Claims (10)

1. Metoda za kvantifikaciju kimerne cfDNK u uzorku pacijenta, pri čemu se metoda sastoji od: (a) dobivanja uzorka cirkulirajuće slobodne DNK (cfDNK) iz uzorka krvi, seruma ili plazme dobivenog od pacijenta; (b) provođenja multipleks digitalnog PCR-a koji se sastoji najmanje od prve amplifikacije i druge amplifikacije, naznačeno time, što prva amplifikacija cilja prvi genomski lokus jedinstvene kopije i dovodi do proizvodnje prvog amplikona, a druga amplifikacija cilja drugi genomski lokus jedinstvene kopije i dovodi do proizvodnje drugog amplikona, naznačeno time, što se prvi i drugi amplikon razlikuju po dužini za najmanje 50 baznih parova; (c) utvrđivanja omjera prvog ili drugog amplikona u ukupnom amplificiranom proizvodu dobivenom pomoću digitalnog PCR-a iz koraka (b) radi pružanja faktora korekcije za dio uzorka cfDNK pogodan za amplifikaciju, i (d) korekcije dijagnostičkog digitalnog PCR-a provedenog na uzorku cfDNK radi procjene razine kimerne DNK prisutne u uzorku primjenom faktora korekcije utvrđenog u koraku (c).
2. Metoda prema patentnom zahtjevu 1, naznačena time, što se korak (b) dodatno sastoji od amplifikacije reakcije koja cilja DNK koja je dodana uzorku krvi, seruma ili plazme prije ekstrakcije.
3. Metoda prema patentnom zahtjevu 1 ili 2, naznačena time, što se prvi i drugi amplikon razlikuju po dužini za 100 ili više nukleotida.
4. Metoda prema bilo kojem patentnom zahtjevu od 1 do 3, naznačena time, što duži od prvog i drugog amplikona ima dužinu od 150 do 400 baznih parova, a kraći od prvog i drugog amplikona ima dužinu od 50 do 150 baznih parova.
5. Metoda prema bilo kojem patentnom zahtjevu od 1 do 4, naznačena time, što je uzorak cfDNK dobiven od pacijenta koji je imao transplantaciju.
6. Metoda prema bilo kojem patentnom zahtjevu od 1 do 4, naznačena time, što je uzorak cfDNK dobiven od pacijenta koji ima rak.
7. Metoda prema bilo kojem patentnom zahtjevu od 1 do 4, naznačena time, što je uzorak cfDNK dobiven od trudne pacijentice.
8. Metoda prema bilo kojem patentnom zahtjevu od 1 do 7, naznačena time, što je pacijent čovjek.
9. Metoda prema bilo kojem patentnom zahtjevu od 1 do 8, naznačena time, što je kimerna DNK dobivena od darivatelja za transplantaciju ili fetusa.
10. Metoda prema bilo kojem patentnom zahtjevu od 1 do 8, naznačena time, što kimerna DNK potječe iz stanica raka.
