Description
コラーゲン/ORC凍結乾燥スポンジ材料をベースとする抗酸化性の創傷用包帯材料を以下の通りに作製する。
メチレンブルー染料をクリスタルバイオレット(塩基性染料)に置き換えた以外は、実施例1と同様の手順を実施した。クリスタルバイオレットは、スラリー中に以下の濃度で取り入れられた:0%(参照例),1mg/mL,0.5mg/mLおよび0.1mg/mL。
メチレンブルー染料をフラビン3,6−ジアミノアクリジンヘミスルフェート(flavin 3,6-Diaminoacridine hemisulfate)(塩基性染料)に置き換えた以外は、実施例1と同様の手順を実施した。このフラビンは、スラリー中に以下の濃度で取り入れられた:0%(参照例),1mg/mL,0.5mg/mLおよび0.1mg/mL。

酢酸銀を0.05Mの酢酸に溶解し、全固体をベースにして1重量%の銀を含む最終スラリーを造るのに十分な量のこの溶液をORC/コラーゲンスラリーに加えることにより、実施例1〜7に記載されたようにして作製された創傷用包帯材料に銀を取り入れた。
酸素を含有するフリーラジカルと反応し、これを除去する創傷用包帯材料の能力は、WO94/13333(参照によりその全体が本明細書に明示的に包含される)に開示されたDPPHテストによって評価される。このテストは、Blois M.S., Nature,181:1199(1958年)および、Banda P.W.等,Analytical Letters,7:41(1974年)に記載された方法を本テストの状況に合わせ適用したものである。
実施例8〜14で作製したスポンジの殺菌作用を、緑膿菌および黄色ブドウ球菌について抑制域を観察することによってテストする。

(2) 実施態様1に記載の創傷用包帯材料であって、
前記基質が、コラーゲン、酸化セルロース、キトサン、ガラクトマンナン、グリコサミノグリカン、ポリラクチド/ポリグリコリド、および、これらの混合物からなる群から選ばれた生体吸収性の固体材料を含む、創傷用包帯材料。
(3) 実施態様2に記載の創傷用包帯材料であって、
前記基質が、コラーゲン、酸化再生セルロース、キトサン、および、これらの混合物からなる群から選ばれた生体吸収性の固体材料を含む、創傷用包帯材料。
(4) 実施態様1〜3のいずれかに記載の創傷用包帯材料であって、
前記生体吸収性の固体基質が、織物、編み物、不織布、凍結乾燥スポンジ、溶媒乾燥スポンジ、および、これらの組合せからなる群から選ばれたものである、創傷用包帯材料。
(5) 実施態様1〜4のいずれかに記載の創傷用包帯材料であって、
前記抗酸化性の染料が、アニリン染料(aniline dyes)、アクリジン染料(acridine dyes)、チオニン染料(thionine dyes)、ビス−ナフタレン染料(bis-naphthalene dyes)、チアジン染料(thiazine dyes)、アゾ染料(azo dyes)、アントラキノン(anthraquinones)、および、これらの混合物からなる群から選ばれたものである、創傷用包帯材料。
(6) 実施態様1〜5のいずれかに記載の創傷用包帯材料であって、
前記抗酸化性の染料が、ゲンチアナバイオレット(gentian violet)、アニリンブルー(aniline blue)、メチレンブルー(methylene blue)、クリスタルバイオレット(crystalviolet)、アクリフラビン(acriflavine)、9−アミノアクリジン(9-aminoacridine)、アクリジンイエロー(acridine yellow)、アクリジンオレンジ(acridine orange)、プロフラビン(proflavin)、キナクリン(quinacrine)、ブリリアントグリーン(brilliant green)、トリパンブルー(trypan blue,)、トリパンレッド(trypan red)、マラカイトグリーン(malachite green)、アザクリン(azacrine)、メチルバイオレット(methyl violet)、メチルオレンジ(methyl orange)、メチルイエロー(methyl yellow)、エチルバイオレット(ethyl violet)、アシッドオレンジ(acid orange)、アシッドイエロー(acid yellow)、アシッドブルー(acid blue)、アシッドレッド(acid red)、チオフラビン(thioflavin)、アルファズリン(alphazurine)、インディゴブルー(indigo blue)、メチレングリーン(methylene green)、および、これらの混合物からなる群から選ばれたものである、創傷用包帯材料。
(7) 実施態様1〜6のいずれかに記載の創傷用包帯材料であって、
前記抗酸化性の染料が、前記材料の乾燥重量に基づいて、0.2〜2重量%の量で存在する、創傷用包帯材料。
(8) 実施態様1〜7のいずれかに記載の創傷用包帯材料であって、
前記材料が銀塩をさらに含み、前記染料が当該銀塩を光安定化している、創傷用包帯材料。
(9) 実施態様8に記載の創傷用包帯材料であって、
前記基質がアニオン性ポリマーを含み、前記銀塩がAg+とアニオン性ポリマーとの塩を含む、創傷用包帯材料。
(10) 実施態様8または9に記載の創傷用包帯材料であって、
前記組成物が、当該組成物の乾燥重量に基づいて、0.01〜5重量%の銀を含む、創傷用包帯材料。
(11) 実施態様1〜10のいずれかに記載の創傷用包帯材料であって、
前記材料がシートの形態を有する、創傷用包帯材料。
(12) 実施態様1〜11のいずれかに記載の創傷用包帯材料であって、
前記材料が、殺菌され、微生物の侵入しない容器で包装されたものである、創傷用包帯材料。
(13) 実施態様1〜12のいずれかに記載の創傷用包帯材料であって、
前記材料が、本願明細書で定義した抗酸化活性についてのジフェニルピクリルヒドラジル(DPPH)検査で、少なくとも15%のフリーラジカル活性を有する、創傷用包帯材料。
(14) 実施態様1〜13のいずれかに記載の材料の使用であって、
創傷治療用薬剤の作製のための使用。
(15) 実施態様14に記載の使用であって、
前記創傷が慢性の創傷であり、好ましくは、静脈の潰瘍、褥瘡性潰瘍および糖尿病性潰瘍の諸潰瘍からなる群から選ばれた創傷である、使用。
(16) 抗酸化性の創傷用包帯材料の製造方法であって、
生体吸収性の基質材料を抗酸化性の染料で染めるステップを含む、製造方法。
(17) 実施態様16に記載の方法であって、
実施態様1〜13のいずれかに記載の創傷用包帯材料の製造のための、製造方法。
(18) 創傷用包帯であって、
実施態様1〜13のいずれかに記載の抗酸化性の創傷用包帯材料を含んでなる、創傷用包帯。
(19) 実施態様18に記載の創傷用包帯であって、
前記材料は、滅菌され、微生物の侵入しない容器で包装されたものである、創傷用包帯。
Claims (16)
- 創傷用包帯材料であって、
10 -4 Mのジフェニルピクリルヒドラジル(DPPH)中に多糖を分散した0.5%(w/v)の多糖分散体の4時間後の、524nmにおける吸光度の減少率として測定した抗酸化活性に関するDPPH検査で、少なくとも15%のフリーラジカル活性を有し、
生体吸収性の固体基質を含んでなり、当該基質が、凍結乾燥スポンジまたは溶媒乾燥スポンジであって、酸化再生セルロース、コラーゲン、キトサンおよびこれらの混合物からなる群から選ばれ、抗酸化活性に関するDPPH検査でフリーラジカル活性を有する抗酸化性の染料で染められている生体吸収性の固体材料を含む、
創傷用包帯材料。 - 請求項1に記載の創傷用包帯材料であって、
前記抗酸化性の染料が、アニリン染料(aniline dyes)、アクリジン染料(acridine dyes)、チオニン染料(thionine dyes)、ビス−ナフタレン染料(bis-naphthalene dyes)、チアジン染料(thiazine dyes)、アゾ染料(azo dyes)、アントラキノン(anthraquinones)、および、これらの混合物からなる群から選ばれたものである、創傷用包帯材料。 - 請求項1または2に記載の創傷用包帯材料であって、

前記基質がアニオン性ポリマーを含み、前記銀塩がAg+とアニオン性ポリマーとの塩を含む、創傷用包帯材料。 - 請求項5または6に記載の創傷用包帯材料であって、
前記創傷用包帯材料が、当該創傷用包帯材料の乾燥重量に基づいて、0.01〜5重量%の銀を含む、創傷用包帯材料。 - 請求項1〜7のいずれかに記載の創傷用包帯材料であって、
前記創傷用包帯材料がシートの形態を有する、創傷用包帯材料。 - 請求項1〜8のいずれかに記載の創傷用包帯材料であって、
前記創傷用包帯材料が、殺菌され、微生物の侵入しない容器で包装されたものである、創傷用包帯材料。 - 請求項1〜9のいずれかに記載の創傷用包帯材料の使用であって、
