Description
本発明は、酸化再生セルロース(ORC)などの陰イオン性ポリサッカライドと銀との間で形成される錯体を含む創傷被覆材(wound dressing material)、および、その製造法および創傷治療への使用法に関する。
アルギネート、ヒアルロン酸およびその塩、および、酸化再生セルロース(ORC)などの酸化セルロースなどの陰イオン性ポリサッカライドは、創傷被覆材における使用が既知である。アルギネートおよびORCは、創傷に適用されると、止血性があり、ORCは、皮膚潰瘍などの慢性創傷の治癒を促進することが立証されてきた。
a)陰イオン性ポリサッカライドを銀塩溶液で処理し、当該陰イオン性ポリサッカライドと銀との錯体を生成する段階と、
b)前記錯体を水性アスコルビン酸に分散させ、酸性化分散物(acidified dispersion)を形成する段階と、続いて、
c)前記分散物を凍結乾燥または溶媒乾燥し、スポンジ材を形成する段階と、
を含む、調製法に関する。
銀とのORCの錯体は、以下のとおりに調製された。
本発明に従った創傷被覆剤としての使用に適した凍結乾燥コラーゲン/ORCスポンジは、次のように調製された。
実施例1の方法が繰り返されたが、国際公開第2004/024197号に記述されているように、前記スラリーの酸性化には、酢酸が使用された。生じたスポンジ材は、柔らかく柔軟であったが、昼光に曝されると、ピンク色に変色した。
実施例1の方法が繰り返されたが、前記スラリーの酸性化には、クエン酸が使用された。生じたスポンジ材は、比較的硬質で、もろく、自然の昼光に曝されると、赤色/褐色に変色した。
実施例1の方法が繰り返されたが、前記スラリーの酸性化には、乳酸が使用された。生じたスポンジ材は、比較的稠密で、もろく、自然の昼光に曝されると、安定したオフホワイト(クリーム)色を発した。
上記実施例からの本被覆材の抗増殖作用が、次のとおりに、国際公開第2004/024197号に記述されている方法によって評価された。
(1) 医薬用抗菌スポンジ材の調製方法において、
陰イオンポリサッカライドを銀塩溶液で処理し、前記陰イオン性ポリサッカライドと銀との錯体を生成する段階と、
前記錯体を水性アスコルビン酸に分散させ、酸性化分散物を形成する段階と、
続いて、前記分散物を凍結乾燥または溶媒乾燥し、前記スポンジ材を形成する段階と、
を含む、方法。
(2) 実施態様1に記載の方法において、
前記イオン性ポリサッカライドが酸化再生セルロース(ORC)から本質的に成る、
方法。
(3) 実施態様1または2に記載の方法において、
前記分散物が、前記分散物の中に分散された1種類以上の追加の医学的に許容可能なポリマー材料をさらに含む、
方法。
(4) 実施態様3に記載の方法において、
前記分散物が、実質的に銀を含まないORC、および、コラーゲンをさらに含み、
前記分散物中の銀濃度が、総ポリマー固体量に基づいて0.1重量%〜2重量%である、
方法。
(5) 実施態様1ないし4のいずれかに記載の方法において、
前記分散物が、約3〜約4のpH、および、約0.5%〜約2%の固体濃度を有する、
方法。
(6) 光安定化抗菌スポンジ材(photostabilized antimicrobial sponge material)において、
実施態様1〜5のいずれかに記載の方法によって入手される、または、入手可能な、光安定化抗菌スポンジ材。
(7) 光安定化抗菌スポンジ材において、
銀(I)イオンと錯体形成された陰イオン性ポリサッカライド、
を含み、 前記スポンジは、アスコルベートをさらに含み、かつ、実質的に白色であり、
この色は、25℃で24時間のあいだ空気中で自然の昼光に曝露された時に、目視可能な変色に対して実質的に安定である、
光安定化抗菌スポンジ材。
(8) 創傷被覆材において、
実施態様6または7に記載の抗菌スポンジ材、
を含む、創傷被覆材。
(9) 実施態様8に記載の創傷被覆材において、
前記創傷被覆材が、無菌状態であり、微生物不透過性の容器中に包装される、
創傷被覆材。
(10) 抗菌材料へのアスコルビン酸の使用において、
前記抗菌材料は、光曝露時の変色に対して前記材料を安定化させるために、医学的に許容可能な多価陰イオン性酸の銀(I)塩を含む、
使用。
Claims (9)
- 医薬用抗菌スポンジ材の調製方法において、
陰イオン性ポリサッカライドを銀塩溶液で処理し、前記陰イオン性ポリサッカライドと銀との錯体を生成する段階と、
前記錯体を水性アスコルビン酸に分散させ、酸性化分散物を形成する段階と、
続いて、前記分散物を凍結乾燥または溶媒乾燥し、前記スポンジ材を形成する段階と、
を含み、
前記陰イオン性ポリサッカライドが、酸化セルロースを含む、方法。 - 請求項1に記載の方法において、
前記イオン性ポリサッカライドが、酸化再生セルロース(ORC)から成る、
方法。 - 請求項1または2に記載の方法において、
前記分散物が、前記分散物の中に分散された1種類以上の追加の医学的に許容可能なポリマー材料をさらに含む、
方法。 - 請求項3に記載の方法において、
前記分散物が、銀を含まないORC、および、コラーゲンをさらに含み、
前記分散物中の銀濃度が、総ポリマー固体量に基づいて0.1重量%〜2重量%である、
方法。 - 請求項1ないし4のいずれかに記載の方法において、
前記分散物が、3〜4のpH、および、0.5%〜2%の固体濃度を有する、
方法。 - 光安定化抗菌スポンジ材において、
請求項1ないし5のいずれかに記載の方法によって入手される、または、入手可能な、光安定化抗菌スポンジ材。 - 創傷被覆材において、
請求項6に記載の抗菌スポンジ材、
を含む、創傷被覆材。 - 請求項7に記載の創傷被覆材において、
前記創傷被覆材が、無菌状態であり、微生物不透過性の容器中に包装される、
創傷被覆材。 - 抗菌材料へのアスコルビン酸の使用において、
前記抗菌材料は、光曝露時の変色に対して前記材料を安定化させるために、医学的に許容可能な酸化セルロースの銀(I)塩を含む、
