Description
天然に生じない融合タンパク質を、好ましくは、遺伝子工学的手法によって製造する。その手法では、融合パートナーをコードする1個の核酸配列、特にDNA配列及び、ハイドロフォビン部分をコードする1個の核酸配列、特にDNA配列を結合することで、結合した核酸配列の遺伝子発現により所望のタンパク質が宿主生物中に発生する。このような方法は、本出願人の先行出願であるDE102005007480.4に開示されている。
本発明により使用させるハイドロフォビンの作成及び試験
ポリメラーゼ連鎖反応をオリゴヌクレオチドHa1570およびHa1571(Ha1572/Ha1573)を用いて行った。鋳型DNAは、Bacillus subtilis細菌のゲノムのDNAを使用した。得られたPCR断片は、Bacillus subtilisのyaaD/yaaE遺伝子およびそれらの末端でいずれの場合も、それぞれNcolおよびBglll制限酵素の開裂部分のコード配列を含んでいた。PCR断片を精製し、制限エンドヌクレアーゼNcolおよびBglllで切断した。このDNA断片を挿入物として使用し、予め制限エンドヌクレアーゼNcolおよびBglllで直鎖化したベクターpQE60(Qiagen社)にクローニングした。そして、得られたベクターpQE60YAAD#2/pQE60YaaE#5はそれぞれ、YAAD::His6およびYAAE::HIS6から成るタンパク質の発現に使用される。
ポリメラーゼ連鎖反応をオリゴヌクレオチドKaM416及びKaM417を用いて行った。鋳型DNAは、Aspergillus nidulansカビのゲノムDNAを使用した。得られたPCR断片はハイドロフォビン遺伝子dewA及びN末端のXa因子プロテアナーゼ開裂部位のコード配列を含んでいた。PCR断片を精製し、制限エンドヌクレアーゼBamHIで切断した。このDNA断片を挿入物として使用し、予め制限エンドヌクレアーゼBglllで直鎖化したベクターpQE60YAAD#2にクローニングした。
プラスミド#513をプラスミド#508と同様に、オリゴヌクレオチドKaM434及びKaM435を使用してクローニングした。
プラスミド#507をプラスミド#508と同様に、オリゴヌクレオチドKaM417及びKaM418を使用してクローニングした。鋳型DNAには、人工的に合成したDNA配列であるハイドロフォビンBASF1(別表参照)
プラスミド#506をプラスミド#508と同様に、オリゴヌクレオチドKaM417及びKam418を使用してクローニングした。鋳型DNAは人工的に合成したDNA配列であるハイドロフォビンBASF2(別表参照)を用いた。
プラスミド#526をプラスミド#508と同様に、オリゴヌクレオチドKaM464およびKam465を使用してクローニングした。鋳型DNAは、スエヒロタケ(Schyzophyllum commune)の相補的DNA(別表参照)を用いた。
yaadハイドロフォビンDewA−His6を発現するE.coil株を15mlグライナー試験管中の3mlLB液体培地に接種した。37℃、200rpmで振とう培養した。それぞれの場合に2本のバッフル付き1Lエルレンマイヤーフラスコ中の250mlLB培地(+100μg/mlアンピシリン)に前培養の培養液1mlを接種し、37℃、180rpmで9時間振とう培養した。20Lファーメンター中の13.5LのLB培地(+100μg/mlアンピシリン)に前培養(水に対して測定したOD600nm1:10)の0.5Lを接種した。140mlの100mMIPTGをOD600nmが〜3.5の時点で添加した。3時間後、ファンメンターを10℃に冷却し、培養地を遠心分離により除去した。細胞沈殿物を次の精製工程で使用する。
(C−末端His6タグをプロセッシングするハイドロフォビン融合タンパク質の精製)
100gの細胞沈殿物(100〜500mgのハイドロフォビン含有)を50mMリン酸ナトリウム緩衝液(pH7.5)にて総容量200mlになるようにして、再懸濁した。懸濁液を、Ultraturrax typeT25(Janke and Kunkel;IKA−Labortechnik社製)で10分間処理し、次いで核酸を分解する目的で、50単位のベンゾナーゼ(Merck,Darmstadt社;オーダーNo.1.01697.0001)で室温にて1時間反応させた。細胞が破壊される前に、ガラスカートリッジ(P1)を使用してろ過を行った。細胞の破壊及び残存したゲノムのDNAをせん断する目的で、1500barで高圧ホモジナイザー(Microfluidizer M−110EH;Microfluidizer社)で2回処理した。ホモジネートを遠心分離(Sovall RC−5B, GSAローター,250ml遠心ビーカー、4℃、60分、12000rpm、23000g)し、上清を氷冷し、沈殿物を100mlリン酸ナトリウム緩衝液(pH7.5)で再懸濁した。遠心分離及び再懸濁を3回繰り返して、3回目に1%SDSを含むリン酸緩衝液を用いた。再懸濁後、溶液を1時間攪拌し、続いて最後の遠心分離(Sovall RC−5B, GSAローター,250ml遠心ビーカー、4℃、60分、12000rpm、23000g)を行った。SDS―PAGE分析によると、ハイドロフォビンは最後の遠心分離後の上清に存在している(図1)。本実験は、ハイドロフォビンがE.coilの細胞内におそらく封入体(including bodies)の形態で存在していることを示す。50mlのハイドロフォビンを含有する上清を50mMTris−Cl緩衝液(pH8.0)で平衡化した50mlのnickel−Sepharose High Performance17−5268−02カラム(Amersham社)にかけた。カラムを50mMTris−Cl緩衝液(pH8.0)で洗浄し、続いて200mMイミダゾールを含む50mMTris−Cl緩衝液(pH8.0)で溶出した。イミダゾールを除く目的で、溶液を50mMTris−Cl緩衝液(pH8.0)で透析した。

図1のハイドロフォビンは分子量約53kDを有する。何本かのより小さい分子量のバンドはハイドロフォビンの分解産物を示す。
基質:
ガラス(窓ガラス、Sueddeutshe Glass、Mannheim、Germany):
ハイドロフォビン濃縮物:100μg/ml、
50mM酢酸ナトリウム(pH4.0)+0.1質量%Tween20中でガラススライドを1晩(温度80℃)培養し、
続いて、ハイドロフォビン被覆されたガラススライドを蒸留水中で洗浄し、
続いて、1質量%n−ドデシル硫酸ナトリウム(SDS)水溶液において80℃で10分間培養し、
蒸留水で蒸留したもの。
セラミック表面の汚れ除去被覆用のハイドロフォビンの使用
使用される溶液
性能試験は、実施例8にしたがって製造された融合タンパク質yaad−Xa−dewA−his(SEQ ID NO:19)の水溶液を用いて行った。溶液中のハイドロフォビンの濃度:100μg/ml(0.01質量%)
使用されるセラミック表面
光沢があり白色で10cm×15cmのセラミックタイル(Novocker社製)をエタノールと水で拭いたもの。
上記試験を、IKW ballast soil(Seifen、Fette、Oele、Wachse(SOEFW)−Journal、Volume 124、14/98、page 1029)を使用して行った。
2gの上記100μg/mlの濃度のハイドロフォビン水溶液を1個のタイルへ滴下し(1.3μmハイドロフォビン/cm2)、表面を完全にカバーするように布で丁寧に広げた。その後、そのタイルを24時間空気乾燥した。
処理されたタイルについて水滴に対する接触角を測定したところ56°(10回の測定値の平均)であった。対して、未処理のタイルの接触角は、20°であった。従って、そのタイルは、明らかに疎水化されていた。
Claims (9)
- 硬質無機建材、自然石、人造石又はセラミックから成る群から選択される材料の表面を処理するためのハイドロフォビンの使用方法
- 前記処理を、少なくとも1種のハイドロフォビンとともに溶媒を含む配合物を使用して行うことを特徴とする請求項1に記載の使用方法。
- 表面を処理する方法であって、
前記表面を少なくとも1種のハイドロフォビンと接触させる工程を含み、
前記表面が硬質無機建材、自然石、人造石又は、セラミックスから成る群から選択される材料の表面を含むことを特徴とする方法。 - 前記処理を、少なくとも1種のハイドロフォビンとともに溶媒を含む配合物を使用して行うことを特徴とする請求項3に記載の使用方法。
- 前記溶媒は、水を含むことを特徴とする請求項4に記載の方法。
- 前記配合物中のハイドロフォビンの量が、前記配合物の全成分の合計を基準として0.0001質量%から1質量%であることを特徴とする請求項4又は5に記載の方法。
- 少なくとも1種のハイドロフォビンで被覆された表面であって、
硬質無機建材、自然石、人造石又は、セラミックから成る群から選択される材料の表面を含むことを特徴とする表面。 - 汚れ忌避性を有することを特徴とする請求項7に記載の表面。
- 疎水性を有することを特徴とする請求項7又は8に記載の表面。
