Claims (34)
- 세포군 내의 60% 이상의 세포가 영장류 만능 줄기 (primate pluripotent stem, pPS) 세포주에서 분화된 것인 특징을 갖고 하기 목록으로부터 3 가지 이상의 추가적인 특징을 갖는, 시험관 내에서 배양될 수 있는 분화된 세포군:ㆍ항체로 검출가능한 α1-안티트립신 (AAT) 의 발현;ㆍ항체로 검출가능한 알부민의 발현;ㆍ항체로 검출가능한 α-페토단백질 (fetoprotein) 발현의 부재;ㆍRT-PCR 로 검출가능한 아시알로당단백질 수용체 (ASGR) 의 발현;ㆍ글리코겐 저장의 증거;ㆍ시토크롬 p450 활성의 증거;ㆍ글루코오스-6-포스파타아제 활성의 증거; 및ㆍ간세포의 형태학적 특색.
- 영장류 만능 줄기 (pPS) 세포주; 및 상기와 동일한 pPS 세포주에서 분화된 것을 특징으로 하고 하기 목록으로부터 3 가지 이상의 추가적인 특징을 갖는 분화된 세포군으로 이루어진 2 가지 단리된 세포군 세트:ㆍ항체로 검출가능한 α1-안티트립신 (AAT) 의 발현;ㆍ항체로 검출가능한 알부민의 발현;ㆍ항체로 검출가능한 α-페토단백질 (fetoprotein) 발현의 부재;ㆍRT-PCR 로 검출가능한 아시알로당단백질 수용체 (ASGR) 의 발현;ㆍ글리코겐 저장의 증거;ㆍ시토크롬 p450 활성의 증거;ㆍ글루코오스-6-포스파타아제 활성의 증거; 및ㆍ간세포의 형태학적 특색.
- 제 1 항 또는 제 2 항에 있어서, 상기 pPS 세포가 인간 배아 줄기 (hES) 세포인 세포군(들).
- 하기를 포함하는, 시험관 내에서 배양될 수 있는 분화된 세포를 수득하기 위한 영장류 만능 줄기 (pPS) 세포의 처리 방법:a) 인간 만능 줄기 세포의 배양물을 제공;b) 분화된 세포를 농축시키는 조건 하에서 간세포 분화 제제를 함유하는 배양 배지 중의 기질 상에서 상기 세포를 배양.
- 제 4 항에 있어서, 하기 특징 중 1 가지 이상을 가지는 방법:ㆍ세포를 배양하여 배상체를 형성하는 단계; 상기 배상체를 세포외 매트릭스 상에 플레이팅하는 단계; 간세포 분화 제제를 상기 배상체의 플레이팅과 동시에 또는 그 후에 상기 배지에 첨가하는 단계; 및 그 후 분화된 세포를 농축시키는 조건 하에서 상기 플레이팅된 세포를 간세포 분화 제제와 함께 배양하는 단계에 의하여 pPS 세포 배양물을 제조함;ㆍ상기 간세포 분화 제제를, 세포외 매트릭스는 함유하나 영양 세포는 본질적으로 없는 pPS 세포의 배양물에 첨가하고; 그 후 분화된 세포를 농축시키는 조건 하에서 상기 pPS 세포를 상기 간세포 분화 제제와 함께 배양함;ㆍ상기 간세포 분화 제제는 n-부티레이트와 같은 히스톤 디아세틸라아제 억제제임;ㆍ상기 세포를, 상기 간세포 분화 제제와의 배양과 동시적으로 또는 순차적으로 1 가지 이상의 간세포 성숙 인자와 함께 배양하는 것으로서, 이때 상기 간세포 성숙 인자 중 하나 이상이 디메틸 술폭시드 (DMSO), 디메틸아세트아미드 (DMA); 헥사메틸렌 비스아세트아미드 또는 기타 폴리메틸렌 비스아세트아미드와 같은 유기 용매, 또는, 글루코코르티코이드, 표피 성장 인자 (EGF), 인슐린, TGF-α, TGF-β, 섬유아세포 성장 인자 (FGF), 헤파린, 간세포 성장 인자 (HGF), IL-1, IL-6, IGF-I, IGF-II 또는 HBGF-1 과 같은 시토카인 또는 호르몬 중 하나임.
- 삭제
- 제 4 항 또는 제 5 항에 있어서, 상기 pPS 세포가 인간 배아 줄기 (hES) 세포인 방법.
- 제 1 항 또는 제 2 항에 따른, 또는 제 4 항 또는 제 5 항의 방법에 따라 제조된 분화된 세포를 화합물과 배합하고 상기 화합물이 세포에 독성인지 여부를 결정하는 것을 포함하는, 화합물의 간세포 독성 스크리닝 방법.
- 제 1 항 또는 제 2 항에 따른, 또는 제 4 항 또는 제 5 항의 방법에 따라 제조된 분화된 세포를 화합물과 배합하고, 상기 화합물과의 접촉에 따른 세포 내 임의의 표현형 또는 대사적 변화를 측정하여 상기 변화를 간세포 기능의 조절능과 연관시키는 것을 포함하는, 화합물의 간세포 독성 스크리닝 방법.
- 세포가 플루이드 중의 독소를 제거하거나 변형시킬 수 있게 하는 조건 하에서 제 1 항 또는 제 2 항에 따른, 또는 제 4 항 또는 제 5 항의 방법에 따라 제조된 분화된 세포를 상기 플루이드와 접촉시키는 것을 포함하는, 생체 밖에서의 플루이드의 해독 방법.
- 제 1 항 또는 제 2 항에 따른, 또는 제 4 항 또는 제 5 항의 방법에 따라 제조된 분화된 세포를 포함하는, 개인에서의 간세포 기능의 복원 또는 보충용 약학 제제.
- 제 1 항 또는 제 2 항에 따른, 또는 제 4 항 또는 제 5 항의 방법에 따라 제조된 분화된 세포가, 환자로부터 유입되고 장치에 의해 해독된 후 상기 환자에게 복귀되는 혈액을 해독시키도록 위치되는 간 보조 장치.
- 제 5 항에 있어서, 세포외 매트릭스가 라미닌, 콜라겐 또는 엥겔브레드-홀름-스왐 (Engelbreth-Holm-Swarm) 세포의 매트릭스인 방법.
