Abstract

Esta invención se refiere a composiciones y métodos para modificar factores de transcripción de Entrenudos Cortos (SHI) que regulan la fertilidad de la flor, el número de semillas y/o el peso de las semillas en plantas. Reivindicación 1: Una planta o parte de ella que comprende por lo menos una mutación en un gen de factor de transcripción de Entrenudos Cortos (SHI) endógeno que codifica un factor de transcripción de SHI que comprende un dominio de unión de ADN de dedos de zinc (dominio de ZnF), donde la mutación interrumpe la unión del factor de transcripción de SHI a ADN. Reivindicación 25: Una célula vegetal que comprende un sistema de edición que comprende: (a) una proteína efectora asociada con CRISPR; y (b) un ácido nucleico guía (ARNg, ADNg, ARNcr, ADNcr) que tiene una secuencia espaciadora con complementariedad a un gen objetivo endógeno que codifica un factor de transcripción de Entrenudos Cortos (SHI). Reivindicación 34: Una célula vegetal que comprende una mutación en un sitio de unión de ADN de un gen de factor de transcripción de Entrenudos Cortos (SHI) que evita o reduce la unión del factor de transcripción de SHI codificado al ADN, donde la mutación es una sustitución, una inserción y/o una deleción que se introduce usando un sistema de edición que comprende un dominio de unión de ácido nucleico que se une a un sitio objetivo en el gen de factor de transcripción de SHI, donde el gen de factor de transcripción de SHI: (a) comprende una secuencia que tiene por lo menos 80% de identidad de secuencia con cualquiera de las secuencias de nucleótidos de SEQ ID Nº 69, 70, 72 o 73; o comprende una región que tiene por lo menos 80% de identidad con cualquiera de las secuencias de nucleótidos de SEQ ID Nº 75 - 87; o (b) codifica un polipéptido que comprende una secuencia que tiene por lo menos 80% de identidad de secuencia con la secuencia de aminoácidos de SEQ ID Nº 71 o SEQ ID Nº 74 y/o un polipéptido que comprende una región que tiene por lo menos 80% de identidad de secuencia con cualquiera de las secuencias de aminoácidos de SEQ ID Nº 88 - 97. Reivindicación 88: Un ácido nucleico guía que se une a un sitio objetivo en un gen de factor de transcripción de Entrenudos Cortos (SHI), donde el sitio objetivo comprende una secuencia que tiene por lo menos 80% de identidad con cualquiera de las secuencias de nucleótidos de SEQ ID Nº 75 - 87; o que codifica una secuencia que tiene por lo menos 80% de identidad de secuencia con cualquiera de las secuencias de aminoácidos de SEQ ID Nº 88 - 92.
