Abstract
Description
Claims (25)
- 1.非開裂HIVenvおよび医薬的に許容される賦形剤からなる滅菌組成物の 治療的有効投与量を個体に投与することからなるAIDSの予防または治療方法 。
- 2.該非開裂HIVenvが全長gp120またはその断片からなる第1項の方 法。
- 3.該非開裂HIVenvが全長gp160またはその断片からなる第1項の方 法。
- 4.HIVに対する暴露または暴露の危険後に該組成物を個体に投与する第1項 の方法。
- 5.一系列の接種の1つとして該組成物を投与する第1項の方法。
- 6.該系列が第1項の組成物とは異なる少なくとも1つのHIV抗原による接種 を含む第5項の方法。
- 7.HIVに対するワクチンの製造における非開裂HIVenvの使用。
- 8.医薬的に許容される担体中の非開裂HIVenvからなるHIVワクチン。
- 9.該非開裂HIVenv内の開裂部位以外のアミノ酸残基が置換されているか 、欠失しているか、あるいは別のアミノ酸残基の隣接する挿入を有する第8項の ワクチン。
- 10.1および30N−末端アミノ酸残基の間からなる断片が欠失している第9 項のワクチン。
- 11.開裂HIVenv断片を本質的に含有しない非開裂HIVenv調製物。
- 12.少なくとも50%が開裂HIVenv断片を含有しない第11項の調製物 。
- 13.非開裂gp120を含有する組成物。
- 14.非開裂gp160を含有する組成物。
- 15.次の操作: a.HIVenvの第1の調製物を、第2の(抗体結合非開裂HIVenv)調 製物の形成を可能とするに足る時間、開裂部位をまたくHIVenvエピトープ を指向する抗体と接触させる; b.上記第2の調製物を抗体の結合していないあらゆるHIVenvから分離す る; c.該第2の調製物から非開裂HIVenvを回収する;からなる非開裂HIV envの製造方法。
- 16.開裂部位をまたぐHIVenvエピトープを指向する抗体を担体マトリッ クス上に結合し、HIVenvおよび非開裂HIVenvを含有する溶液をその カラムに通し、非開裂HIVenvを上記マトリックス結合抗体に選択的に吸着 させ、吸着した抗体−非開裂HIVenvマトリックスを洗浄することによって 、非吸着物資を除去し、非開裂HIVenvを溶出させるアフィニティークロマ トグラフィーからなる非開裂HIVenvの単離方法。
- 17.非開裟HIVenvをコード化するDNAで形質転換された非開裂HIV envの複製および発現に適した哺乳類細胞を低血清培地中で生育させ、非開裂 HIVenvを回収することからなる非開裂HIVenvの製造方法。
- 18.該培地が約0〜3%血清を含有する第17項の方法。
- 19.6E10、13H8、11G5、10F6、10D8および6D8からな る群から選択されるモノクローナル抗体の特徴を有する抗体。
- 20.モノクローナル抗体5B3、5B6、5C2、6D8、6E10、7F1 1、7G11、9E3、10C1、10D8、10F6、11G5、13H8、 14F12、および15G7からなる群から選択される、検出可能なマーカーま たは水不溶性マトリックスに共有結合したモノクローナル抗体。
- 21.モノクローナル抗体5B3、5B6、5C2、6D8、6E10、7F1 1、7G11、9E3、10C1、10D8、10F6、11G5、13H8、 14F12、および15G7からなる群から選択される、滅菌された医薬的に許 容される賦形剤中のモノクローナル抗体から構成される、患者への投与に適した 組成物。
- 22.該モノクローナル抗体が毒素に結合している第21項の組成物。
- 23.HIV感染を有するか、もしくはHIV感染を有する危険にある患者に第 21項の組成物の治療的に許容される投与量を投与することからなる方法。
- 24.該組成物を一組の少なくとも3回の連続的接種において投与し、第1回の 接種後、2〜8週間の期間内に第2回の接種を行い、該第2回の接種後、該第1 回の接種後5ヵ月〜2年の期間内に第3回の接種を行う第1項の方法。
- 25.該組成物中の非開裂HIVenvの投与量が約10μg〜1mgである第 1項の方法。
