Abstract
PURPOSE:To perform immunoassay with high accuracy by incorporating a chemical material which can be a trigger for a chain of chemical reactions into liposome and constituting an immunoassay reagent from this liposome, a chemical reaction reagent and complement. CONSTITUTION:The chemical material (beta-1, 3 glucan, etc.) which can be a trigger for a chain of chemical reactions is incorporated into the liposome consisting of lipid bilayers and the antibody or the antigen is immobilized to the surface. The immunoassay reagent is prepd. by incorporating this liposime, the chemical reaction reagent (extracted matter of horseshoe crab blood cells, etc.) and the complement therein. For example, the beta-1, 3 glucan (3) is included in the liposome (1) and the human IgG(2) which is the antibody is immobilized therein. This reagent, antihuman IgG which is the object to be measured and the extracted matter of horseshoe crab blood cells are mixed. The viscosity of the resultant reaction mixture is measured and the beta-1, 3 glucans released from the liposome are calculated. The concn. of the antihuman IgG is measured from the calculated value. Since this reagent is used, the immunoassay with high accuracy is enabled.
