Abstract

The process comprises the following steps: cultivating biologically and hydroponically in an open container and in the absence of pesticides and additives, seeds of vegetable varieties for example soya, wheat, oats, rice, millet, corn, barley, rye, alfalfa, radish, peanut, peas and/or beans, at a temperature between 18 and 20 °C, and with environment humidity comprised between 70 and 75 % under artificial light, ultraviolet light and ultrasounds, during preferably between five and fifteen days; obtaining an extract of the seeds once germinated; and mixing the extract with other additives and/or excipients. The cosmetic and/or pharmaceutical product obtained has a minimum proportion of 0.01 % by weight of the extract of germinated seeds and is applicable as bath gel, liquid soap, shampoo, conditioner shampoo, hair tonic, after-shave balm, high protection solar cream, medium protection solar milk, face tonic, hydrating cream, serum against skin ageing, make-up products, etc.

State of the art -

In the skin are fibrous proteins (mainly collagen and elastin) and the glycosaminoglycans that constitute the structural elements of this tissue par excellence. The synthesis, that is, the manufacture and development, of these fibrous proteins takes place in cells known as fibroblasts, thanks to the presence of oxygen and other necessary nutrients, such as, for example, adenosine triphosphate, nucleotide best known in the art by ATP. - two -

With the passage of time, the synthesis of new molecules in the skin is happening more slowly, because the metabolism of fibroblasts decreases with age. The synthesis processes of these fibrous proteins slow down and the difficulty of accessing the compounds necessary for the synthesis increases. Also, the existing fibers suffer a series of alterations in their structure, which cause the appearance of skin flaccidity and wrinkles in the skin, ultimately associated with the maturity and aging of the person.

To prevent and mitigate the effects of aging and wrinkled skin until now, the use of cosmetic and pharmaceutical products based on hydrolysates of animal placenta extract has been used almost exclusively.

These dermatological products based on placental extracts base their reparative action on a set of molecules with biological activity, capable of penetrating the innermost layers of the skin, providing the energy necessary to increase the synthesis of the said fibrous proteins constituting the skin tissue, producing the beneficial effect on human skin of preventing and delaying the appearance of signs of skin aging.

Currently, due to ethical and legal considerations, all cosmetic and pharmaceutical products of animal origin are gradually being replaced by substances of plant origin, in particular seed extracts, the action of which is based on them principles and substances that placenta extracts, acting in the same way.

Although the use of seed and germ extracts has long been widely known in fields such as food, being ancestral in some regions, such as in eastern countries, its use is currently being extended to other fields of the technique, like cosmetics. Thus, a multiplicity of documents and publications describe cosmetic and pharmaceutical procedures and products applicable to the prevention of skin aging, based on the use of hydrolyzates of seed germ extracts, such as, for example, patents JP-91/179050, FR- 92/7830 and ES-91/2496.

In all these applications, seed germ extracts are used, prior to germination, in order to stimulate the metabolism of the skin.

The use of seed extracts after germination is also known, either as antioxidant agents or as a means of preventing the occurrence of so-called free radicals, such as the procedure described in Japanese patent JP-93/332374.

However, until now the application of germinated seed extracts as metabolic stimulating agents is not known.

The purpose of the present invention is to provide a novel process for the preparation of a pharmaceutical and / or cosmetic preparation, as well as the product obtained from such a procedure, that fills the vacuum existing in the art.

Explanation of the invention. -

For this purpose, a first object of the present invention is a process for the preparation of a pharmaceutical and / or cosmetic product, of novel concept, which in essence is characterized in that said step of obtaining the extract is carried out after germination of the seeds grown biologically in water, in the absence of pesticides and additives, for a predetermined period of time.

According to another characteristic of the process of the invention, the germination of the seeds is hydroponically stimulated in an open container culture, at a temperature between 18 and 20 ° C, an ambient humidity between 70 and 75%, under artificial light , ultraviolet light and ultrasound.

According to a preferred embodiment of the process object of the present invention, said predetermined period of time is between five and fifteen days.

Next, the procedure of the present invention, the pharmaceutical and / or cosmetic products obtained with such a procedure are explained in detail, and the tests performed "in-vivo" and "in-vitro" for the verification of pharmacological efficacy and Cosmetic of the germinated seed extracts of the present invention.

The procedure consists in itself of a stage of cultivation and germination of seeds, a stage of obtaining an extract of the germinated seeds (germinated) and a stage of addition of components to the germinated.

The sprouts used are preferably obtained by the so-called hydroponic system. As is known, the hydroponic system consists, in a generic way, in the practice of plant crops in the absence of soil and in an artificial environment.

However, another known method other than hydroponic, either conventional or any other, can be used since it has been found that the system itself does not affect the final composition of the extracts.

The hydroponic germination system was applied to two biological crops: one of wheat seeds, and another formed by a mixture of wheat, soy, alfalfa and radish seeds. Both cultures were maintained with only water and in the absence of pesticides and other chemical agents, at a temperature between 18 and 20 ° C, with a - 11 - ambient humidity between 70 and 75% and with artificial light. In both crops, the seeds underwent a germination process from the first day in the same container without the need to handle the product once the process was finished. In order to maintain the constancy of air quality, during the germination process it was applied to ultrasound and ultraviolet light cultures.

The hydroponic method is advantageous with respect to the other possible cited as it allows the growth of sprouts with the formation of a microenvironment, not caked and with longitudinal growth, as well as with a lower concentration of water present, which favors its conservation. On the other hand, the fact that sprouts are deposited in open tanks facilitates metabolic processes. In this case, the synthesis of chlorophyll has been verified by direct light.

In addition, the hydroponic culture system is the most suitable for measuring the optimum levels of vital components in the germinated seed that must be active ingredients in the future extract, that is, monosaccharides, oligosaccharides, peptides, amino acids, minerals and vitamins.

The germination process was interrupted when the measured concentrations of these elements had a maximum. Germination could have been just ^¬ interrupted when the rate of increase of such - 12 - concentrations begin to decrease. Preferably, germination stops after five to fifteen days of cultivation. In the particular tests carried out, germination was interrupted on the seventh day of its beginning.

After germination, the seeds of both crops were subjected to rapid freezing at temperatures below -20 ° C, and were kept frozen until they were extracted.

The extraction of the germinated seeds began by grinding and disintegrating them in their frozen form. Then the extraction was carried out by maceration using a hydroalcoholic extraction solvent containing 20% by weight of 98% ethyl alcohol, maintaining a plant-solvent extraction ratio of 1: 2, at a maximum temperature of 80 ° C for 15 minutes. The extraction process lasted for 24 hours.

After separating the extraction liquids, this same extraction process was repeated, but with a plant-solvent extraction ratio of 1: 1.

Finally, the extraction liquids were joined, performing a filtration on paper first and then a filtration by membrane filters until particle sizes of less than 1 millimeter were obtained.

The product obtained after filtration was a fully transparent, crisp, active substance with - 13 - absence of visible particles.

To evaluate the action of germinated seed extracts on cell metabolism, "in vitro" tests were performed using two different models: that of hepatic mitochondria; and that of human skin fibroblast cell cultures, measuring oxygen consumption by an electrode called "Clark." Both models are scientifically recognized as appropriate for studying and measuring the stimulating action of compounds on cell metabolism.

The results obtained in both showed that with a concentration of 0.007% of the active product a stimulation on the activity of mitochondria and fibroblasts exceeding 70% was obtained, which demonstrates the excellent action of the extracts according to the invention on cell metabolism when, for example, to obtain similar stimulations with animal placenta extracts, concentrations of around 10% are necessary.

To verify the "in vivo" efficacy of both germinated seed extracts, or active ingredients, "non-invasive" biophysical testing techniques were carried out.

First, the hydration of the skin surface was measured, using two cosmetic formulations, one of which contained 3% of the active substance, while the other was a formula- - 14 - control or placebo tion. Through a corneometer (CM 825 Courage & Khazaka) the capacitance values of the skin surface were recorded. This method is based on the dielectric constant of water, usually 81 units of capacitance, while the rest of substances have values lower than 7 units, so that the measured capacitance changes are a measurable function of water levels, and therefore of skin hydration. The experiment was designed as an experimental, randomized and "double blind" study. The results obtained show that the use of cosmetic and pharmaceutical formulations containing germinated seed extracts increase skin hydration.

The results of the hydration tests were similar for both extracts, that is, the one formed from wheat seed crops and the one formed by a mixture of wheat, soy, alfalfa and radish seeds, both showing an increase in the hydration of a 10% versus the placebo formula.

Parallel to the skin hydration test, the effectiveness of the germinated seed extracts in a cosmetic formulation was also evaluated, measuring on the one hand their effect on the mechanical properties of the skin (elasticity) and then performing an image analysis, which It is the technique used to observe the effect on the skin surface and in particular on wrinkles, before and after treatment. - fifteen -

The skin elasticity of the skin was analyzed by measuring its degree of recovery after applying a certain suction pressure performed using a device known in the art as "Cutometer" (model SEM 474 of Courage & Khazaka).

The test was performed by applying a cosmetic formulation of the extract according to the invention and a placebo formulation on the cheek of 6 volunteers, applying low pressure (300 mbar). The measurement was made three times to each volunteer before starting the treatment and another three after the end of it. The values of the parameters R9 (experimental elasticity value), R0 (maximum extension value) and Rl (recovery capacity) were obtained, which are shown in Table I below:

23 -REIVINDICATION
