Description
人は、食物用途およびエネルギー用途のためにトウモロコシ(corn)を栽培する。人は、ダイズおよびワタを含む他の多くの作物も栽培する。昆虫は、植物を食べて損傷し、それによってこれらの人の努力を台無しにする。害虫(insect pest)を防除する(control)ために毎年何10億ドルも費やされ、害虫が与える損傷に対してさらに数10億ドルが失われる。合成有機化学殺虫剤は、害虫を防除するために用いられる主な手段であったが、バチルス・チューリンゲンシス(Bacillus thuringiensis)(Bt)に由来する殺虫性タンパク質(insecticidal protein)などの生物殺虫剤(biological insecticide)が一部の領域で重要な役割を演じてきた。Bt殺虫性タンパク質遺伝子による形質転換を通して昆虫抵抗性植物(insect-resistant plant)を作出できたことにより、現代農業が革新し、殺虫性タンパク質およびその遺伝子の重要性および価値が高まっている。
本発明は、Cry1Caタンパク質と組み合わせたVip3Abタンパク質の使用に一部関する。これらのタンパク質の両方とも生成する植物(およびそのような植物を植える地所(acreage))は、本発明の範囲内に含まれる。
本発明は、Vip3AbおよびCry1Caがフォールアーミーワーム(FAW;スポドプテラ・フルジペルダ(Spodoptera frugiperda))の腸において結合のために互いに競合しないという意外な発見に一部関する。したがって、これらのタンパク質のいずれか単独への抵抗性をFAWが発達させることを遅らせるか阻止するために、Vip3Abタンパク質は、トランスジェニックトウモロコシ(および他の植物、例えばワタおよびダイズ)でCry1Caタンパク質と組み合わせて用いることができる。Cry抵抗性のフォールアーミーワームによる傷害から植物(メイズ植物(maize plant)および/またはダイズ植物など)を保護することにおいて、対象のタンパク質の組は効果を発揮することができる。すなわち、本発明の1つの用途は、Vip3AbまたはCry1Caへの抵抗性を発達させるかもしれないフォールアーミーワーム集団に起因する損害および収量減からトウモロコシおよび他の経済的に重要な植物種を保護することである。

Cry1CaおよびVip3Ab1プロトキシンをコードする遺伝子を蛍光菌(Pseudomonas fluorescens)発現株で発現させ、完全長タンパク質は不溶性封入体として単離された。洗浄した封入体は、20mM CAPS緩衝液、pH11、+10mM DDT、+0.1%2−メルカプトエタノールを含む緩衝液で37℃において2時間撹拌することによって可溶化した。溶液を37℃で10分間の27,000×gで遠心分離し、上清を0.5%(w/v)TCPK処理トリプシン(Sigma)で処理した。この溶液を室温でさらなる1時間混合しながらインキュベートし、濾過し、次に20mM CAPS、pH10.5で平衡させたPharmacia Mono Q 1010カラムにロードした。ロードしたカラムを2カラム容量の緩衝液で洗浄した後に、15カラム容量の20mM CAPS中の0〜0.5MのNaClの直線濃度勾配を用いて、切断された毒素を1.0ml/分の流速で溶出させた。精製されたトリプシン切断Cryタンパク質は、約0.2〜0.3MのNaClで溶出した。タンパク質の純度は、SDS PAGEおよびクーマシーブリリアントブルー色素を用いる可視化によって検査した。場合によっては、精製された毒素の合わせた分画を濃縮してSuperose 6カラム(直径1.6cm、長さ60cm)にロードし、サイズ排除クロマトグラフィーによってさらに精製した。モノマーの分子量の単一のピークを含む分画を合わせ、濃縮して、約60,000kDaの分子量を有するタンパク質の95%を超えて均質である調製物を生成した。
Cry1Caは従来の方法を用いて放射標識するのが非常に困難であることを以前の研究は示したが、選定された数例ではそれは受容体結合アッセイで放射標識され、良好に機能するであろう。我々は、Cry1Fa(Prov.69919)を活発に放射標識する働きをした方法である、125I放射標識フルオレセイン−5−マレイミドを用いてCry1Caを放射標識することに決めた。フルオレセイン−5−マレミドのヨウ素化、およびCry1Caとのこの放射標識化学物質の以降のコンジュゲーションは、タンパク質のシステイン特異的放射標識をもたらす。したがって、そのような標識化手法は、標識される残基で高度に特異的である。Cry1Caコア毒素セグメント(残基29〜619)は、210位および438位に2つのシステインアミノ酸残基を含む。Palmer et al. (1997)は、フルオレセイン−5−マレイミドのフェニル環を放射ヨウ素標識し、次にスルフヒドリル基(例えば遊離システイン残基によって提供される)を含むタンパク質と反応させ、タンパク質の遊離システインのアルキル化をもたらし、したがって放射能標識タンパク質を提供することができることを実証した。トリプシン切断されたCry1Caコア毒素は2つのシステイン残基を含み、したがってこれらの2つの(特異的な)部位でのタンパク質のアルキル化および放射標識のための基質を提供する。
相同的および非相同的競合結合アッセイは、150μg/mlのBBMVタンパク質および2nMの125I放射性標識Cry1Caコア毒素タンパク質を用いて実施した。反応混合液に加えられた相同的競合的非放射性標識Cry1Caコア毒素タンパク質の濃度は0.1、1、10、100および1000nMであった。非相同的トリプシン切断Vip3Abタンパク質を10および1,000nMで試験し、真の結合競合を保証するために放射性Cry1Caコア毒素タンパク質と同時にタンパク質を加えた。インキュベーションを28℃で1時間実行し、BBMVに未結合の(すなわち、昆虫受容体タンパク質に結合していない)125I標識Cry1Caコア毒素タンパク質の量を、16,000×gで8分間のBBMV混合物の遠心、および生じたペレットから上清を除去することによって結合したタンパク質から分離する。ペレットを氷冷結合緩衝液(PBS;11.9mM Na2HPO4、137mM NaCl、2.7mM KCl、pH7.4プラス0.1%ウシ血清アルブミン;Sigma−Aldrich、St.Louis、MO)で3回洗浄して、残りのいかなる未結合の125I標識Cry1Caを完全に除去する。遠心管の底を切り取り、この部分に含まれるタンパク質ペレットを13×100 mmガラス培養管に入れ、ガンマ計数器で10分間数えてペレット分画中に含まれる結合した放射活性の量を得た。結合したタンパク質分画中の放射活性量は、昆虫受容体に結合したCryタンパク質の量の指標となる(全結合)。1,000nMの非放射標識Cry1Caコア毒素タンパク質の存在下でペレットで得られたカウント数によって、非特異的結合を表した。BBMVに特異的に結合した(特異的な結合)放射標識Cry1Caの量は、全結合レベルを非特異的な結合から引くことで測定された。100パーセントの全結合は、いかなる競合者Cry1Faコア毒素タンパク質もない場合の結合の量であるとみなされた。データは、競合的非標識リガンドの濃度に対する特異的結合125I Cry1Caの百分率で表す。
結果(図1)は、相同的非標識Cry1Caタンパク質が、BBMVタンパク質への特異的結合から放射標識Cry1Caコア毒素タンパク質を用量依存的に効果的に追い出したことを示す。Vip3Abは、示す濃度(10または1,000nM)のいずれでも、結合した125I標識Cry1Caコア毒素タンパク質をその受容体タンパク質(複数可)から追い出さなかった。試験されたVip3Abの最も高い濃度(1,000nM)は、アッセイで用いられた放射標識Cry1Caより500倍高い濃度に相当し、Vip3AbがS.フルジペルダ(S. frugiperda)BBMVで放射標識Cry1Caの結合と効果的に競合しないことを実証する。
本発明は、以下の態様を包含する。

前記第三のタンパク質が、Cry1FaおよびCry1Beからなる群から選択され、前記植物が、Cry2A、Cry1I、DIG−3およびCry1Abからなる群から選択される第四および第五の殺虫性タンパク質をコードするDNAをさらに含む、上記[2]に記載のトランスジェニック植物。
