Description
(1)焼酎廃水を培地として使用する、スクワレン産生藻類の培養方法。
(2)前記スクワレン産生藻類が従属栄養性藻類である、(1)に記載の培養方法。
(3)前記スクワレン産生藻類が、オーランチオキトリウム(Aurantiochytrium)属の株である、(1)又は(2)のいずれかに記載の培養方法。
(4)前記スクワレン産生藻類が、オーランチオキトリウムsp.(Aurantiochytrium sp.)18W−13aである、(1)〜(3)のいずれか1項に記載の培養方法。
(5)前記焼酎廃水が、遠心分離した焼酎粕を濾過することにより取得される、(1)〜(4)のいずれか1項に記載の培養方法。
(6)前記焼酎廃水に、糖質、有機酸、無機酸、有機塩基、無機塩基、ビタミン、アミノ酸、ペプチド、タンパク質、ミネラルのいずれか1つ以上が添加される、(1)〜(5)のいずれか1項に記載の培養方法。
(7)前記焼酎廃水に、NaCl、KCl、CaCl2・H2O、MgSO4・7H2O及びMgCl2・6H2Oの1つ以上の塩類が、合計で5〜34‰(g/培地L)の量で添加される、(1)〜(6)のいずれか1項に記載の培養方法。
(8)前記塩類の添加量が合計で7〜10‰(g/培地L)である、(7)に記載の培養方法。
(9)(1)〜(8)のいずれか1項に記載の培養方法で培養したスクワレン産生藻類を回収し、当該回収されたスクワレン産生藻類からスクワレンを抽出する、スクワレン製造方法。
(10)焼酎廃水を(1)〜(8)に記載の培養方法で培地として使用することによる、焼酎廃水を浄化する方法。
本実験ではスクワレン産生藻類としてオーランチオキトリウムsp. 18W−13aを用いた。当該株は、焼酎廃水中の栄養分を同化してスクワレンを産生する。焼酎廃水は、霧島酒造から提供されたものを用いた。
霧島酒造から提供された焼酎廃水を、5000rpm、15分で遠心分離後、No.2の濾紙で濾過して、清澄な焼酎廃水を得た。当該廃水を、蒸留水で25%、50%、75%に希釈した。当該希釈廃水及び希釈をしていない廃水に、主要塩類を25%海水相当の塩分(8.5‰)となるように添加した(NaCl 5.85 g/L, KCl 0.19 g/L, CaCl2・H2O 0.37 g/L, MgSO4・7H2O 1.23 g/L, MgCl2・6H2O 1.02 g/L)。これらを、焼酎廃水の希釈系列とした。
焼酎廃水以外に、大豆廃水を培地とした場合のオーランチオキトリウムsp. 18W−13aの増殖及びスクワレン産生能力の評価を行った。大豆廃水は日本全国またはアジア圏で広く入手しやすく、炭素源や窒素源が豊富に含まれるため、培地源として適当であると思われた。また、焼酎廃水同様、これを有効利用する方法及び効率的に処理する方法の開発が求められている。
焼酎廃水以外に、オイルパーム廃液を培地とした場合のオーランチオキトリウムsp. 18W−13aの増殖及びスクワレン産生能力の評価を行った。オイルパーム廃液は日本では入手困難だが、東南アジア圏においてその処理が大問題となっている食品廃水であるため、これを有効利用する方法及び効率的に処理する方法の開発が求められている。
スクワレン産生株は、焼酎廃水中で増殖し、スクワレンを産生する過程で、焼酎廃水中の有機物を同化する。本実験において、スクワレン産生株オーランチオキトリウムsp. 18W−13aの培養の過程における、DOC(Dissolved organic carbon、溶存態有機炭素)、DTN(Dissolved total nitrogen、溶存態全窒素)及びDTP(Dissolved total phosphorus、溶存態全リン)を測定して、当該培養による焼酎廃水中の有機物除去作用の評価を行った。
Claims (10)
- 焼酎廃水を培地として使用する、スクワレン産生藻類の培養方法。
- 前記スクワレン産生藻類が従属栄養性藻類である、請求項1に記載の培養方法。
- 前記スクワレン産生藻類が、オーランチオキトリウム(Aurantiochytrium)属の株である、請求項1又は2のいずれかに記載の培養方法。
- 前記スクワレン産生藻類が、オーランチオキトリウムsp.(Aurantiochytrium sp.)18W-13aである、請求項3に記載の培養方法。
- 前記焼酎廃水が、遠心分離及び/又は濾過により清澄される、請求項1〜4のいずれか1項に記載の培養方法。
- 前記焼酎廃水に、糖質、有機酸、無機酸、有機塩基、無機塩基、ビタミン、アミノ酸、ペプチド、タンパク質、ミネラルのいずれか1つ以上が添加される、請求項1〜5のいずれか1項に記載の培養方法。
- 前記焼酎廃水に、NaCl、KCl、CaCl2・H2O、MgSO4・7H2O及びMgCl2・6H2Oの1つ以上の塩類が、合計で5〜34‰(g/培地L)の量で添加される、請求項1〜6のいずれか1項に記載の培養方法。
- 前記塩類の添加量が合計で7〜10‰(g/培地L)である、請求項7に記載の培養方法。
- 請求項1〜8のいずれか1項に記載の培養方法で培養したスクワレン産生藻類を回収し、当該回収されたスクワレン産生藻類からスクワレンを抽出する、スクワレン製造方法。
- 焼酎排水を請求項1〜8のいずれか1項に記載の培養方法で藻類の培地として使用することによる、焼酎排水を浄化する方法。
