Abstract
Description
도 2는 잡음 전류와 이온 채널 전류를 밀봉 저항과 막전위의 변동 전압 ΔVm의 함수로서 나타낸다.
도 3은 제 1 실시예에 의한 전극부의 상세한 구조를 단면도로 나타낸다.
도 4는 제 1 실시예에서 실제로 제작한 전극부의 사진을 나타낸다.
도 5는 종래의 AgCl/Ag 전극(「Normal AgCl/Ag electrode」라고 표기)을 사용한 경우와 본 발명의 전극부(「Stable AgCl/Ag electrode」라고 표기)를 사용했을 경우의 인가 전압 0mV에서의 전류변동의 계측 결과를 나타낸다.
도 6(a)는 제 2 실시예에 의한 플레이너 패치 클램프 장치의 요부 단면도를 나타내고, 도 6(b)는 복수의 원형의 오목부(24)와, 이들을 서로 연결하는 가는 홈의 평면도를 나타낸다.

Claims (13)
- (1) 전기절연성 기판에 그 양측의 표면을 연통시키며, 세포를 통과시키지 않지만 액체를 통과시킬 수 있는 미세한 관통 구멍을 1개이상 형성하고, (2) 상기 관통 구멍의 제 1 표면측과 제 2 표면측에는 각각 도전성 액체를 유지하기 위한 액저류부와, 그 액저류부의 도전성 액체에 대해서 통전 가능하게 배치된 전극부를 형성하고, (3) 제 1 표면측의 액저류부가 세포 배치용 액저류부로 되고, 또한 (4) 상기 제 1 표면측 및 제 2 표면측의 전극부가 이하의 (a)∼(c)의 요소를 구비하는 것을 특징으로 하는 플레이너 패치 클램프 장치.
(a) 상기 액저류부에 도전성 액체가 도입되었을 때에 그 도전성 액체에 접하게 되는 용기벽의 적어도 일부가 무기 다공질 재료로 구성되어 있는 전극 용기.
(b) 상기 전극 용기내에 수용되며, 귀금속(Nm)의 표층부에 그 귀금속의 염화물(NmCl)층을 형성한 전극.
(c) 상기 전극 용기내에 충전되며, 상기 귀금속의 염화물(NmCl) 및 알칼리 금속염화물이 포화 농도로 용해된 전극 용액. - 제 1 항에 있어서,
상기 제 1 표면측의 액저류부는 광불투과성의 재료에 의해 구성되며 세포를 배치하기 위한 제 1 액저류부와, 제 1 표면측의 전극부가 배치된 제 2 액저류부와, 이들 주액저류부 및 부액저류부를 연통시키는 좁은 통액로로 이루어지는 것을 특징으로 하는 플레이너 패치 클램프 장치. - 제 1 항 또는 제 2 항에 있어서,
상기 제 2 표면측의 액저류부가 도전성 액체를 도입 및 배출하기 위한 통액로와 연통되고, 또한 이 통액로에 제 2 표면측의 전극부가 배치되어 있는 것을 특징으로 하는 플레이너 패치 클램프 장치. - 제 1 항 내지 제 3 항 중 어느 한 항에 있어서,
상기 제 1 표면측에 액저류부를 복수 형성하고, 이들 액저류부에는 신경 세포의 세포체보다 큰 폭과 배양 상태의 세포체의 이동을 저지할 수 있는 깊이를 갖는 오목부를 형성하고 있는 것을 특징으로 하는 플레이너 패치 클램프 장치. - 제 1 항 내지 제 4 항 중 어느 한 항에 있어서,
상기 플레이너 패치 클램프 장치가 이하의 (A) 및/또는 (B)의 구성을 구비하는 것을 특징으로 하는 플레이너 패치 클램프 장치.
(A) 제 2 표면측의 액저류부가 액체 흡인 디바이스에 연결되어 있고, 이 액체 흡인 디바이스에 의해 제 2 표면측의 액저류부에 음압을 부하할 수 있게 되어 있다.
(B) 기판의 관통 구멍에 있어서의 제 1 표면측의 개구부 둘레가장자리에 세포 고정력을 갖는 세포외 매트릭스 형성 물질을 부착시키고 있다. - 이하의 (a')∼(c')의 요소를 구비하는 것을 특징으로 하는 플레이너 패치 클램프 장치용 전극부.
(a')용기벽의 적어도 일부가 무기 다공질 재료로 구성된 전극 용기.
(b')상기 전극 용기내에 수용되며, 귀금속(Nm)의 표층부에 그 귀금속의 염화물(NmCl)층을 형성한 전극.
(c')상기 전극 용기에 충전되며, 상기 귀금속의 염화물(NmCl) 및 알칼리 금속염화물이 포화 농도로 용해된 전극 용액. - 제 6 항에 있어서,
상기 용기벽의 적어도 일부를 구성하는 무기 다공질 재료가 다공질 유리 또는 다공질 세라믹스인 것을 특징으로 하는 플레이너 패치 클램프 장치용 전극부. - 제 6 항 또는 제 7 항에 있어서,
상기 귀금속(Nm)이 은(Ag) 또는 백금(Pt)이며, 알칼리 금속염화물이 염화칼륨(KCl)인 것을 특징으로 하는 플레이너 패치 클램프 장치용 전극부. - 제 6 항 내지 제 8 항 중 어느 한 항에 있어서,
상기 전극이 이하 (C) 또는 (D)인 것을 특징으로 하는 플레이너 패치 클램프 장치용 전극부.
(C) 전극 용기 내부로 돌출된 봉상 전극으로서, 귀금속(Nm)으로 이루어지는 심재의 표층부에 귀금속 염화물(NmCl)층을 형성하고 있다.
(D) 전극 용기의 벽부의 내주면에 형성된 통상 전극으로서, 용기벽부측의 바닥층이 귀금속(Nm)의 증착층이며, 전극 용액에 접하는 표면층이 귀금속 염화물(NmCl)의 증착층이다. - 제 1 항 내지 제 5 항 중 어느 항에 기재된 플레이너 패치 클램프 장치의 제 2 표면측의 액저류부에 도전성 액체를 도입함과 아울러 제 1 표면측의 액저류부에는 측정 대상인 세포를 분산시킨 도전성 액체를 도입해서 제 1 표면측의 액저류부와 제 2 표면측의 액저류부를 도전적으로 연통시키고, 상기 세포를 제 1 표면측의 액저류부의 소정 위치에 배치한 상태에서 제 1 표면측 및 제 2 표면측의 전극부의 전극간에 전압을 인가하여 측정 대상인 세포의 이온 채널 전류를 계측하는 것을 특징으로 하는 세포 이온 채널 전류 계측 방법.
- 제 10 항에 있어서,
상기 플레이너 패치 클램프 장치의 기판의 제 1 표면측에는 제 4 항에 기재된 구성의 복수의 액저류부를 형성하고, 측정 대상인 세포가 신경 세포이며, 도전성 액체가 상기 신경 세포의 세포 배양액인 것을 특징으로 하는 세포 이온 채널 전류 계측 방법. - 제 11 항에 있어서,
상기 신경 세포에는 미리 Ca 이미징을 위한 Ca 프로브가 도입되어 있고, 그 세포 이온 채널 전류를 적어도 세포 활동 전위의 발생시 또는 활동 전위의 전파시에 발생하는 형광의 관찰인 Ca 이미징을 포함하는 수단에 의해 측정하는 것을 특징으로 하는 세포 이온 채널 전류 계측 방법. - 제 12 항에 있어서,
상기 Ca 프로브가 도입된 신경 세포를 상기 플레이너 패치 클램프 장치의 제 1 표면측에 복수 내지 다수 배치함과 아울러 이들 신경 세포의 시냅스가 서로 연접된 신경 세포 네트워크로 해 두고, 이들 중 단일의 신경 세포에 전류 주입 또는 전압 인가함으로써 상기 복수 내지 다수의 신경 세포에 있어서의 상기 Ca 이미징에 의한 측정을 행하는 것을 특징으로 하는 세포 이온 채널 전류 계측 방법.
