The three gameplay modes are based off each of the three Darkstalkers games this release is adapted from Darkstalkers 3 (1997), Vampire Savior 2: The Lord of Vampire (1997), and Vampire Hunter 2: Darkstalkers' Revenge (1997). Initially the only gameplay mode which is available is "Vampire Savior" but by choosing a character that the player wishes to customize and use for "Original Character" mode and leveling up your character high enough will unlock the "EX Options" that will allow the player to switch between this mode and the "Vamipire Savior 2" and "Vampire Hunter 2".
While each three games contains six battles, however for Vampire Savior, the final battle is a story-based mode that includes dialogue before the battle begins, while many of these involve the main villain Jedah, for the non-Jedah final battles, he is instead the fifth opponent in the second-to-last opponent. As they were not part the original game, the final battles for Huitzil, Pyron, and Donovan from their battle against Jedah is original to the game.
For the Vampire Hunter 2, it instead follows the same battle final battle set of as the original of facing Huitzil, then Pyron. For Vampire Savior 2 it instead always places Jedah as last, and Pyron before him.
Lastly the characters that appeared in those games are the only ones that can be fought, so aside from the aforementioned characters omitted from Vampire Savior, there is Q-Bee, Lilith, B.B. Hood, and Jedah omitted from Vampire Hunter 2, and Rikuo, Sasquatch, and J. Taliban omitted from Vampire Savior 2 (unless if the player is using one of these aforementioned characters for that particular mode).
