Abstract: RAPD markers were used to measure the genetic diversity of 119 individuals of Ixodes ricinus collected from Lithuania and Norway. The samples were analysed within and also between the populations. We analysed 74 loci in each of 6 populations. Our results show high levels of diversity within the populations. The percentage of polymorphic loci of the six analysed populations: Birzai, Vilnius, Kretinga, Tjore, Kjosvik and Odderoya were 68.9%, 58.1%, 78.38%, 62.2%, 44.6 % and 68.9%, respectively. The percentage of polymorphic loci in the Lithuanian populations was 93.2%, and in the Norwegian populations 81.08%. The genetic distance ranged from 0.019 to 0.079 within Norwegian populations and from 0.005 to 0.0967 within Lithuanian populations and between the countries from 0.022 to 0.146. Large cage and field studies were conducted to compare the efficacy of 2 American Biophysics Corporation mosquito traps, the standard professional (PRO) trap and a new counterflow geometry (CFG) trap. The PRO trap utilizes conventional downdraft technology and the CFG trap uses a patent-pending technology. In large cage studies, similarly baited CFG traps captured approximately 1.7 times as many laboratory-reared Aedes taeniorhynchus as the PRO trap. The CFG trap baited with CO2 + octenol resulted in significantly reduced landing counts compared to all other treatments; mean landing count was reduced from 233.8 (12.99/min), when no trap was present, to 24.7 (1.37/min). In field studies against natural populations of woodland species, the CFG trap captured 7.8 times more mosquitoes than the PRO trap overall, and approximately 11 times more Anopheles crucians, Anopheles quadrimaculatus, and Culex erraticus.

Plant viruses have evolved a wide array of strategies to ensure efficient transfer from one host to the next. Any organism feeding on infected plants and traveling between plants can potentially act as a virus transport device. Such organisms, designated vectors, are found among parasitic fungi, root nematodes and plant-feeding arthropods, particularly insects. Due to their extremely specialized feeding behavior – exploring and sampling all plant tissues, from the epidermis to the phloem and xylem – aphids are by far the most important vectors, transmitting nearly 30% of all plant virus species described to date. Several different interaction patterns have evolved between viruses and aphid vectors and, over the past century, a tremendous number of studies have provided details of the underlying mechanisms. This article presents an overview of the different types of virus-aphid relationships, state-of-the-art knowledge of the molecular processes underlying these interactions, and the remaining black boxes waiting to be opened in the near future.Les virus de plantes ont développé une grande diversité de stratégies pour assurer leur transmission efficace d’un hôte malade vers un nouvel hôte sain. Tout organisme se nourrissant sur une plante infectée, et capable de se déplacer vers d’autres plantes, peut potentiellement transporter des virus. De tels organismes sont dénommés « vecteurs » et se trouvent parmi les champignons phytopathogènes, les nématodes des racines, et les arthropodes, en particulier les insectes. Du fait de leur comportement alimentaire très particulier, explorant et échantillonnant différents tissus végétaux (épiderme, mésophylle et tissus vasculaires), les pucerons sont de loin les vecteurs de virus les plus performants, transmettant près de 30 % des espèces de virus de plantes décrites à ce jour. Plusieurs mécanismes d’interaction virus-puceron très différents ont évolué et, depuis plus d’un siècle, un effort de recherche très important a permis de les décortiquer, au moins pour certaines espèces virales modèles. Cet article présente une synthèse de ces connaissances, les trajets des virus dans leurs pucerons vecteurs, les mécanismes cellulaires et moléculaires de leurs interactions et les grandes inconnues qui persistent et qui devront être élucidées dans les années à venir.

In this article, literature records of argasid and ixodid ticks feeding on humans worldwide are provided in view of increased awareness of risks associated with tick bites. Ticks can cause paralyses, toxicoses, allergic reactions and are vectors of a broad range of viral, rickettsial, bacterial and protozoan pathogens. Approximately 12 argasid species (Argas and Ornithodos) are frequently found attached to humans who intrude into tick-infested caves and burrows. Over 20 ixodid tick species are often found on humans exposed to infested vegetation: four of these are Amblyomma species, 7 Dermacentor spp., 3 Haemaphysalis spp., 2 Hyalomma spp. and 6 Ixodes species. Personal protection methods, such as repellents and acaricide-impregnated clothing are advised to minimize contact with infected ticks. Acaricidal control of ixodid ticks is impractical because of their wide distribution in forested areas, but houses infested with soft ticks can be sprayed with acaricidal formulations. Attached ticks should be removed without delay. The best way is to grasp the tick as close to the skin as possible with fine tweezers and pull firmly and steadily without twisting. Finally, despite the fact that most people who are bitten destroy the offending tick in disgust, it is recommended that they preserve specimens in ethanol for taxonomic identification and detection of pathogens by molecular methods.

Lentiviral vectors have emerged over the last decade as powerful, reliable and safe tools for stable gene transfer in a wide variety of mammalian cells. Unlike other vectors derived from oncoretroviruses, they allow for stable gene delivery into most nondividing primary cells, including neurons. This is why LVs are becoming the most useful and promising tools in the field of neuroscience, not only for research, but also for future gene and cell therapy approaches. Lentivectors (LVs) derived from HIV-1 have gradually evolved to display many desirable features aimed at increasing both their safety and their versatility. These latest designs are reviewed in this unit. This unit also describes protocols for production and titration of LVs that can be implemented in a research laboratory setting, with an emphasis on standardization to improve transposability of results between laboratories.
