<3-7> 오미자 추출물 제조<3-7> Preparation of Omija extract

<3-8> 산수유 추출물 제조<3-8> Manufacture of Corn oil extract
산수유는 금산 관내에서 전남 구례에서 재배되는 산수유(열매)를 구입하여 사용하였다. 산수유 1 kg을 10배량의 정제수를 첨가하여 5분간 초음파를 한 후 교반기를 사용하여 80℃에서 6시간 3회 추출한 후, 감압 농축하여 4 brix 농도의 산수유 추출물을 제조하여 분말을 얻은 후 냉동 보관하여 사용하였다.Cornus japonica was purchased from Jeonnam Gurye in Jinsan, and was used for the purchase. 1 kg of corn oil was added with 10 times of purified water and ultrasonicated for 5 minutes. 상기 실시예 3을 통해 준비한 각각의 추출물의 과산화수소에 의해 유도된 산화적 스트레스로부터 SH-SY5Y(한국세포주은행) 신경세포 생존율을 측정하였다.The survival rate of SH-SY5Y (Korean Cell Line Bank) neurons was measured from the oxidative stress induced by hydrogen peroxide in each of the extracts prepared in Example 3 above.
자세하게는, SH-SY5Y 세포를 사용하여 10% FBS가 들어간 DMEM에서 37℃, 5% CO2조건에서 배양한 후 실험조건에 따라 세포수를 측정하여 96 웰 플레이트에 일정량을 넣어 준 후 과산화수소 대한 약물(시료 추출물)의 보호효과 측정을 위해서 과산화수소를 처리하기 4시간 전에 시료를 전처리 하였다. 세포생존도 측정은 과산화수소 처리 후 24시간 때에 실시 후 세포생존도 분석은 각각의 방법에 따라 수행하였다. 세포생존도 분석 방법으로는 WST-1 assay를 수행하였다.In detail, SH-SY5Y cells were cultured in DMEM containing 10% FBS at 37 ° C and 5% CO 2 , and then the number of cells was measured according to the experimental conditions, and a certain amount was added to a 96-well plate. 상기 팽화 홍삼 조건별 시료 및 약용작물 6종 중 과산화수소에 의해 유도된 산화적 스트레스로부터 SH-SY5Y 신경세포 생존율에서 팽화 가공 방법에 따른 조건선정은 팽화홍삼 2(RGP2)로 선정하였고, 약용작물은 구기자 및 산수유를 최종 선발하였다. 따라서 3종의 물 추출물을 혼합하여 과산화수소에 의해 유도된 산화적 스트레스로부터 신경세포 생존율을 통하여 단일 처리구보다 활성을 상승시키는 최적의 혼합비를 선정하였다.From the oxidative stress induced by hydrogen peroxide in six specimens and medicinal crops by the condition of paddy red ginseng, the condition for the expansion of SH-SY5Y neurons was selected according to the puffing processing method, and the selected paddy red ginseng 2 (RGP2) And marine oil were finally selected.
