Description
本出願は、2013年3月14日出願の米国仮出願番号61/785,038の利益を主張する。上記出願は、参照により本明細書に組み込まれる。
吸着剤:吸着剤は、クロマトグラフィーを実施するために使用される、固相担体に付着した少なくとも1個の分子であるか、またはそれ自体が固相である少なくとも1個の分子である。
本発明は、タンパク質に結合する第2のタンパク質を含有する試料からの組換えタンパク質の精製方法を提供する。本方法は、組換えタンパク質が触手状陰イオン交換マトリックスクロマトグラフィー媒体に結合するような条件下で、試料を触手状陰イオン交換マトリックスクロマトグラフィー媒体に供し、続いて溶離液中でクロマトグラフィー媒体に結合した組換えタンパク質を溶離することを伴う。本発明の方法を使用して、本発明の発明者らは、第2の汚染タンパク質の少なくとも75%を除去しながら、溶離液中の組換えタンパク質の少なくとも85%を回収することができた。実際には、本発明の方法は、多くの条件において、第2の汚染タンパク質の少なくとも80%を除去すると同時に、溶離液中の組換えタンパク質の少なくとも90%を回収することができるようなものであった。そのような優れた結果は、工業生産において典型的に使用されるいかなる他の陰イオン、陽イオン、または疎水性相互作用クロマトグラフィーでも不可能であった。特定の機序に縛られることを望むものではないが、陰イオン交換マトリックスクロマトグラフィー媒体の触手状の側面が、本発明の方法に、任意の他の種類の陰イオン交換樹脂または実際には大半の樹脂で観測されるよりも予想外に優れた結果をもたらすと考えられる。
Claims (17)
- 組換えタンパク質と前記タンパク質に結合する第2のタンパク質とを含む試料から前記組換えタンパク質を精製するための方法であって、前記組換えタンパク質が触手状(tentacle)陰イオン交換マトリックスクロマトグラフィー媒体に結合するような条件下で前記試料を前記触手状陰イオン交換マトリックスクロマトグラフィー媒体に供し、続いて溶離液中で前記クロマトグラフィー媒体に結合した前記組換えタンパク質を溶離することを含み、それによって前記組換えタンパク質の少なくとも85%が前記溶離液中で回収され、前記第2のタンパク質の少なくとも75%が前記溶離液から除去され、前記第2のタンパク質がプロテインA、プロテインGあるいはプロテインLG、あるいはIgG抗体のF C 部位に結合する能力を有するそれらの組換え形態であり、および前記組換えタンパク質が腫瘍壊死因子受容体Fc融合タンパク質である、方法。
- 前記触手状陰イオン交換マトリックスクロマトグラフィー媒体が強陰イオン官能基を含有する、請求項1に記載の方法。
- 前記強陰イオン官能基がトリメチル−アンモニウムエチル(TMAE)である、請求項2に記載の方法。
- 前記触手状陰イオン交換マトリックスクロマトグラフィー媒体の樹脂基質が、メタクリレートポリマー樹脂またはポリビニルスチレンポリマー樹脂である、請求項1〜3のいずれか一項に記載の方法。
- 前記クロマトグラフィー媒体が、Fractogel(登録商標)EMD TMAE HiCapである、請求項1〜4のいずれか一項に記載の方法。
- 前記第2のタンパク質が、プロテインAまたはプロテインGである、請求項1に記載の方法。
- 前記試料が、プロテインAクロマトグラフィー媒体を介して前記タンパク質のアフィニティ精製から獲得される、請求項1〜6のいずれか一項に記載の方法。
- 前記タンパク質がエタネルセプトである、請求項1に記載の方法。
- 前記試料が、約pH8で前記触手状陰イオン交換マトリックスクロマトグラフィー媒体に供される、請求項8に記載の方法。
- 前記組換えタンパク質が前記触手状陰イオン交換マトリックスクロマトグラフィー媒体に結合された後、かつ前記組換えタンパク質が溶離される前に、前記触手状陰イオン交換マトリックスクロマトグラフィー媒体が洗浄工程に供される、請求項1〜9のいずれか一項に記載の方法。
- 前記洗浄工程が、前記触手状陰イオン交換マトリックスクロマトグラフィー媒体を約pH8の緩衝液で洗浄することを含む、請求項10に記載の方法。
- 前記緩衝液が、約pH8の25mMトリス(ヒドロキシメチル)アミノメタン(トリス)から本質的になる、請求項11に記載の方法。
- 前記組換えタンパク質が、約7.2〜約7.5のpHの溶離緩衝剤中で前記触手状陰イオン交換マトリックスクロマトグラフィー媒体から溶離される、請求項1〜12のいずれか一項に記載の方法。
- 前記溶離緩衝剤が25mMトリスHCl、pH7.2である、請求項13に記載の方法。
- 前記溶離緩衝剤が25mMトリスHCl、pH7.5、及び約150mM〜約200mMのNaClである、請求項13に記載の方法。
- 前記組換えタンパク質が、前記触手状陰イオン交換マトリックスクロマトグラフィーの前または後にさらなる精製工程に供される、請求項1〜15のいずれか一項に記載の方法。
- 前記組換えタンパク質が薬学的組成物に製剤化される、請求項1〜16のいずれか一項に記載の方法。
