Abstract

The present invention is based on the fact that an oil, e.g. corn oil, is not soluble in aqueous sample, e.g. a plasma sample. If the aqueous sample contains proteins which are natural binders for a ligand which have hydrophobic properties, then the ligand is present in the free state in both the oil phase and the water phase, while bound ligand is only present in the water phase. If a labeled version of the ligand is present in the system, it is possible to determine the percentage of free ligand in the aqueous sample under equilibrium conditions, provided that the partition coefficient of free ligand also is determined. The percentage of free ligand, together with the total concentration of the ligand, yields the free concentration of the ligand.
