技术领域
本发明涉及在其出口处具有漩涡破坏器的一次性容器及使用所述容器的系统。
背景技术
在本发明之前,在使用不锈钢容器的系统中处理流体。这些容器在使用之后进行消毒以重复使用。消毒程序昂贵而又麻烦,并且浪费时间。
为了在生产中提供较高的灵活性,并减少实现有效再生所需要的时间,制造商已经开始使用一次性无菌包用于每个生产批次。使用这些一次性包的一个实例是在通过生物处理来生产蛋白质的系统中,在所述生物处理中蛋白质从生长细胞中获得并随后回收。在流体离开所述包的包出口处产生一个问题。当除去流体时,在包内存在的圆锥体形气体形成了一个或多个圆锥形涡旋。这是不理想的,因为涡旋会引起流体和气体的混合,导致不理想地形成泡沫。涡旋还会将空气带入充满蛋白质的流体中。这是不理想的,因为空气会使蛋白质变性。空气带入流体流也会给处理设备例如计量器、传感器、泵和过滤器带来问题。
因此,理想的是提供一种用于流体的一次性容器,该容器具有构件用于使容器出口处的起泡最小化或者防止起泡。
发明内容
本发明提供了一种用于流体的具有入口和出口的一次性容器,该容器具有使出口处起泡最小化或者防止起泡的装置。
本发明还提供了一种系统,其使用所述容器,并在所述容器的下游具有一个单元操作(unit operation),例如常规过滤(NFF)单元等,所处理的流体在此被回收。例如,来自所述容器的流出物可以被引导通过澄清深层过滤器、预过滤器或微孔膜过滤器。具体的单元操作的实例包括澄清、缓冲和介质制备以及病毒去除。
所述出口设置有包含固体表面的漩涡破坏器,其先引导流体离开所述出口。在所述固体表面附近设置开口,其允许流体进入所述出口。先将流体引导离开出口可以最小化或防止在出口处形成一个或多个涡旋。

发明详述
本发明的一次性容器由单层或多层弹性壁形成,所述弹性壁由聚合物组合物形成,所述聚合物组合物例如聚乙烯,包括超高分子量聚乙烯、线性低密度聚乙烯、低密度或中密度聚乙烯;聚丙烯;乙烯乙烯醇共聚物(EVOH);聚氯乙烯(PVC);聚乙酸乙烯酯(PVA);乙烯醋酸乙烯共聚物(EVA共聚物);多种热塑性塑料的混合物;不同热塑性塑料的共挤出物;不同热塑性塑料的多层层合体等。“不同”是指包括不同的聚合物类型,比如聚乙烯层,和一个或多个EVOH的层,以及相同的聚合物类型但具有不同的特性,比如分子量、线性或支化聚合物填料等。典型地使用医用等级,优选地使用对动物无害的塑料(animal-free plastics)。它们通常通过例如蒸汽、环氧乙烷或辐射(例如β或γ辐射)来灭菌。大多数具有良好的抗拉强度、低气体迁移并且是透明或至少半透明的。所述容器设置有入口和出口。所述出口在出口附近设置有漩涡破坏器,并通过例如热封或粘合剂固定于所述容器的内表面。任选地,所述漩涡破坏器可以成型成出口结构。在更优选的实施方案中,所述一次性容器位于固体支持容器内,以易于容器填充或倒空流体。
参考图1,本发明的容器10具有入口9并含有流体12,所述容器包括漩涡破坏器14固定于容器10的出口11的内表面16上。设置泵18以引导流体12通过出口11到达下游单元操作,例如所示的过滤单元20。使用本发明的系统的工艺的实例是由生长细胞来生产蛋白质,其中细胞溶解以释放细胞内蛋白质。随后通过过滤从蛋白质中分离出细胞碎片,从而分离蛋白质。引导滤出液通过管道22到达使用点或者进一步处理。
图2显示了本发明的漩涡破坏器30。该漩涡破坏器包括固体表面32,固体表面32可以是任何形状,包括如图所示的圆形或多边形。基底34密封于容器10的内壁16上。漩涡破坏器30位于出口11附近。固体表面32由连接于固体表面32和基底34的支持体36支持。漩涡破坏器30先引导流体如箭头38和40所示离开出口11,然后引导其如箭头37和39所示通过支持体36之间的空间到达出口11。通过用该漩涡破坏器操作,最小化或防止了涡旋的形成。所述漩涡破坏器也可以形成出口11的一部分。
Claims (4)
1.用于流体的一次性容器,其包含:
由弹性壁形成的封闭容积,
贯穿所述壁之一的入口,
贯穿所述壁之一的出口,
和位于所述出口附近,并固定于所述壁的内表面的漩涡破坏器,所述漩涡破坏器适于先引导流体离开所述出口,随后再引导流体通过所述出口。
2.流体处理系统,其包含:
权利要求1所述的容器,
所述容器下游的单元操作,和
一个或多个管道以使流体从所述容器流到所述单元操作。
3.根据权利要求1所述的容器,其中所述漩涡破坏器包含由空间隔离的固体支持物固定于基底的固体表面。
4.根据权利要求2所述的系统,其中所述漩涡破坏器包含由空间隔离的固体支持物固定于基底的固体表面。
