Abstract
Description
도 2는 항 IL-6R 단클론 항체 제조를 위한 폴리 파아지 ELISA 분석 결과를 나타낸다.
도 3은 항 IL-6R 단클론 항체 제조를 위한 모노 파아지 ELISA 분석 결과를 나타낸다.
도 4는 모노 파아지 클론에 대한 핑거프린팅 결과를 나타낸다.
도 5는 IL-6R에 특이적으로 결합하는 6개 서로 다른 모노 파아지 클론을 단계적으로 희석해서 IL-6R에 대한 친화도가 어느 정도까지 보이는지 ELISA로 확인한 것으로, 희석 정도에 따라 결합력의 변화를 보였으나, 모두 높은 결합 친화도를 보였다.
도 6은 도 5에서의 결과 중 IL-6R full 단백질을 이용하여 파아지 클론을 선별 결과를 나타내며, 결과적으로 B10 의 결합력이 가장 높으며 D2, A7, A10, A3의 순서로 결합력을 보임을 알 수 있다.
도 7은 모노 파아지 클론들이 전체 IgG 형태로 전환되었음을 웨스턴 블랏을 통해 확인한 것이다.
도 8은 모노 파아지 클론 A3, A7, A10 의 IgG 항체를 정제한 결과를 나타낸다.

도 14의 (A) 는 모플라스미드에 대하여 제한효소로 절단(enzyme cut) 후 전기영동으로 확인한 결과이고, (B) 는 증폭 및 클로닝이 가능하도록 모플라스미드를 E.coli 숙주에 도입하여 새로운 세포주를 제작한 후 제한효소로 절단하여 전기영동으로 확인한 결과이고, (C) 는 아라비노스를 이용하여 미니서클 벡터를 제조하였음을 전기영동으로 확인한 결과를 나타낸다.
도 15는 미니서클 벡터가 GFP 를 발현하도록 구성한 후 293T 세포주에 도입하여 GFP 발현 여부를 확인한 결과를 나타낸다.
도 16은 미니서클 벡터가 GFP 를 발현하도록 구성한 후 쥐에 투여하여 in vivo 에서 GFP 발현 여부를 확인한 결과를 나타낸다.
도 17의 (A)는 모플라스미드에 항 IL-6R 항체(A7로 명명)를 발현하는 DNA 를 삽입한 후 그 경쇄(LC) 및 중쇄(HC)의 발현을 확인한 것이고, (B) 는 모플라스미드에 항 IL-6R 항체를 발현하는 DNA 를 삽입한 다음 아라비노스로 미니서클 벡터로 변환시킨 후 그 발현을 확인한 것이고, (C) 는 상기 미니서클 벡터가 항 IL-6R 항체를 발현한다는 것을 제한효소 절단을 통하여 확인한 결과를 나타낸다.
도 18은 미니서클 벡터가 항 IL-6R 항체(A7로 명명)를 발현한다는 것을 293T 세포주에서 GFP 발현을 통해 확인한 결과를 나타낸다. 각 그림에서 LD 는 저용량(low dose) 을, HD 는 고용량(high dose) 을 나타낸다.
도 19의 (A)는 미니서클 벡터가 항 IL-6R 항체(A7로 명명)를 발현한다는 것을 293T 세포주에서 GFP 발현을 통해 다시 한 번 확인한 결과를 나타낸다. (B)는 상기 세포의 배양 상층액 내에 존재하는 항-IL-6R 의 농도를 확인한 결과를 나타낸다.
도 20은 줄기세포에 GFP 를 발현하는 모플라스미드와 미니서클 벡터를 각각 도입하였을 때, 미니서클 벡터의 유전자 발현 효율이 더 우수함을 나타낸다.
도 21은 A7 항체가 류마티스 관절염 활막세포(fibroblast-like synoviocytes, FLS)의 증식을 억제하는 효과를 나타낸 것이다.
도 22는 A7 항체가 류마티스 관절염 활막세포(FLS)의 이동을 억제하는 효과를 나타낸 것이다.

도 24는 콜라겐 유발성 관절염(collagen-induced arthritis, CIA) 동물 모델에 A7 항체를 투여했을 때 각 동물에 대하여 발의 붓기 점수를 나타낸다. Wild type 은 정상 동물, CIA 는 콜라겐 유발성 관절염 동물을 의미한다.

Claims (15)
- (a) 프로모터;
IL-6R(Interleukin-6 receptor)에 특이적인 단클론 항체를 코딩하는 유전자; 및
전사 터미네이터를 포함하는 유전자 발현 카세트를 포함하고,
(b) 상기 유전자 발현 카세트의 외부에 위치한 부위 특이적 재조합 영역을 포함하며, 및
(c) 복제 개시점 및 선별마커 유전자는 포함하지 않는
비바이러스성 벡터가 도입된 줄기세포로서,
상기 IL-6R에 특이적인 단클론 항체는
서열번호 1 의 아미노산 서열로 이루어진 중쇄 CDR1, 서열번호 2 의 아미노산 서열로 이루어진 중쇄 CDR2 및 서열번호 3 의 아미노산 서열로 이루어진 중쇄 CDR3을 포함하는 중쇄 가변 영역; 및
서열번호 4 의 아미노산 서열로 이루어진 경쇄 CDR1, 서열번호 5 의 아미노산 서열로 이루어진 경쇄 CDR2 및 서열번호 6 의 아미노산 서열로 이루어진 경쇄 CDR3을 포함하는 경쇄 가변 영역을 포함하는 것인, 줄기세포.

- 제14항에 있어서, 상기 자가 면역 질환이 류마티스성 관절염, 인슐린 의존성 당뇨병, 다발성 경화증, 루푸스, 건선, 염증성 장질환, 궤양성 대장염, 중증 근무력증, 다발성 근육염, 피부근염, 크론병, 자가면역혈구감소증, 맥관염 증후군 또는 전신 홍반성 낭창인 조성물.
