The astaxanthin extract is extracted from haematococcus pluvialis, haematococcus pluvialis powder is weighed and placed in an extraction kettle, ethanol is added according to the feed-liquid ratio of 1:1g/ml, the temperature of the extraction kettle is set to be 40-42 ℃, the pressure is set to be 30Mpa, the flow rate of CO2 is set to be 20L/h, the temperature of a separation kettle is set to be 30 ℃, the pressure is set to be 8-9MPa, the temperature of a separation kettle is set to be 35 ℃, continuous dynamic extraction is carried out for 2h, dark red liquid is collected, and then the ethanol is removed through rotary evaporation, so that the astaxanthin extract is obtained.
The following examples employ a method of preparation of vaccinium myrtillus extract:
mixing the washed and crushed Vaccinium myrtillus with water, adding mixed enzyme into the crushed mixture of Vaccinium myrtillus to adjust pH value of the mixture to 4.5, placing in water bath at 55 deg.C for enzymolysis for 3h, adding sugar 16% of the mixture, inoculating mixed bacteria to the adjusted enzymolysis product, sealing, fermenting, collecting supernatant, and performing reduced pressure ultrafiltration to obtain Vaccinium myrtillus extract.
The seaweed extracts used in the following examples were prepared:
cleaning and crushing seaweed, mixing the seaweed with water, adding a mixed enzyme to adjust the pH value of a system to 4.5, carrying out enzymolysis for 4 hours in water bath at 55 ℃, adding sugar accounting for 12% of the mass of the crushed mixed solution after enzymolysis, inoculating the adjusted enzymolysis product into mixed bacterial suspension, carrying out sealed fermentation, carrying out ultrafiltration sterilization to obtain a primary fermentation liquid, taking supernatant, and carrying out reduced pressure ultrafiltration to obtain a seaweed extract. The seaweed material is kelp.
The mixed fungal enzymes in the preparation method of the vaccinium myrtillus extract and the seaweed extract are cellulase, hemicellulase, pectinase and acid protease, the addition amounts of the cellulase, the hemicellulase, the pectinase and the acid protease are respectively based on the mass ratio of the mixed solution, the addition amount of the cellulase is 0.4%, the addition amount of the hemicellulase is 0.4%, the addition amount of the pectinase is 0.2% and the addition amount of the acid protease is 0.4%.
The mixed bacterial suspension in the preparation method of the vaccinium myrtillus extract and the seaweed extract is yeast suspension, lactobacillus plantarum suspension and bifidobacterium suspension, and the enzymolysis product is uniformly stirred with the prepared yeast suspension, lactobacillus plantarum suspension and bifidobacterium suspension according to the mass ratio of 60:1.5:1.5: 1.5; the yeast suspension is 20% yeast suspension prepared from yeast dry powder and sterile normal saline; the lactobacillus plantarum bacterial suspension is a bacterial suspension with the concentration of 20 percent prepared by sterile normal saline for lactobacillus plantarum dry powder; the bacterial suspension of the bifidobacterium is 20 percent of bacterial suspension prepared by bifidobacterium dry powder and sterile normal saline.
The preparation of centella asiatica extract used in the following examples:
cleaning fresh or dried herba Centellae, pulverizing, adding purified water and pectase (addition amount: 4mg/g) at a ratio of 1:50g/ml, performing enzymolysis at 45 deg.C for 2 hr, performing heat reflux extraction for 2 hr, repeatedly extracting for 1 time, mixing filtrates, cooling the filtrate to room temperature, centrifuging at 4 deg.C and 10000r/min for 15min to obtain supernatant, and performing reduced pressure ultrafiltration to obtain herba Centellae extract.
The rosemary extracts used in the following examples were prepared:
drying fresh rosemary at 40 ℃ for 10h, then crushing, adding purified water and pectinase (the addition amount is 5mg/g) according to the material-liquid ratio of 1:40-60g/ml, carrying out enzymolysis at 45 ℃ for 2 hours, carrying out hot reflux extraction for 2 hours, repeatedly extracting for 1 time, combining 2 times of filtrates, cooling the filtrate to room temperature, centrifuging at 4 ℃ at 10000r/min for 15min, obtaining supernatant, and carrying out reduced pressure ultrafiltration to obtain the rosemary extract.
Other raw materials and equipment which are not specially described are all commercial products.
Example 1
The anti-aging composition comprises the following raw materials in parts by weight: 7 parts of astaxanthin microcapsules, 6 parts of vaccinium myrtillus extracts, 6 parts of seaweed extracts, 3.5 parts of centella asiatica extracts and 3.5 parts of rosemary extracts.
The anti-aging composition of this example was tested to have a DPPH radical scavenging capacity of 88.2%, superoxide anion radical scavenging capacity of 52%, and hydroxyl radical scavenging capacity of 23.9%; after the anti-aging composition is placed for half an hour, the moisture retention rate is 84.7 percent; has a certain inhibition rate on tyrosinase, and the inhibition effect is as follows: 38.5 percent.
The method for detecting the DPPH free radical scavenging capacity comprises the following steps: 1mL of the anti-aging composition mixture and 2mL of an 80% aqueous solution of DPPH in ethanol (2X 10) were sequentially added to a colorimetric tube4mol/L), fully oscillating and mixing uniformly, measuring the light absorption value at the wavelength of 517nm by using a 1cm cuvette, placing the cuvette in the dark for reacting for half an hour, immediately measuring the OD value at the wavelength after the reaction is finished, taking the OD value as a test group, and recording A1(ii) a Mixing 2mL of the PPH solution and 1mL of 80% ethanol solution, measuring the absorbance under the same condition to obtain a blank group A0(ii) a Mixing 1mL of the antiaging composition mixture with 2mL of 80% ethanol solution, measuring absorbance under the same condition, and recording as control group A2. Each set of experiments was averaged in triplicate. DPPH radical clearance was calculated using the following formula: DPPH.radical scavenging ratio (%) [1- (A)1-A2)/A0]×100。
The method for detecting the scavenging capacity of the superoxide anion free radicals comprises the following steps: 1mL of the anti-aging composition mixture and the control solution were weighed into a graduated tube, followed by addition of 0.5mL of phosphate buffer (0.5mol/L) at pH 7.8, addition of 1.5mL of methionine reagent (0.13mol/L), addition of 0.3mL of NBT solution (0.75mmol/L), and addition of 0.3mL of riboflavin (0.02mmol/L) to mix well, followed by reaction at 35 ℃ under 4000lx for 25 min. After the reaction, the reaction was terminated in the dark, and after the reaction was completed, the OD value was measured immediately at 560nm, and each test sample was recorded using distilled water as a blankOD value of (1). Each set of experiments was averaged in triplicate and recorded in detail. Superoxide anion clearance was calculated using the following equation: o is2-Clearance (%) ═ a1-A2)/A1X 100, wherein: a1 is the absorbance of the blank control, A2 is the absorbance of the mixture of the experimental composition.
The method for detecting the scavenging capacity of the hydroxyl free radical comprises the following steps: adding 1mL of the anti-aging composition mixed solution into a 10mL graduated test tube, taking 1mL of distilled water as a blank control, adding 1.0mL of hydrogen peroxide with the concentration of 6mmol/L, adding 1.0mL of salicylic acid with the concentration of 20mmol/L, and adding 0.5mL of ferrous sulfate with the concentration of 1.5 mmol/L. Heating in a thermostatic waterbath at 37 deg.C for 1h, adjusting to zero with distilled water, measuring absorbance A with spectrophotometer at wavelength of 562nm, and repeating for 3 times. The hydroxyl radical clearance is calculated using the following formula: hydroxyl radical scavenging rate (%) - (A)1-A2)/A1]X 100, wherein: a. the1Average absorbance for blank; a. the2Is the average absorbance of the anti-aging composition mixture.
The moisture retention rate detection method comprises the following steps: placing the humectant in a dryer, and placing the allochroic silica gel at the bottom of the dryer. Soaking facial mask paper with a certain specification in the anti-aging composition mixed solution to uniformly soak the anti-aging composition mixed solution, taking out after 1h, weighing and counting, then placing the facial mask paper in a weighed beaker, uniformly placing the beaker close to the wall, then placing the beaker with the soaked facial mask paper in a dryer, placing the beaker in a safe and clean environment, and weighing after a certain period of time. In the test, the determination time interval is set as 0.5h, and then the moisture retention rate of the anti-aging composition mixed liquor is calculated. The calculation formula is as follows: moisture retention rate is M2/M1X 100%, wherein: m1For the weight of water before standing, M2Is the weight of water after standing.
Example 2
The anti-aging composition comprises the following raw materials in parts by weight: 8 parts of astaxanthin microcapsules, 7 parts of vaccinium myrtillus extracts, 7 parts of seaweed extracts, 4 parts of centella asiatica extracts and 4 parts of rosemary extracts.
The anti-aging composition of this example was tested to have a DPPH radical scavenging capacity of 91.3%, superoxide anion radical scavenging capacity of 55.2%, and hydroxyl radical scavenging capacity of 25.4%; after the anti-aging composition is placed for half an hour, the moisture retention rate is 86.2 percent; has a certain inhibition rate on tyrosinase, and the inhibition effect is as follows: 42.1 percent.
Example 3
The anti-aging composition comprises the following raw materials in parts by weight: 9 parts of astaxanthin microcapsules, 8 parts of vaccinium myrtillus extracts, 8 parts of seaweed extracts, 4.5 parts of centella asiatica extracts and 4.5 parts of rosemary extracts.
The anti-aging composition of this example was found to have a DPPH radical scavenging ability of 92.8%, superoxide anion radical scavenging ability of 58.6%, and hydroxyl radical scavenging ability of 27.2%; after the anti-aging composition is placed for half an hour, the moisture retention rate is 87.5 percent; has a certain inhibition rate on tyrosinase, and the inhibition effect is as follows: 43.5 percent.
Comparative example 1
The anti-aging composition comprises the following raw materials in parts by weight: 5 parts of bilberry extract, 5 parts of seaweed extract, 5 parts of asiatic centella extract and 5 parts of rosemary extract.
The anti-aging composition of this comparative example was tested to have a DPPH radical scavenging capacity of 65.2%, superoxide anion radical scavenging capacity of 39.5%, and hydroxyl radical scavenging capacity of 12.5%; after the anti-aging composition is placed for half an hour, the moisture retention rate is 78.6 percent; has a certain inhibition rate on tyrosinase, and the inhibition effect is as follows: 35.6 percent.
Comparative example 2
The anti-aging composition comprises the following raw materials in parts by weight: 5 parts of astaxanthin microcapsules, 5 parts of seaweed extract and 4 parts of centella asiatica extract.
The anti-aging composition of this comparative example was tested to have a DPPH radical scavenging capacity of 62.4%, superoxide anion radical scavenging capacity of 34.5%, and hydroxyl radical scavenging capacity of 10.6%; after the anti-aging composition is placed for half an hour, the moisture retention rate is 80.5 percent; has a certain inhibition rate on tyrosinase, and the inhibition effect is as follows: 22.6 percent.
Comparative example 3
The anti-aging composition comprises the following raw materials in parts by weight: 5 parts of astaxanthin extract, 4 parts of bilberry extract, 4 parts of seaweed extract, 2 parts of centella asiatica extract and 2 parts of rosemary extract.
The anti-aging composition of this comparative example was tested to have a DPPH radical scavenging capacity of 60.4%, superoxide anion radical scavenging capacity of 35.1%, and hydroxyl radical scavenging capacity of 9.6%; after the anti-aging composition is placed for half an hour, the moisture retention rate is 77.8 percent; has a certain inhibition rate on tyrosinase, and the inhibition effect is as follows: 20.5 percent.
As can be seen from the analysis of the above examples and comparative examples, the anti-aging composition obtained by the examples of the present invention has a good effect. The anti-aging composition of the embodiment has good anti-oxidation activity effect and can play a role in repairing cells and delaying aging in view of the DPPH free radical scavenging capacity, superoxide anion free radical scavenging capacity and hydroxyl free radical scavenging capacity. The whitening effect of the anti-aging composition is proved by detecting the tyrosinase activity inhibition rate, the obtained embodiment has high tyrosinase inhibition rate which can reach 43.5%, and the anti-aging composition is proved to have the whitening effect at the same time.
The anti-aging composition ingredients of comparative examples 1 and 2 lack 1-2 ingredients, and the DPPH radical scavenging ability, superoxide anion radical scavenging ability, hydroxyl radical scavenging ability, and tyrosinase inhibition rate were significantly lower than those of the examples. This is because different antioxidants have different mechanisms and functional sites during their use, and they have synergistic effects and cannot be substituted for each other, and the antioxidant properties are limited when the antioxidant structure is relatively single. The invention has synergistic effect among different types of antioxidants, the characteristics of all components are complementary, the coordination and the synergy are realized, the oxidation resistance is obviously improved, and the invention has stronger anti-aging performance.
The anti-aging composition of comparative example 3, in which non-microencapsulated astaxanthin was used, was remarkably reduced in oxidation resistance because astaxanthin, although having strong oxidation resistance, is very unstable and easily oxidatively denatured upon contact with air, while microencapsulation improved stability and water solubility of astaxanthin.
The composition obtained in the embodiment can be prepared into one of facial masks, essence, lotion, skin-care gel and cream according to the existing preparation method of cosmetics and the selection of corresponding auxiliary agents.
Application example 1
The mask containing the anti-aging composition of example 1 further comprises an auxiliary agent, wherein the auxiliary agent comprises 5 parts of butanediol, 4 parts of glycerol, 3 parts of propylene glycol, 1.5 parts of betaine, 0.8 part of bioglycan, 0.8 part of 1, 2-pentanediol, 1.5 parts of sodium hyaluronate, 0.2 part of allantoin, 1 part of hydrolyzed collagen, 0.03 part of disodium EDTA, and the balance of deionized water to 100 parts.
The preparation method of the mask containing the anti-aging composition comprises the following steps:
(1) adding water, butanediol, glycerol, propylene glycol, betaine, bioglycan gum, 1, 2-pentanediol, sodium hyaluronate, allantoin, hydrolyzed collagen and EDTA disodium into a reactor in sequence according to the weight parts, heating to 80-85 ℃, stirring until the materials are completely dissolved, and preserving heat for 20min to obtain a mixed solution.
(2) And (2) stirring the mixed solution prepared in the step (1), cooling to 30-35 ℃, adding the anti-aging composition according to the weight parts, uniformly stirring, and standing for 10 hours to obtain the facial mask nutrient solution.
(3) Filling the facial mask nutrient solution into a packaging bag containing facial mask paper after irradiation disinfection to obtain the facial mask containing the anti-aging composition.
Application example 2
The mask containing the anti-aging composition of example 2 further comprises an auxiliary agent, wherein the auxiliary agent comprises 6.5 parts of butanediol, 6 parts of glycerol, 5 parts of propylene glycol, 2 parts of betaine, 1.1 parts of bioglycan, 1.1 parts of 1, 2-pentanediol, 2.5 parts of sodium hyaluronate, 0.3 part of allantoin, 2 parts of hydrolyzed collagen, 0.04 part of disodium EDTA, and the balance of deionized water to 100 parts.
The preparation method of the facial mask containing the anti-aging composition is the same as the application example 1.
Application example 3
The mask containing the anti-aging composition of example 3 further comprises an auxiliary agent, wherein the auxiliary agent comprises 6 parts of butanediol, 5 parts of glycerol, 4 parts of propylene glycol, 2 parts of betaine, 1.1 parts of bioglycan, 1.1 parts of 1, 2-pentanediol, 2.5 parts of sodium hyaluronate, 0.3 part of allantoin, 2 parts of hydrolyzed collagen, 0.04 part of disodium EDTA, and the balance of deionized water to 100 parts.
The preparation method of the facial mask containing the anti-aging composition is the same as the application example 1.
Application example 4
The essence containing the anti-aging composition of example 3 further includes an auxiliary agent, which is 7 parts of sodium hyaluronate, 2 parts of glycerin, 1.5 parts of bioglycan, 3 parts of polyethylene glycol, 0.3 part of allantoin, 2 parts of hydrolyzed collagen, 0.4 part of xanthan gum, 0.04 part of disodium EDTA, and the balance up to 100 parts of deionized water.
The preparation method of the essence containing the anti-aging composition comprises the following steps:
(1) adding water, glycerol, bioglycan, polyethylene glycol, allantoin, hydrolyzed collagen, xanthan gum and EDTA disodium into a reactor in sequence according to the weight parts, heating to 80-85 ℃, stirring until the materials are completely dissolved, and preserving heat for 20min to obtain a mixed solution.
(2) And (2) stirring the mixed solution prepared in the step (1), cooling to 30-35 ℃, adding sodium hyaluronate according to the weight parts, uniformly stirring the anti-aging composition, and standing for 10 hours to obtain the anti-aging essence.
Application example 5
The skin care gel containing the anti-aging composition of example 3 further comprises an auxiliary agent, wherein the auxiliary agent is 7 parts of sodium hyaluronate, 1 part of glycerol, 3 parts of polyethylene glycol, 0.3 part of allantoin, 0.02 part of sodium alginate, 0.01 part of carbomer, 0.04 part of disodium EDTA, and the balance of deionized water to 100 parts.
The preparation method of the skin care gel containing the anti-aging composition comprises the following steps:
(1) adding sodium hyaluronate, glycerol, polyethylene glycol, an anti-aging composition and a proper amount of deionized water according to the weight parts, and uniformly stirring to obtain a solution 1.
(2) And weighing the sodium alginate in parts by weight, and dispersing the sodium alginate in a proper amount of deionized water to form gel liquid 1.
(3) And weighing the carbomer in parts by weight, and dispersing the carbomer into a proper amount of deionized water to form gel liquid 2.
(4) And respectively adding the solution 1, the gel solution 2 and the EDTA disodium into the balance of deionized water, and fully stirring and uniformly mixing to obtain the anti-aging skin care gel.
Sensory evaluation of mask
20 volunteers were selected by a double-blind random method, and after skin was cleaned, 1 piece of the facial mask of application example 1 was spread and attached to the face, and after 15 minutes, was peeled off and washed clean with clear water. Sensory evaluation volunteers claim: the human body is healthy after being aged 25-55 years, volunteers can accurately express the use feelings after being trained, and the evaluation standard is shown in Table 1.

The facial mask liquid disclosed by the invention is good in color, light purple, light in delicate fragrance, free of peculiar smell, fine and smooth, free of tight, mild and irritation to the face after being used, comfortable and cool in feeling, free of greasy adhesion, free of any adverse reaction and good in safety.
The astaxanthin microencapsulation can shield peculiar smell and improve stability and water solubility. Besides the functions of antioxidation, whitening and anti-aging, the natural plant extract has the effects of bacteriostasis and corrosion prevention, avoids the addition of chemical preservatives and further ensures the pure naturalness of the mask liquid.
Sensory evaluation of mask for anti-aging
Compared with other skin care products, the facial mask has the obvious characteristics of rapidly supplementing moisture and nutrient substances to the skin and having good instant effect. Therefore, the test adopts a double-blind random test method to evaluate the human body efficacy, a multifunctional skin tester is used for measuring the skin state parameters (including the skin water content and the skin elasticity) of the volunteer before use (0 h) and after use (4 h and 8h), the skin water content and the elasticity value change value are obtained by subtracting the value before use from the detection value after use, and the effect is better when the value is larger.
The using method comprises the following steps: after skin cleaning, 1 piece of the facial mask of application example 1 is spread and pasted on a face, and is taken off after 15 minutes, and is washed clean by clear water.
The volunteer requires: 30 people, age 25-55 years old, healthy, volunteers trained, can express their own use experience accurately.

As can be seen from the above table, after the facial mask is used for 4 hours and 8 hours, the moisture content and elasticity value of the skin of the experimental group and the control group are greatly improved after the product of the invention is used, and the moisture content and elasticity value of the skin fall back to 8 hours, which indicates that the prepared facial mask has good effect of improving the moisture content and elasticity of the skin and the duration time is longer than 8 hours. Meanwhile, the embodiment is more improved compared with the comparative example, and the falling speed is slow, so that the added astaxanthin and various natural plant extracts have the obvious effects of improving the moisture content and the elasticity value of the skin. Experiments show that the facial mask prepared by adding the astaxanthin and other plant extracts has the obvious effects of improving the water content and the elasticity of the skin, and no one of all volunteers has the symptoms of skin allergy and the like.
The anti-aging composition disclosed by the invention has the advantages that the anti-oxidation activity is obviously enhanced after the combination of various natural substances, and the anti-aging composition prepared by the invention achieves a remarkable anti-oxidation effect by forming a synergistic anti-oxidation effect in various modes such as repairing, regenerating, oxygen absorbing, metal ion complexing and the like just because various substances such as astaxanthin, blueberry extract, seaweed extract and the like have different anti-oxidation action mechanisms, and has the effects of obviously removing fine wrinkles and improving the skin elasticity. Therefore, the anti-aging composition disclosed by the invention can generate a good effect of immediately improving skin, can promote cell metabolism, is natural and non-irritant in the added raw materials, and can be widely applied to anti-aging skin care products.

Claims (7)

1. An anti-aging composition characterized by: the raw materials comprise the following components in parts by weight: 5-15 parts of astaxanthin microcapsules, 5-10 parts of vaccinium myrtillus extracts, 5-10 parts of seaweed extracts, 2-5 parts of centella asiatica extracts and 2-5 parts of rosemary extracts;
the vaccinium myrtillus extract is prepared by mixing washed and crushed vaccinium myrtillus with water, adding mixed enzyme into the vaccinium myrtillus crushed mixed solution to adjust the pH value of the mixed solution to 4.0-5.0, placing the mixed solution in a water bath at 50-60 ℃ for enzymolysis for 2-4h, then adding sugar which accounts for 14% -20% of the mass of the mixed solution, inoculating mixed bacteria suspension into the adjusted enzymolysis product, taking supernate after sealed fermentation, and carrying out reduced pressure ultrafiltration to obtain the vaccinium myrtillus extract;
the seaweed extract is prepared by cleaning and crushing seaweed, mixing the crushed seaweed with water, adding a mixed enzyme to adjust the pH value of a system to 4.5, carrying out enzymolysis for 2-4h in a water bath at 50-60 ℃, adding sugar accounting for 8-14% of the mass of the crushed mixed solution after enzymolysis, inoculating the adjusted enzymolysis product to a mixed bacteria suspension, carrying out sealed fermentation, taking supernatant, and carrying out reduced pressure ultrafiltration to obtain the seaweed extract;
the mixed enzyme is cellulase, hemicellulase, pectinase and acid protease, the addition amount of the cellulase is 0.2-0.6% of the mass of the mixed solution, the addition amount of the hemicellulase is 0.2-0.6% of the mass of the mixed solution, the addition amount of the pectinase is 0.16-0.22% of the mass of the mixed solution, and the addition amount of the acid protease is 0.2-0.6% of the mass of the mixed solution;
the mixed bacteria suspension is yeast bacteria suspension, lactobacillus plantarum suspension and bifidobacterium suspension, and the mass ratio of the enzymolysis product to the yeast bacteria suspension, the lactobacillus plantarum suspension and the bifidobacterium suspension is 50-100:1-2:1-2: 1-2.

The anti-aging composition according to claim 1, wherein: the raw materials are as follows by weight: 7-12 parts of astaxanthin microcapsules, 6-9 parts of vaccinium myrtillus extracts, 6-9 parts of seaweed extracts, 3.5-4.5 parts of centella asiatica extracts and 3.5-4.5 parts of rosemary extracts.
3. The anti-aging composition according to claim 2, wherein: the raw materials are as follows by weight: 8-10 parts of astaxanthin microcapsules, 7-8 parts of vaccinium myrtillus extracts, 7-8 parts of seaweed extracts, 4-4.5 parts of centella asiatica extracts and 4-4.5 parts of rosemary extracts.

The anti-aging composition according to claim 3, wherein: the astaxanthin microcapsule is prepared by mixing an astaxanthin extract with a wall material, then carrying out ultrasonic emulsification, adjusting the pH value of a mixed solution to 4.0, cooling and solidifying the microcapsule, and carrying out centrifugal separation;
