Diluted whole blood was layered onto Ficoll gradients (Pharmacia LKB Biotech, Piscataway, NJ) and centrifuged at 400xg for 30 minutes at 22oC. Cells were recovered from the interface of the gradient and washed twice in RPMI 1640 medium (Life technologies Inc., Gaithersburg, MA).
