Abstract
【解決手段】6−ジアゾ−5−オキソ−L−ノルロイシン等のグルタミナーゼ阻害剤を有効成分として含有する抗炎症剤。

Description
1.グルタミナーゼ阻害剤を有効成分として含有する抗炎症剤。
2.グルタミナーゼ阻害剤がDON又はCB−839である前記1に記載の抗炎症剤。
3.炎症に由来する疾患の予防、改善又は治療に用いられる前記1又は2に記載の抗炎症剤。
4.前記疾患が、2型糖尿病、高血糖、インスリン抵抗性、高血圧、アテローム性動脈硬化、NAFLD、脂肪肝、脂質異常症、関節炎、アルツハイマー、炎症性大腸炎、喘息及び心疾患から選ばれる少なくとも1である、前記1〜3のいずれか1に記載の抗炎症剤。
5.血糖値上昇、血圧上昇、血中中性脂肪上昇、動脈硬化、関節炎、慢性膝関節痛若しくは認知症の抑制、予防又は改善作用、脂質代謝の改善作用、並びに記憶学習機能及び認知機能の少なくとも一方の増強又は改善作用から選ばれる少なくとも1の作用を有する前記1又は2記載の抗炎症剤。
6.前記1〜5のいずれか1に記載の抗炎症剤を含有する飲食品。
7.前記1〜5のいずれか1に記載の抗炎症剤を含有する医薬。
野生型C57BL/6雄性マウス(三協ラボサービス社製)について、通常食(以下、NDとも略す。)を摂取させた普通餌群(20匹)及び脂肪含有量が60kcal%である高脂肪食(以下、HFDとも略す。)(Research Diets社製、D12492)を摂取させた高脂肪食群(20匹)を調製した。0、3、6及び10日目にマウス(肥満や耐糖能異常は未発症)各5匹の脂肪組織に関して、網羅的な発現変動遺伝子を抽出し、マイクロアレイを用いて階層クラスタリング解析を行った。その結果、初期段階(3日目)で普通餌群と高脂肪食群を振り分け可能なGroup12を検出した。
動物実験により、CB−839の投与による効果を解析した。5週齢雄性野生型C57BL/6マウス(三協ラボサービス社製)を12時間の明暗サイクルを一定にして制御された温度で維持した。マウスには食物と水を自由に摂取させた。
動物実験により6−ジアゾ−5−オキソ−L−ノルロイシン(DON)の投与による効果を解析した。4週齢の野生型C57BL/6雄性マウス(三協ラボサービス社製)を12時間の明暗サイクルを一定にして制御された温度で維持した。マウスには食物と水を自由に摂取させた。
RT−PCRにより、CB−839の免疫学的効果を調べた。TRISure(日本ジェネティクス社製)を用いて、凍結eWATからtotal RNAを単離した。組織(100mg)を300μLのTRISureでホモジナイズした。TRISure(600μL)とクロロホルム(200μL)を加え、十分に混合した。

- グルタミナーゼ阻害剤を有効成分として含有する抗炎症剤。
- グルタミナーゼ阻害剤がDON又はCB−839である請求項1に記載の抗炎症剤。
- 炎症に由来する疾患の予防、改善又は治療に用いられる請求項1又は2に記載の抗炎症剤。
- 前記疾患が、2型糖尿病、高血糖、インスリン抵抗性、高血圧、アテローム性動脈硬化、NAFLD、脂肪肝、脂質異常症、関節炎、アルツハイマー、炎症性大腸炎、喘息及び心疾患から選ばれる少なくとも1である、請求項1〜3のいずれか1項に記載の抗炎症剤。
