Description
すなわち、本発明の医療用テープ剤は、支持体の片面に、粘着剤層と剥離ライナーが、順次積層されたテープ剤であって、該粘着剤層の粘着剤が、(A)メタクリル酸2−アセトアセトキシエチルエステル及びその他のビニルモノマーを構成成分とする共重合体を含む粘着性ポリマーA、(B)アルキル基の炭素原子数が4個〜10個の(メタ)アクリル酸アルキルエステルとその他のビニルモノマーを構成成分とし、前記粘着性ポリマーAとは異なる共重合体を含む粘着性ポリマーB、の2種類の粘着剤の混合物であることを特徴とするものである。
さらに、本発明の医療用テープ剤は粘着剤層に、ミリスチン酸イソプロピルなどの親油性の油状物を可塑剤として含有させることが好ましく、経皮吸収性薬物も粘着剤層中に配合することができる。
本発明の医療用テープ剤は、経皮吸収性薬物を配合した経皮吸収製剤としての用途のほかに、薬物を配合せずにテーピング材などの医療用材としての用途にも用いることが可能である。
本発明の粘着剤層に含まれる(A)成分の粘着性ポリマーAとして、メタクリル酸2−アセトアセトキシエチルエステルとその他のビニルモノマーを構成成分として含む共重合体を医療用テープ剤製造時の加熱乾燥時に揮散する溶媒中に溶解又は分散させたものを使うことが好ましい。
モノマーとしては、メタクリル酸2−アセトアセトキシエチルエステルと共重合するものであればよく、ビニルモノマーの中の一つは例えば(メタ)アクリル酸ブチル、(メタ)アクリル酸ヘキシル、(メタ)アクリル酸2−エチルヘキシルエステル、及び(メタ)アクリル酸オクチルエステルなどのようなアルキル基の炭素原子数が4個〜10個の(メタ)アクリル酸アルキルエステルが挙げられ、さらにその他のメタクリル酸メチル、ジアセトンアクリルアミド、テトラエチレングリコールジ(メタ)アクリレート、ヘキサエチレングリコールジ(メタ)アクリレート、(メタ)アクリル酸2−ヒドロキシエチルエステル等から選択される1種又は2種以上を用いることができる。
アルキル基の炭素原子数が4個〜10個の(メタ)アクリル酸アルキルエステルとして、(メタ)アクリル酸ブチル、(メタ)アクリル酸ヘキシル、(メタ)アクリル酸2−エチルヘキシルエステル、及び(メタ)アクリル酸オクチルなどを挙げることができ、この中から適宜1種又は2種以上を選択して用いることができる。アルキル基の炭素原子数が4個〜10個の(メタ)アクリル酸アルキルエステルの含量は共重合体総量の40質量%以上であることが好ましい。
本発明のテープ剤に用いる粘着性ポリマーAと粘着性ポリマーBの製造法を下記の粘着剤1〜5で示す。
[粘着剤1の製造]
ジムロート冷却器、温度計、窒素ガス吹き込み管及び攪拌翼を備えた2リットルの4つ口フラスコに、アクリル酸2−エチルヘキシルエステル157.5g、メタクリル酸2−
アセトアセトキシエチルエステル35g、ジアセトンアクリルアミド80.5g、メタクリル酸メチル76g、テトラエチレングリコールジメタクリレート1.3gを仕込み、溶剤として酢酸エチル525gを加えて均一になるまで溶解した。100ml/分の流量の窒素ガスを吹き込みながら75℃まで昇温した。75℃で30分間維持した後、重合開始剤として過酸化ベンゾイル0.21gを5gの酢酸エチルに溶解して添加し、外温を85℃に設定した。開始剤投入後、3、5、7時間後にトルエン300gを3回100gづつ分割して投入した。重合中は、100ml/分の流量の窒素ガスを吹き込み続けた。最後のトルエン投入から12時間後に、過酸化ベンゾイル0.35gを追加触媒として投入し、その後、外温設定を95℃にして12時間、熱処理した後に冷却して粘着剤1を得た。
粘着剤1の溶液の物性
溶液粘度(B型粘度計で測定):30,000mPa・s
固形分(150℃ 1時間処理):28.5%
粘着剤1の製造で示した方法と同様にして、粘着剤2及び粘着剤3を製造した。
[粘着剤4、5の製造]
メタクリル酸2−アセトアセトキシエチルエステルを含まない粘着性ポリマーBの粘着剤4及び粘着剤5は、粘着剤1の製造で示した方法と同様にして製造した。
[粘着剤6]
メタクリル酸2−アセトアセトキシエチルエステルを含まない粘着性ポリマーBの粘着剤6として、市販の溶剤型アクリル系粘着剤のニッセツPE300(商品名;日本カーバイド工業製)を用いた。
ねじ口ビンに、粘着剤1を40g量りとり、次いで粘着剤6を7.1g量りとり、ビン中で1時間以上攪拌した。乾燥した後の被膜重量が約140mg/10cm2となるように、コーティング試験機(LTE−S、Wener Mathis AG社製)を用いて、支持体(ポリエステルフィルム)上に本液を塗工・乾燥した後、剥離ライナー(シリコン処理が施されたポリエステルフィルム)のシリコン面が粘着剤と接するように被覆し、実施例1の医療用テープ剤を得た。
ねじ口ビンに、粘着剤1を40g、次いで粘着剤6を7.1g、最後にミリスチン酸イソプロピル3.6gをそれぞれ量りとり、ビン中で1時間以上攪拌した。乾燥した後の被膜重量が140mg/10cm2となるように、コーティング試験機(LTE−S、Wener Mathis AG社製)を用いて、支持体(ポリエステルフィルム)上に本液を塗工・乾燥した後、剥離ライナー(シリコン処理が施されたポリエステルフィルム)のシリコン面が粘着剤と接するように被覆し、実施例2の医療用テープ剤を得た。
実施例1又は2の製造法と同様な方法で、実施例3〜21及び比較例1〜5を製造した。
ねじ口ビンに、粘着剤1を40g、次いで粘着剤6を7.1g、最後にケトプロフェン1.6gをそれぞれ量りとり、ビン中で1時間以上攪拌した。乾燥した後の被膜重量が140mg/10cm2となるように、コーティング試験機(LTE−S、Wener Mathis AG社製)を用いて、ポリエステルフィルム製の支持体上に本液を塗工・乾燥した後、シリコン処理が施されたポリエステルフィルム製の剥離ライナーのシリコン面が粘着剤と接するように被覆し、実施例22の医療用テープ剤を得た。得られた製剤中のケトプロフェンは10質量%であった。
ねじ口ビンに、粘着剤1を38g、次いで粘着剤6を6.8g、さらに、ミリスチン酸イソプロピルを3.8g、最後にケトプロフェンを2gそれぞれ量りとり、ビン中で1時間以上攪拌した。乾燥した後の被膜重量が140mg/10cm2となるように、コーティング試験機(LTE−S、Wener Mathis AG社製)を用いて、ポリエステルフィルム製の支持体上に本液を塗工・乾燥した後、表面をシリコン処理が施されたポリエステルフィルム製の剥離ライナーのシリコン面が粘着剤と接するように被覆し、実施例23の医療用テープ剤を得た。得られた製剤中のケトプロフェンは10質量%であった。
比較例6では、ねじ口ビンに、粘着剤6、ケトプロフェンを量りとり、ビン中で1時間以上攪拌し、後は実施例22と同様に医療用テープ剤とした。比較例7では更にミリスチン酸イソプロピルを加えて医療用テープ剤とした。比較例8及び9では比較例6及び7の成分の外にイソシアネート架橋剤を加えて医療用テープ剤とした。
本発明の実施例1〜23と、比較例1〜9の医療用テープ剤を用いて粘着特性を、凝集力、貼着力、及び油状物質の相溶性の観点から下記の(官能)試験法で評価した。
1)凝集力試験
医療用テープ剤の剥離ライナーを剥がし、粘着剤面を指で触り、下記の評価基準に基づいて評価する。
評価基準
○:天然ゴムラテックスを使用した市販のインドメタシン含有テープ剤S、市販のフェルビナク含有テープ剤Fと同程度の凝集力
△:スチレン−イソプレン−スチレン共重合体を使用した市販のケトプロフェン含有テープ剤M、市販のフルルビプロフェン含有テープ剤Yと同程度の凝集力
×:市販品以下の凝集力
−:凝集力が著しく低いため(半固形の状態)評価不能
医療用テープ剤の剥離ライナーを剥がし、粘着剤面を指で触り、下記の評価基準に基づいて評価する。
評価基準
○:スチレン−イソプレン−スチレン共重合体を使用した市販のケトプロフェン含有テープ剤M、市販のフルルビプロフェン含有テープ剤Yと同程度の貼着力
△:天然ゴムラテックスを使用した市販のインドメタシン含有テープ剤S、市販のフェルビナク含有テープ剤Fと同程度の貼着力
×:市販品以下の貼着力
−:凝集力が著しく低いため(半固形の状態)評価不能
医療用テープ剤の剥離ライナーを剥がし、剥離ライナーの表面に液状の物質が付着していないことを光学顕微鏡で観察して、下記の評価基準に基づいて評価する。
評価基準
○:剥離ライナー表面上に液状物質なし
×:剥離ライナー表面上に液状物質あり
−:凝集力が著しく低いため(半固形の状態)評価不能
/:油状物質を配合していないため未評価
Claims (9)
- 支持体の片面に、粘着剤層と剥離ライナーが、順次積層された医療用テープ剤において、該粘着剤層の粘着剤が、
(A)メタクリル酸2−アセトアセトキシエチルエステル及びその他のビニルモノマーを構成成分として含有する共重合体である粘着性ポリマーA、
(B)アルキル基の炭素原子数が4個〜10個の(メタ)アクリル酸アルキルエステルとその他のビニルモノマーを構成成分として含有し、前記粘着性ポリマーAとは異なる共重合体である粘着性ポリマーB、
の2種類の粘着剤の混合物を含むことを特徴とする医療用テープ剤。 - 支持体の片面に、粘着剤層と剥離ライナーが、順次積層された医療用テープ剤において、該粘着剤層の粘着剤が、
(A)メタクリル酸2−アセトアセトキシエチルエステル、アルキル基の炭素原子数が4個〜10個の(メタ)アクリル酸アルキルエステル、及びその他のビニルモノマーを構成成分として含有する共重合体である粘着性ポリマーA、
(B)アルキル基の炭素原子数が4個〜10個の(メタ)アクリル酸アルキルエステルとその他のビニルモノマーを構成成分として含有し、前記粘着性ポリマーAとは異なる共重合体である粘着性ポリマーB、
の2種類の粘着剤の混合物を含むことを特徴とする請求項1に記載した医療用テープ剤。 - 粘着剤層が、さらに上記(A)成分の粘着性ポリマーAと(B)成分の粘着性ポリマーBを混合した粘着剤と相溶する油状成分からなる可塑剤を含むことを特徴とする請求項1又は2に記載した医療用テープ剤。
- 前記粘着性ポリマーAのメタクリル酸2−アセトアセトキシエチルエステルの含量が、前記粘着性ポリマーAに含まれる共重合体の総重量の1質量%〜50質量%であることを特徴とする請求項1〜3のいずれか1項に記載した医療用テープ剤。
- 前記粘着性ポリマーAのメタクリル酸2−アセトアセトキシエチルエステルの含量が、前記粘着性ポリマーAに含まれる共重合体の総重量の10質量%〜45質量%であること
を特徴とする請求項1〜4のいずれか1項に記載した医療用テープ剤。 - 前記(A)成分の粘着性ポリマーAと、前記(B)成分の粘着性ポリマーBの含有比率が、1:9〜4:1である請求項1〜5のいずれか1項に記載した医療用テープ剤。
- 前記可塑剤が、ミリスチン酸イソプロピル、セバシン酸ジエチル、アジピン酸ジイソプロピル、中鎖脂肪酸トリグリセリド、及びヒマシ油からなる群から選ばれる1種又は2種以上の油状物であることを特徴とする請求項3〜6のいずれか1項に記載した医療用テープ剤。
- 前記粘着剤層中に、経皮吸収性薬物を含有してなる請求項1〜7のいずれか1項に記載した医療用テープ剤。
- 前記(A)成分の粘着性ポリマーAが、メタクリル酸2−アセトアセトキシエチルエステル、アクリル酸2−エチルヘキシルエステル、メタクリル酸メチルエステル、ジアセトンアクリルアミド、及びテトラエチレングリコールジメタクリレートを構成成分として含有する共重合体であることを特徴とする請求項1〜8のいずれか1項に記載した医療用テープ剤。
