Claims (7)
- (ⅰ) 미지의 염기서열을 갖는 원핵 또는 진핵세포의 게놈 DNA의 샷건 라이브러리(shotgun library)를 지지체상에 고정화하여 DNA 칩(chip)을 제조하고;(ⅱ) 프로브(probe)로서, 말단이 각각 다른 형광물질로 직접 표지(label)된, 원핵 또는 진핵세포로부터 분리된 2 종 이상의 mRNA를 한 개의 DNA 칩상의 게놈 DNA와 동시에 혼성화(hybridization)시키고;(ⅲ) DNA 칩을 세척하여 혼성화되지 않은 mRNA를 제거하고;(ⅳ) DNA 칩상의 형광을 판독하는 단계를 포함하는,유전자의 존재 또는 발현양상을 확인하는 방법.
- 삭제
- 제1항에 있어서, 형광물질로 표지된 mRNA가 바이오티닐화(biotinylated) mRNA를 스트렙타비딘(streptavidin)-결합 형광물질로 표지하여 제조된 것인 방법.

- 제1항에 있어서, 단계 (ⅱ)에서 원핵세포로부터 분리된 mRNA가(ⅰ) 원핵세포로부터 총 RNA를 추출하고;(ⅱ) 추출된 총 RNA를 rRNA에 상보적인 프라이머로 역전사하여 rRNA-rDNA 하이브리드를 얻고;(ⅲ) 얻어진 rRNA-rDNA 하이브리드를 RNaseH 및 DNaseⅠ으로 처리하여 제거하는:단계를 포함하는 방법에 의해 제조된 것인 방법.

- (ⅰ) 지지체상에 미지의 염기 서열을 갖는 원핵 또는 진핵세포의 게놈 DNA의 샷건 라이브러리가 고정화된 DNA 칩;(ⅱ) 프로브로서, 말단이 각각 다른 형광물질로 직접 표지된, 원핵 또는 진핵세포로부터 분리된 2 종 이상의 mRNA; 및,(ⅲ) DNA 칩상의 게놈 DNA와 mRNA간의 혼성화를 가능하게 하는 완충용액을 포함하는,제1항에 따른 유전자의 존재 또는 발현양상을 확인하는 방법을 수행하기 위한 키트.
