실시예 1 및 실시예 2의 조성물의 미백 효과를 확인하기 위하여 합성된 멜라닌의 생성을 저해하는지 확인하였다. 구체적으로, B16F1 세포를 6 well plate에 분주한 후, 세포 배양 조건에서 24시간 배양하였다. 이후 대조군과 시험물질인 각 조성물 1%를 세포에 처리하였고, 멜라닌 합성을 위해 α-MSH를 최종 10 nM가 되도록 같이 첨가하여 24시간 동안 추가 배양하였다. 이때 조성물 내 포함된 10 % 스트렙토코쿠스 테르모필루스발효물, 30 mg/L 클로로제닉애씨드, 10 mg/L 페룰릭애씨드 및 10 mg/L 레스베라트롤 단일 물질 4종류도 각각 1 %씩 같이 처리하였다. 이어 배양액을 제거하고 PBS를 이용하여 3 회 세척한 뒤 1 N NaOH 용액 300 μL씩 첨가하여 90℃ 1시간 동안 세포를 용해시켰다. 세포 내 멜라닌 양은 492 nm에서 흡광도를 측정하여 구하였으며, 그 결과를 표 3에 나타내었다.In order to confirm the whitening effect of the compositions of Examples 1 and 2, it was confirmed whether they inhibited the production of synthesized melanin.
