Figura 4 mostra que a expressão de GT468 promove proliferação de células de câncer de mama. A Figura 5 mostra que GT468 é alvejado por anticorpos de antagonizar função. A análise de proliferação de células de MCF-7 e BT-549 após incubação com quantidades diferentes de anticorpo anti-GT468 e anticorpo de controle (controle de isotipo) por 48 h.

A Figura 7 mostra um teste ELISA de peptídeo para determinar a especificidade de anticorpos criados contra GT468 em sobrenadantes de hibridoma. Sobrenadantes de hibridoma são somente reativos com peptídeos utilizados para imunização.
A Figura 8 mostra coloração de células CHO transfectadas com uma construção GT468-eGFP por sobrenadantes de hibridoma contendo anticorpos criados contra GT468. Os sobrenadantes de hibridoma colorem especificamente células que expressam GT468-eGFP.
A Figura 9 mostra que GT468 é alvejado por anticorpos monoclonais de antagonizar função. A análise de proliferação de linhagens de célula de câncer diferentes após incubação com sobrenadantes de hibridoma por 72 h.
A Figura 10 mostra que lisado bruto (CrELISA) (A) ou ELISA especifico de peptídeo (B) para determinar a especificidade de anticorpos criados contra GT468 em sobrenadantes de hibridoma. Sobrenadantes de hibridoma somente são reativos com o lisado de GT468 (A) ou o peptídeo respectivo utilizado para imunização (B).

A Figura 12 mostra um teste de Western blot para determinar a especificidade de anticorpos criados contra GT468 em sobrenadantes de hibridoma. Todos os sobrenadantes de hibridoma mostraram reatividade específica com lisados de células HEK293 transfectadas com plasmídeo de expressão pcDNA3.1 GT468, ao passo que lisados de células transfectadas simuladas não mostraram sinal. O sinal desvanecido de sobrenadante de hibridoma 23-33A-1 no lisado simulado é devido a derramamento do lisado GT468 HEK.

Sobrenadantes de hibridoma 22-1A-1, 23-33A-1, e 23-19A-1 mostraram cada um, ligação a dois peptideos de sobreposição indicando reatividade com um epitopo linear de GT468. Os padrões de ligação de 22-2A-1 e 22-9B-1 indicam reatividade a um epitopo de conformação (epitopo descontinuo) da proteína GT468.
A Figura 14 mostra que GT468 é alvejado por anticorpos monoclonais de antagonizar função. A análise de proliferação de linhagens de célula de câncer diferentes após incubação com sobrenadantes de hibridoma purificado por 72 h ou 120 h.
A Figura 15 mostra uma análise citométrica de fluxo para determinar a especificidade de anticorpos criados contra GT468 em sobrenadantes de hibridoma. Sobrenadantes de hibridoma mostraram coloração específica de células transfectadas GT468aall6-212, ao passo que nenhuma coloração foi observada em células tranfectadas simuladas.
A Figura 16 A, B mostra uma análise citométrica de fluxo para determinar a ligação específica dos anticorpos monoclonais com células de tumor não fixas expressando endogenamente GT468 (células de câncer de mama MCF-7, MDA-MB-468 e SK-BR-3). Células de câncer gástrico Np-C4 não expressando GT468 foram utilizadas como controle negativo. Os anticorpos são capazes de se ligar a células de tumor que expressam GT468.
A Figura 17 mostra um teste de Western blot para determinar a especificidade de anticorpos criados contra GT468 em sobrenadantes de hibridoma. Todos os sobrenadantes de hibridoma mostraram reatividade especifica com lisados de células HEK293 transfectadas com um plasmídeo de expressão de GT468, ao passo que lisados de células transfectadas simuladas não mostraram sinal.
A Figura 18 mostra que GT468 é alvejado por anticorpos monoclonais de antagonizar função. A análise de proliferação de linhagens de célula de câncer diferentes após incubação com sobrenadantes de hibridoma purificado por 72 h.
A Figura 19 mostra um ensaio clonogênico para análise da atividade de inibição de anticorpos monoclonais para GT468 em formação de colônia de células SK-BR-3 expressando endogenamente GT468. A incubação das células com anticorpo reduziu ou inibiu a formação de colônia.
A Figura 20 A-B mostra a quantificação de expressão de GT468 em tecidos normais utilizando RT-PCR em tempo real. Os tecidos de três indivíduos foram testados para cada tipo de tecido normal. Somente quantidades residuais de transcritos GT468 poderiam ser detectados em tecidos normais após 40 ciclos de RT-PCR. O único tecido normal que excede o corte de expressão (linha tracejada, expressão média de todos os tecidos normais + 3 STDs (99% percentil)) foram placenta e testículos. Barras de erro, STD. Além de câncer de mama (vide a figura 1), expressão elevada de GT468 foi encontrada em amostras de câncer de pulmão, câncer de ovário, câncer gástrico, câncer de próstata, câncer pancreático, câncer de célula renal, câncer hepático, sarcoma, câncer de tiróide, e câncer de cabeça e pescoço.
A Figura 21 mostra a análise western blot de expressão de GT468 em linhagens de célula de câncer. A expressão de GT468 foi detectada em células HEK293 transfectadas com plasmídeo de expressão de GT468 (controle positivo), SK-BR-3 (câncer de mama), BEWO (coriocarcinoma placental), JAR (coriocarcinoma placental), HCT-15 (câncer de cólon), LnCaP (câncer de próstata), HeLA (câncer cervical), MDA-MB-468 (câncer de mama), JEG-3 (coriocarcinoma placental), JIMT-1 (câncer de mama), LA1-55n (Neuroblastoma), PC-3 (câncer de próstata), BT-20 (câncer de mama), e NC1-H929 (mieloma), MelHO (melanoma maligno) e NpC4 (câncer gástrico) foram testados negativos.
A Figura 22 mostra que GT468 é alvejado por anticorpos monoclonais de antagonizar função. Análise de proliferação de células de câncer de mama MDA-MB-468 positiva de GT468 e células de câncer gástrico NpC4 negativa de GT468 após incubação com sobrenadantes de hibridoma purificado por 72 h.
A Figura 23 mostra que Western blot para determinar a especificidade de anticorpos criados contra GT468 em sobrenadantes de hibridoma. Sobrenadantes de hibridoma mostraram reatividade específica com lisados de células HEK293 transfectadas com um plasmídeo de expressão GT468, ao passo que lisados de células transfectadas simuladas não mostraram sinal.
A Figura 26 mostra o tratamento com anticorpos monoclonais anti-GT468 atenua crescimento de tumor xenoenxerto de coriocarcinoma placental BEWO em camundongos nude. Após injeção de células BEWO s.c. os animais foram tratados com anticorpos monoclonais purificados (200 μg) duas vezes por semana por 2 semanas.
A Figura 25 mostra um teste ELISA de peptídeo para identificar epítopos de ligação de anticorpo na proteína GT468. Sobrenadantes de hibridoma 29-8B-1, 35-48B-1, 44-3A-2, 49-3A-1, 4908A-1, 51-1A-1, 53-13A-2 e 62-9B1 cada mostraram ligação a um ou mais peptídeos de sobreposição indicando reatividade a um epítopo linear de GT468. Os padrões de ligação de 29-1A-2 e 54-4B-2 indicam reatividade a um epítopo de conformação (epítopo descontínuo) da proteína GT468.
A Figura 26 mostra o tratamento com anticorpos monoclonais anti-GT468 impede significativamente o espalhamento metastático de células de câncer de mama MCF-7 em ensaios de metástase experimental. Após injeção de 1x106 células MCF-7 i.v. os animais foram tratados com anticorpos monoclonais purificados (200 μg) duas vezes por semana.a carga de tumor metastático nos pulmões de animais foi determinada 5 semanas após inoculação por PCR quantitativo utilizando oligonucleotídeos específicos para DNA microsatélite humano.
Resumidamente, as células polimorfonucleares (PMNs), células NK, monócitos, células mononucleares ou outras células efetoras, de doadores saudáveis podem ser purificadas por centrifpação de densidade de Ficoll Hypaque, seguida por lise de eritrócitos de contaminação. As células efetoras lavadas podem ser suspensas em RPMI suplementado com soro de vitelo fetal inativado a calor a 10% ou alternativamente com soro humano inativado a calor a 5% e misturado com células alvo rotuladas 51Cr que expressam GT468 e/ou sendo caracterizadas por associação de GT468 com sua superfície de célula, em várias razões de células efetoras para células alvo. Alternativamente, as células alvo podem ser rotuladas com um ligando de intensificar fluorescência (BATDA). Um quelato altamente fluorescente de európio com o ligando de intensificação que é liberado de células mortas pode ser medido por um fluorímetro. Outra técnica alternativa pode utilizar a transfecção de células alvo com luciferase. Amarelo lúcifer adicionado pode ser então oxidado por células viáveis somente. IgGs de anti-GT468 purificados podem ser então adicionados em várias concentrações. IgG humano irrelevante pode ser utilizado como controle negativo. Ensaios podem ser realizados por 4 a 20 horas a 37°C dependendo do tipo de célula efetora utilizada. As amostras podem ser ensaiadas para citólise por medição de liberação de 51Cr ou a presença do quelato de EuTDA no sobrenadante de cultura. Alternativamente, luminescência resultando da oxidação de amarelo lúcifer pode ser uma medida de células viáveis.
Bactérias XLOLR E. coli foram transformadas com plasmídeo pQE GT468 ou pQE sem inserção (será mencionado como "referência") e cultivadas em meio LB a A600 nm 0,35 E. a expressão de proteína foi induzida com 2 mM IPTG, e as células foram deixadas crescer por um período adicional de 4 h a 37°C. a indução adequada de expressão de proteína e sua cinética foram monitorados por análise de gel Coomassie. Bactérias foram giradas e suspensas novamente em um pequeno volume de PBS pH 7,2 contendo 0.1 mM de inibidor de protease AEBSF-cloridreto (AppliChem). As células foram colocadas em gelo e rompidas por sonicação (Branson Sonic Power A Smithkline) . Lisados de referência e GT468 foram diluídos a uma concentração total de proteína de 2 mg/ml em PBS contendo 0,2 mM AEBSF e 20% (v/v) de glicerol. Alíquotas foram congeladas por choque em nitrogênio e armazenadas a -70°C até uso.
Antes do uso, GT468, bem como os lisados de referência, foi diluído em tampão de revestimento (100 mM HEPES, pH 7,2), a seguir transferido para placas de microcavidade Maxisorp F96 de fundo redondo (50 ul/cavidade, Nunc) e adsorvido por 2 h a 37°C.

No plasmídeo aminoácidos pDisplay-GT468aall6-212 116-212 de GT468 são terminalmente C fundidas a um domínio de transmembrana de receptor PDGF. Essa construção assegura expressão estável de GT468 aall6-212 na membrana de plasma de células. As células foram coloridas com os sobrenadantes em um estado não fixado que detecta ligação a GT468 em sua conformação nativa. Sobrenadantes de hibridoma mostraram coloração específica de células transfectadas de GT468, ao passo que nenhum coloração foi observada em células transfectadas simuladas (figura 15).
Claims (4)
- Anticorpo caracterizado por ser selecionado do grupo que consiste em:
- (i) um anticorpo produzido por ou obtenível de um clone depositado de acordo com o número de acesso DSM ACC2944 (51-1A-1),
- (ii) um anticorpo que é uma forma quimerizada ou humanizada do anticorpo de acordo com (i), e
- (iii) um anticorpo que compreende a porção de ligação de antígeno ou sítio de ligação de antígeno do anticorpo de acordo com (i),
- Hibridoma caracterizado por ser selecionado do grupo que consiste em:
- (i) um hibridoma que produz o anticorpo conforme definido na reivindicação 1, e
- (ii) um hibridoma depositado de acordo com o número de acesso DSM ACC2944 (51-1A-1).
- Conjugado caracterizado por compreender um anticorpo conforme definido na reivindicação 1 acoplado a um agente terapêutico, preferencialmente, em que o agente terapêutico é uma toxina, um radioisótopo, um fármaco ou um agente citotóxico.
- Composição farmacêutica caracterizada por compreender um anticorpo conforme definido na reivindicação 1 e/ou um conjugado conforme definido na reivindicação 3, e um veículo farmaceuticamente aceitável.
