Abstract
Description
도 2는 일 양상에 의한 형광 염료로 표지한 엑소좀을 투과전자현미경으로 관찰한 결과이다.
도 3은 일 양상에 의한 형광 상관 분광법을 이용하여 각 엑소좀 샘플의 시간에 따른 형광 강도 및 형광 상관 함수를 측정한 결과이다.
도 4는 일 양상에 의한 형광 상관 분광법을 이용하여 각 엑소좀 샘플에서 하나의 엑소좀에 결합한 형광 염료의 개수를 계산한 결과이다.

Claims (16)
- 형광 염료가 표지된 세포외소포체(extracellular vesicle) 분자를 제공하는 단계;
상기 형광 염료가 표지된 세포외소포체 분자 및 상기 형광 염료 분자에 대하여 형광 상관 분광법(Fluorescence correlation spectroscopy, FCS)을 수행하여 상기 각각의 분자의 휘도(brightness)를 결정하는 단계; 및
형광 염료가 표지된 세포외소포체 분자의 휘도 대비 형광 염료 분자의 휘도 값을 결정하여 1개의 세포외소포체 당 표지된 형광 염료 분자의 개수를 결정하는 단계를 포함하고,
상기 형광 상관 분광법은 형광 염료로 표지된 세포외소포체 분자를 포함하는 샘플에 여기(excitation) 레이저를 조사한 후, 발생하는 형광 신호를 검출기로 측정하여 형광 상관 함수를 도출하는 단계; 상기 형광 상관 함수를 분석하는 단계; 및 상기 샘플의 형광 휘도 및 상기 샘플에 포함된 세포외소포체 분자 개수를 이용하여 상기 세포외소포체 분자 1개 당 형광 휘도를 계산하는 단계를 포함하며,
상기 형광 염료가 표지된 세포외소포체 분자는 형광 염료가 화학적으로 표지된 것인, 세포외소포체에 표지된 형광 염료를 정량화하는 방법. - 청구항 1에 있어서,
상기 분자의 휘도는 Hz 단위로 표시되는 것이고, 특정 분자의 절대값인 것인 방법. - 삭제
- 청구항 1에 있어서,
상기 1개의 세포외소포체 당 표지된 형광 염료 분자의 개수 값은 정수이고, 정수가 아닌 경우 소수점 첫째 자리에서 반올림된 정수를 결정하는 것인 방법. - 청구항 1에 있어서,
상기 형광 염료가 표지된 세포외소포체 분자는 서로 다른 휘도 값을 갖는 복수 개의 형광 염료가 표지된 세포외소포체 분자인 것인 방법. - 청구항 1에 있어서,
상이한 조건의 변수를 갖는 분자의 휘도를 결정하여 상관 관계 함수를 도출하는 단계를 포함하는 것인 방법. - 청구항 6에 있어서,
상기 상이한 조건의 변수는 세포외소포체의 크기, 또는 개수, 세포외소포체에 처리된 형광 염료의 농도, 및 세포외세포체에의 형광 염료의 표지에 영향을 미칠 수 있는 인자로 이루어진 군으로부터 선택되는 것인 방법. - 삭제
- 청구항 1에 있어서,
상기 형광 염료는 플루오레세인(fluorescein), 플루오레세인 클로로트리아지닐(fluorescein chlorotriazinyl), 플루오레세인 아이소싸이오사이아네이트(Fluorescein isothiocyanate, FITC), 로다민 그린(rhodamine green), 로다민 레드(rhodamine red), 테트라메틸로다민(tetramethylrhodamine), 오레곤 그린(Oregon green), 알렉사 플루오로(Alexa Fluor), JOE, ROX, HEX, 텍사스 레드(Texas Red), TET, TRITC, TAMRA, 시아닌(Cyanine) 계열 염료 및 씨아디카르보시아닌(thiadicarbocyanine) 염료로 이루어진 군으로부터 선택되는 것인 방법. - 청구항 1에 있어서,
상기 형광 염료는 N-하이드록시석시니마이드 에스테르(N-Hydroxysuccinimide ester, NHS ester), 이소티오시아네이트(isothiocyanates), 카르복실산(carboxylic acids) 및 술포닐 클로라이드(sulfonyl chlorides)로 이루어진 군에서 선택된 어느 하나의 반응 유도체인 방법. - 청구항 1에 있어서,
상기 세포외소포체 분자는 엑소좀(exosome), 미세소포체(microvesicle), 인트라루미날 소포체 (intraluminal vesicle), 다중 소포체(multi vesicular body), 다중 소포 엔도좀(multivesicular endosome) 및 엔도좀 또는 원형질막에서 분리된 소포체로 이루어진 군으로부터 선택되는 것인 방법. - 삭제
- 청구항 1의 방법을 통해 미리 결정된 복수의 군의 특정 농도의 형광 염료가 표지된 세포외소포체 분자의 휘도 값에 대한 상관 관계 함수에 미지의 시료의 형광 염료가 표지된 세포외소포체 분자의 휘도 값을 대입하여 상기 미지의 시료의 세포외소포체 분자를 정량화하는 단계를 포함하는 세포외소포체를 정량화하는 방법.
- 인 비트로(in vitro) 또는 인 비보(in vivo) 상에서 처리된 또는 투여된 후보물질을 포함하는 세포외소포체 또는 후보물질인 세포외소포체를 청구항 1 또는 청구항 13의 방법을 이용하여 평가하는 단계를 포함하는 치료제 후보물질을 평가하는 방법.
- 생체 내 타겟 단백질을 검출하는 방법으로서,
상기 타겟 단백질 또는 상기 타겟 단백질과 결합하는 단백질을 발현하는 세포주로부터 분리되고, 형광 염료가 표지된 세포외소포체 분자를 제공하는 단계;
상기 형광 염료가 표지된 세포외소포체 분자에 대하여 형광 상관 분광법(Fluorescence correlation spectroscopy, FCS)을 수행하여 처리된 형광 염료의 농도에 따라 1개의 세포외소포체 분자당 표지된 형광 염료의 개수를 정량화하는 단계; 및
타겟 단백질을 포함하는 분리된 생물학적 시료를 상기 표지된 형광 염료의 개수가 조절되고, 정량화된 세포외소포체 분자와 결합시킨 후 형광을 검출하는 단계를 포함하고,
상기 형광 상관 분광법은 형광 염료로 표지된 세포외소포체 분자를 포함하는 샘플에 여기(excitation) 레이저를 조사한 후, 발생하는 형광 신호를 검출기로 측정하여 형광 상관 함수를 도출하는 단계; 상기 형광 상관 함수를 분석하는 단계; 및 상기 샘플의 형광 휘도 및 상기 샘플에 포함된 세포외소포체 분자 개수를 이용하여 상기 세포외소포체 분자 1개 당 형광 휘도를 계산하는 단계를 포함하며,
상기 형광 염료가 표지된 세포외소포체 분자는 형광 염료가 화학적으로 표지된 것인, 생체 내 타겟 단백질을 검출하는 방법. - 타겟 단백질 또는 상기 타겟 단백질과 결합하는 단백질을 발현하는 세포주로부터 분리되고, 형광 염료가 화학적으로 표지된 세포외소포체 분자를 포함하고, 상기 형광 염료가 화학적으로 표지된 세포외소포체 분자는 청구항 1의 방법으로 표지된 형광 염료의 개수가 조절되고, 정량화된 것인, 생체 내 타겟 단백질을 검출하기 위한 키트.
