Claims (17)
- 비-자연적으로 발생하는 미생물의 자연적으로 발생하는 야생형 대응부 (counterpart)에서의 포스포에놀 피루베이트 카르복시키나제 효소 활성에 비해 증가된 수준의 내인성 pck 유전자에 의해 인코딩된 포스포에놀 피루베이트 카르복시키나제 효소 활성 및 피루베이트 키나제 효소 활성의 감소된 수준을 유발하는 pykA 유전자 또는 그의 상동체에서의 돌연변이를 포함하는 비-자연적으로 발생하는 미생물에 있어서, 상기 증가된 수준의 포스포에놀 피루베이트 카르복시키나제 효소 활성은 포스포에놀 피루베이트 카르복시키나제 효소의 활성을 증가시키는 돌연변이로부터 기인하는 것을 특징으로 하는, 비-자연적으로 발생하는 미생물.
- 제 1항에 있어서,
비-자연적으로 발생하는 미생물의 자연적으로 발생하는 야생형 대응부에서의 포스포에놀 피루베이트 카르복시키나제 효소 활성에 비해 증가된 수준의 내인성 pck 유전자에 의해 인코딩된 포스포에놀 피루베이트 카르복시키나제 효소 활성은, 내인성 pck 유전자의 프로모터 영역에서의 돌연변이 또는 상기 내인성 pck 유전자의 프로모터 영역과 상호작용하는 것으로 알려진 하나 이상의 조절 요소의 유전자 조작 중 어느 하나에 기인한 것을 특징으로 하는, 비-자연적으로 발생하는 미생물. - 제 1항에 있어서,
포스포에놀 피루베이트-의존성 포스포트랜스퍼라제 시스템의 기능성을 저하시키는 하나 이상의 유전자 변형을 더 포함하는 것을 특징으로 하는, 비-자연적으로 발생하는 미생물. - 제 1항에 있어서,
(a) 포스포에놀 피루베이트-의존성 포스포트랜스퍼라제 시스템의 기능성을 저하시키는 하나 이상의 유전자 변형; 및
(b) 비-포스포트랜스퍼라제 시스템 당 이송자의 활성을 증가시키는 하나 이상의 유전자 변형을 더 포함하는 것을 특징으로 하는, 비-자연적으로 발생하는 미생물. - 제 4항에 있어서,
상기 비-포스포트랜스퍼라제 시스템 당 이송자는 갈락토오스 퍼미아제 (galactose permease)인 것을 특징으로 하는, 비-자연적으로 발생하는 미생물. - 제 5항에 있어서,
상기 갈락토오스 퍼미아제의 활성의 증가는 상기 갈락토오스 퍼미아제를 코딩하는 galP 유전자의 복제 수를 증가시키는 것에 의하여 달성되는 것을 특징으로 하는, 비-자연적으로 발생하는 미생물. - 제 5항에 있어서,
상기 갈락토오스 퍼미아제의 활성의 증가는, 하나 이상의 돌연변이를 통해 상기 galP 유전자 억제자의 음성 조절을 완화하는 것에 의하여 달성되는 것을 특징으로 하는, 비-자연적으로 발생하는 미생물. - 제 7항에 있어서,
상기 갈락토오스 퍼미아제의 억제자는 galS 또는 galR 유전자에 의하여 인코딩되는 것을 특징으로 하는, 비-자연적으로 발생하는 미생물. - 제 1항에 있어서,
락테이트 디하이드로게나제 A(lactate dehydrogenases A), 피루베이트 포메이트 리아제(pyruvate formate lyase), 알콜 디하이드로게나제 E(alcohol dehydrogenase E), 포스포트랜스-아세틸라제(phosphotrans-acetylase), 아세테이트 키나제 A(acetate kinase A) 및 피루베이트 옥시다제 B(pyruvate oxidase B)로 이루어진 군으로부터 선택된 발효적 경로에 관련된 유전자들 중 적어도 하나의 기능성을 저하시키는 돌연변이를 더 포함하는 것을 특징으로 하는, 비-자연적으로 발생하는 미생물. - 제 1항에 있어서,
트리카르복실산 회로의 작동과 관련된 말레이트 디하이드로게나제(malate dehydrogenase) 유전자에서의 돌연변이를 더 포함하되, 상기 돌연변이는 말레이트 디하이드로게나제 효소 활성을 증가시키는 것을 특징으로 하는, 비-자연적으로 발생하는 미생물. - 제 1항에 있어서,
RNA 폴리머라제의 알파 서브유닛을 인코딩하는 서열번호: 4로 제시된 rpoA 유전자에서의 돌연변이를 더 포함하되, 상기 돌연변이는 상기 rpoA 유전자의 뉴클레오티드 위치 956에서 시토신 잔기를 티민 잔기로 치환하는 점 돌연변이이며, 이에 의하여 상기 RNA 폴리머라아제의 알파 서브유닛의 아미노산 위치 322에서 프롤린 잔기가 류신 잔기로 치환되는 것을 특징으로 하는, 비-자연적으로 발생하는 미생물. - 제 1항에 있어서,
RNA 폴리머라제의 서브유닛을 인코딩하는 서열번호: 5로 제시된 rpoC 유전자에서의 돌연변이를 더 포함하되, 상기 돌연변이는 상기 rpoC 유전자의 뉴클레오티드 위치 2241에서 구아닌 잔기를 아데닌 잔기로 치환하는 점 돌연변이이며, 이에 의하여 상기 RNA 폴리머라아제의 서브유닛의 아미노산 위치 747에서 메티오닌 잔기가 이소류신 잔기로 치환되는 것을 특징으로 하는, 비-자연적으로 발생하는 미생물. - 제 1항에 있어서,
상기 미생물의 염색체 DNA 내에서의 결실을 더 포함하되, 상기 결실은 ydcC , ydcD, ydcE , 및 ydcF 유전자를 제거하는 것을 특징으로 하는, 비-자연적으로 발생하는 미생물. - 제 1항에 있어서,
세포벽 합성 및 세포 분리에 관련된 단백질을 코딩하는 서열번호: 8로 제시된 ftsI 유전자에서의 돌연변이를 더 포함하되, 상기 돌연변이는 상기 ftsI 유전자의 뉴클레오티드 위치 619에서 시토신 잔기를 티민 잔기로 치환하는 것이며, 이에 의하여 상기 세포벽 합성 및 세포 분리에 관련된 단백질의 아미노산 위치 207에서 아르기닌 잔기가 시스테인 잔기로 치환되는 것을 특징으로 하는, 비-자연적으로 발생하는 미생물. - 제 1항에 있어서,

포스포트랜스퍼라제-의존성 디하이드록시아세톤 키나제 효소의 서브유닛을 인코딩하는 서열번호: 7로 제시된 dhaM 유전자에서의 돌연변이를 더 포함하되, 상기 돌연변이는 상기 포스포트랜스퍼라제-의존성 디하이드록시아세톤 키나제 효소 활성의 감소를 유발하는 틀 이동 돌연변이인 것을 특징으로 하는, 비-자연적으로 발생하는 미생물. - (a) 제 1항 내지 제 16항 중 어느 한 항에 따른 미생물을 배양하는 단계;
(b) 탄소원을 제공하는 단계;
(c) 상기 미생물이 상기 탄소원을 대사하도록 허용하는 단계; 및
(d) 임으로, 숙신산을 분리하는 단계
를 포함하는 것을 특징으로 하는, 숙신산을 생산하는 방법.
