Claims (1)
- 【特許請求の範囲】 1.SRC-1ポリペプチドをコードする、単離、濃縮又は精製した核酸。 2.核酸がヒト核酸である請求の範囲第1項の核酸。 3.核酸が図1に示したアミノ酸配列の少なくとも25の連続するアミノ酸を コードする請求の範囲第1項の核酸。 4.核酸が図1に示したアミノ酸配列の少なくとも50の連続するアミノ酸を コードする請求の範囲第1項の核酸。 5.核酸が図1に示したアミノ酸配列の少なくとも100の連続するアミノ酸 をコードする請求の範囲第1項の核酸。 6.核酸が図1に示したアミノ酸配列の少なくとも425の連続するアミノ酸 をコードする請求の範囲第1項の核酸。 7.試料中のSRC-1ポリペプチドの欠失を探査する核酸プローブ。 8.SRC-1ポリペプチドが図1に示したアミノ酸配列の少なくとも25の連続 するアミノ酸を含む請求の範囲第7項のプローブ。 9.SRC-1ポリペプチドが図1に示したアミノ酸配列の少なくとも50の連続 するアミノ酸を含む請求の範囲第7項のプローブ。 10.SRC-1ポリペプチドが図1に示したアミノ酸配列の少なくとも100の連 続するアミノ酸を含む請求の範囲第7項のプローブ。 11.SRC-1ポリペプチドが図1に示したアミノ酸配列の少なくとも425の連 続するアミノ酸を含む請求の範囲第7項のプローブ。 12.SRC-1ポリペプチドをコードする組換え核酸と、宿主細胞中で転写を開始 する効果のあるベクター又はプロモーターとを含む物質の組成物。 13.組換え核酸が図1に示したアミノ酸配列の少なくとも25の連続するアミ ノ酸をコードする請求の範囲第12項の組成物。 14.組換え核酸が図1に示したアミノ酸配列の少なくとも50の連続するアミ ノ酸をコードする請求の範囲第12項の組成物。 15.組換え核酸が図1に示したアミノ酸配列の少なくとも100の連続するア ミノ酸をコードする請求の範囲第12項の組成物。 16.組換え核酸が図1に示したアミノ酸配列の少なくとも425の連続するア ミノ酸をコードする請求の範囲第12項の組成物。 17.SRC-1ポリペプチドをコードする導入遺伝子を含むヒト以外のトランスジ ェニック哺乳動物。 18.導入遺伝子が図1に示したアミノ酸配列の少なくとも25の連続するアミ ノ酸をコードする請求の範囲第17項のトランスジェニック哺乳動物。 19.導入遺伝子が図1に示したアミノ酸配列の少なくとも50の連続するアミ ノ酸をコードする請求の範囲第17項のトランスジェニック哺乳動物。 20.導入遺伝子が図1に示したアミノ酸配列の少なくとも100の連続するア ミノ酸をコードする請求の範囲第17項のトランスジェニック哺乳動物。 21.導入遺伝子が図1に示したアミノ酸配列の少なくとも425の連続するア ミノ酸をコードする請求の範囲第17項のトランスジェニック哺乳動物。 22.標的遺伝子の転写を増強する方法であって、標的遺伝子を含む細胞にSRC- 1ポリペプチドをコードする核酸を提供する段階を含む方法。 23.遺伝子治療における核酸カセットの発現を調節する分子スイッチを標的遺 伝子を含む細胞に提供する段階であって 分子スイッチが修飾されたリガンド結合ドメインと連結したDNA結合ドメイ ンを含む段階 をさらに含む請求の範囲第22項の方法。 24.スイッチが組織特異的である請求の範囲第23項の方法。 25.分子スイッチを核酸カセットと結合させて、遺伝子治療で使用する核酸カ セット/分子スイッチ複合体を形成する段階をさらに含む請求の範囲第23項の方 法。 26.薬理学的用量の核酸カセット/分子スイッチ複合体を治療対象の動物又は ヒトに投与する段階をさらに含む請求の範囲第25項の方法。 27.修飾したリガンド結合部位に結合する薬理学的用量のリガンドを動物又は ヒトに投与することにより分子スイッチをオン、オフする請求の範囲第26項の方 法。 28.薬理学的用量のリガンドを動物又はヒトに投与した後に核酸配列が転写さ れてタンパク質を産生する請求の範囲第27項の方法。 29.分子スイッチ及び核酸カセットが別々のプラスミド上にあり、標的細胞に 同時注入される請求の範囲第28項の方法。 30.分子スイッチ及びSRC-1ポリペプチドをコードする核酸が別々のプラスミ ド上にあり、標的細胞に同時注入される請求の範囲第29項の方法。 28.核酸カセットと連結した分子スイッチを含む物質の組成物であって、カセ ット/分子スイッチ複合体がカセット中の核酸が標的細胞中で転写でき、かつ必 要に応じ翻訳できるようにベクター中で位置的、経時的に配向され、分子スイッ チがSRC-1ポリペプチドのコード領域をもつプラスミド中の活性プロモーターを 構成的に調節する組成物。 29.標的遺伝子の転写を減弱する方法であって、標的遺伝子を含む細胞にSRC- 1ポリペプチドの最も有力な負の抑制因子を提供する段階を含む方法。 30.遺伝子治療において核酸カセットの発現を調節するための分子スイッチで あって、 修飾したSRC-1ポリペプチドを含み、ポリペプチドが修飾結合ドメインと連結 した天然SRC-1DNA結合ドメイン を含む分子スイッチ。 31.遺伝子治療において核酸カセットの発現を調節するための方法であって、 請求の範囲第30項の分子スイッチを結合させて遺伝子治療で使用する核酸/分子 スイッチ複合体を形成する段階と、薬理学的用量の核酸/分子スイッチ複合体を 治療対象の動物又はヒトに投与する段階とを含む方法。 32.核酸カセットと連結した分子スイッチを含む物質の組成であって、複合体 がカセット中の核酸が標的細胞中で転写でき、かつ必要に応じ翻訳できるように ベクター中で位置的に配向されている組成。 33.SRC-1をコードする配列を含む発現ベクターを細胞に挿入する段階と、こ の細胞を試験管内で培養する段階と、この細胞をかような治療が必要な患者に注 入する段階とを含む、SRC-1関連疾患又は健康状態を治療する方法。
