Abstract

PURPOSE:To carry out continuous culture of cell at high density, by circulating a culture liquid supplied to the cavity of a hollow fiber under pressure and circulating a culture liquid containing a low-molecular substance and a high- molecular valuable substance formed in a cell culture space through a membrane separation apparatus. CONSTITUTION:A culture liquid 5 is introduced into a hollow fiber 1 with a liquid pump 6, pressurized with pressure supplied from a pneumatic pressure applying apparatus 8 and circulated. A part of the circulating liquid is permeated into a cell culture space 4 to produce a low-molecular substance and a high- molecular valuable substance, which are temporarily stored in a chamber 19. The produced liquid 5a is introduced into a membrane separation apparatus 15 with a liquid pump 14 to separate low-molecular metabolic product such as lactic acid and ammonium ion with a separation membrane 16 and the treated liquid is circulated and concentrated. The high-molecular valuable substance is recovered at a definite flow rate with a liquid pump 18. The environment in the cell culture apparatus is maintained as uniformly as possible to enable the continuous culture of cell at high density and the production of valuable substance at high concentration over a long period.
