter CM700dを用いて測定した。測定の際には、毎回同じ部位の皮膚を3回ずつ測定し、その平均値を評価資料として使用した。
試験製品と対照製品の使用による皮膚の明るさの変化を調べるために、使用前、使用4週後、使用6週後、使用8週後の皮膚のメラニン量をMexameterというNarrow−band reflectance spectrophotometerでprobeで568nm(green)、660nm(red)、880nm(infrared)での反射光を利用して測定した。測定の際には毎回同じ部位のメラニン量を3回ずつ測定し、その平均値を評価資料として使用した。
試験製品と対照製品の使用による皮膚の明るさの変化を調べるために、使用前、使用4週後、使用6週後、使用8週後の皮膚を肉眼的に評価した。

- ムレスズメ(Caragana Sinica)根のエタノール抽出物の酢酸エチル分画物を有効成分として含有し、
前記分画物は、組成物の総重量に対して0.01重量%〜10重量%で含有される、
美白用化粧料組成物。 - 前記分画物は、分画物の総重量に対して0.1重量%〜1.0重量%でα−ビニフェリンを含有することを特徴とする、請求項1に記載の美白用化粧料組成物。
