Claims (15)
- 엔도톡신을 검출하는 방법에 있어서, 상기 방법은 다음의 단계들로 구성됨을 특징으로 하는 엔도톡신의 검출방법.a) 박테리오파지 미부 단백질과 시료를 배양하는 단계,b) 박테리오파지 미부 단백질에 결합된 엔도톡신을 검출하는 단계.
- 제 1항에 있어서, 상기 방법은 a)단계 이후와 b)단계 이전에a') 상기 시료로부터 상기 박테리오파지 미부 단백질-엔도톡신 복합체를 분리하는 단계를 더 포함하는 것을 특징으로 하는 방법.
- 제 1항 또는 제 2항에 있어서, 상기 검출은 분광법에 의하여 행해지는 것을 특징으로 하는 방법.
- 시료로부터 엔도톡신을 제거하는 방법에 있어서, 상기 방법은 다음의 단계로 구성됨을 특징으로 하는 엔도톡신 제거방법.a) 상기 시료를 비특이적으로 또는 지향적으로 영구 캐리어에 고정화된 박테리오파지 미부 단백질과 함께 배양하거나 또는 이에 접촉시키는 단계,b) 상기 시료로부터 상기 박테리오파지 미부 단백질-엔도톡신 복합체를 분리하는 단계.
- 제 4항에 있어서, 상기 a) 및 b) 단계는 크로마토그래피 컬럼 관통류법으로 행해지는 것을 특징으로 하는 방법.
- 제 4항에 있어서, 상기 영구 캐리어는 여과 매질, 글래스 입자, 자기입자, 원심 물질, 침강 물질 또는 크로마토그래피 컬럼용 충진 물질로 되는 것을 특징으로 하는 방법.
- 제 4항 내지 제 6항 중 어느 한 항에 있어서, 상기 박테리오파지 미부 단백질은 결합군들을 통하여 상기 영구 캐리어에 고정화되는 것을 특징으로 하는 방법.
- 제 7항에 있어서, 상기 결합군은 렉틴, 수용체, 안티칼린으로 되는 것을 특징으로 하는 방법.
- 제 7항에 있어서, 상기 결합군은 스트렙타비딘 또는 아비딘으로 되고, 상기 박테리오파지 미부 단백질은 비오틴 또는 스트렙-태그와 결합되는 것을 특징으로 하는 방법.
- 제 4항 내지 제 6항 중 어느 한 항에 있어서, 상기 박테리오파지 미부 단백질은 화학적 결합을 통하여 이중결합적으로 상기 영구 캐리어에 고정화되는 것을 특징으로 하는 방법.
- 제 1항 내지 제 10항 중 어느 한 항에 있어서, 상기 박테리오파지 미부 단백질은 스트렙-태그 또는 히스-태그를 가지는 것을 특징으로 하는 방법.
- 제 11항에 있어서, 상기 태그는 SEQ ID NO. 5, 6, 7에 의한 아미노산 서열을 가지는 것을 특징으로 하는 방법.
- 제 11항 또는 제 12항에 있어서, 상기 파지 T4의 p12 단백질은 박테리오파지 미부 단백질로서 사용되는 것을 특징으로 하는 방법.
- 제 1항 내지 제 13항 중 어느 한 항에 있어서, 상기 배양에서의 Ca2+ 농도는 0.1μM 내지 10mM이고, Mg2+ 농도는 0.1μM 내지 10mM인 것을 특징으로 하는 방법.
- 제 1항 내지 제 3항 중 어느 한 항에 있어서, 표적 엔도톡신은 박테리오파지 미부 단백질과의 결합으로부터 제거되고, 이어서 상기 표적 엔도톡신이 검출되는 것을 특징으로 하는 방법.
