Abstract
PURPOSE: To prepare the titled compound useful as an intermediate of pharmaceuticals such as antiphlogistic, analgesic and antipyretic agents, economically, by using a raw material which is easily available in an industrial scale, via a novel compound, 1-(2'.6'-dichlorophenyl)2,3-bis(methylthio)indole.
CONSTITUTION: The novel compound of formula II is prepared by the heat treatment or acid treatment of the compound of formulaI. The heat treatment is preferably carried out at ≥200°C in the absence of solvent, and the acid used in the acid treatment is preferably a mineral acid, a Lewis acid, or an acid halide. The compound of formula II can be converted to the objective compound of formula III by treating with a mineral acid in an aqueous medium preferably in the presence of an organic carboxylic acid compatible with water, e.g. acetic acid, etc. Hydrolysis of the objective compound yields the compound of formula IV whose Na salt is useful as the above-mentioned pharmaceutical. Since the compound of formulaIcan be prepared from a raw material which can be obtained easily in an industrial scale, the above pharmaceutical can be prepared by this process economically, in an industrial scale, using simple reaction procedures and reduced number of steps througout the whole process from an industrially available raw material.
