Description
本発明の組成物
ウシエナメル質からの染色ペリクル除去の尺度としてのペリクル清浄率試験(PCR)
導入
以前の試験(J. Dent. Res., 61:1236, 1982)は、歯磨き剤スラリーによるin-vitro PCR試験の結果が、妥当な信頼度をもって、臨床所見を予測し得ると考えられる可能性があることを示している。
ウシの永久中中切歯を切断して約8x8mm2の唇側エナメル質標本を得た。エナメル質標本を次いで、エナメル質表面のみが曝露されるように、自己重合性メタクリレート樹脂中に、包埋した。次いでエナメル質表面を、宝石加工ホイール上で滑らかにし、軽石粉及び水で磨き、次いで超音波処理して余分な破片を除去した。次に標本を、軽くエッチング(0.12MのHCl中で60秒、飽和NaCO3中で30秒、且つ1.0%フィチン酸中で60秒)して、ステインの蓄積及び付着を促進した。次いで、タンパク源としての胃ムチン並びに染色源としてのコーヒー、茶及びFeCl36H2O(即ち、400ml中に1.35gのコーヒー、1.35gの茶、0.02gのFe及び1.0gのムチン)を含有する染色ブロスに交互に標本を曝露する回転ロッド上に、標本を、最低10日間配置して、標本が十分なステインを確実に発現するようにした。
結果を図1にグラフとして示す。
ウシエナメル質試料に対する磨耗性の尺度としての相対的象牙質磨耗性試験(RDA)
導入
前述の歯磨き剤製剤1及び2を、それらの磨耗性(RDAによって決定)に関して評価し、代替シリカ研磨性材料を含有する類似の比較製剤(本発明の範囲には包含されない)と比較した。
使用した手順は、ISO/ADA推奨の歯磨き剤摩損性の決定手順であった。象牙質標本(8)を、ISO/ADAによって概説された制御条件下で、中性子束の中に配置した。次いで標本を、V-8クロスブラッシング機に取り付けられるよう、メチルメタクリレート中にマウントした。0.5%のCMCグリセリン溶液50ml中10gのISO/ADA標準物質からなるスラリーを用いて、標本を、予調整作動において1500ストローク、ブラッシングした(歯が以前に使用されていたため)。使用したブラシは、ISO/ADAによって規定されたもので、以前の試験において使用された。ブラシ張力は150gであった。予調整作動に続き、試験を、150g及び1500ストロークを用いて、以下の表3に概説するように、各試験材料スラリー(歯磨き剤25g/水40ml)の両側に標準物質スラリー(ADA標準物質10g/0.5%CMC50ml)を配置する、サンドイッチデザインで行った。
RDAの結果を図2にグラフとして示す。更に、各群について、平均値、標準偏差(SD)及びSEMを算出し、同様に図2に示す。

シリカ試料に対し走査型電子顕微鏡法を行って、粒子のサイズ及び形態を決定した。溶融シリカ及び沈降シリカを、比較の目的で含めた。
アルミニウムスタブ上のカーボンパッドにマウントした、金でコーティングした試料のSEMを、10eVで作動しているJeol 6480 LV SEMで行った。
シリカ試料のSEM画像を図3に示す。
Zeodent及びTeco-Sil試料は、マイクロ次元の不規則集合体からなっていた。Zeodent試料は、ナノスケールテクスチャを有するように思われ、Teco-Sil材料は、より滑らかで、またプレート様形態のいくつかの粒子をも呈した。Sunsphere NP-30及びSpheron材料は本質的に、滑らかな球状粒子からなっていた。Spheron P-1500は、他の球状粒子とは異なり、表面のナノメートルサイズの突起により表面が荒くなっていた。
気流を1000℃まで加熱することにより、粉末試料で、TGA-DSCを行って、温度上昇及び相変化(ある場合には)に伴う重量減少を決定した。
分析は、20~30mgの試料に対して、TA-Instruments SDT Q600 simultaneous TGA/DSCを用いて、周囲温度から1000℃までの温度範囲における気流下で、昇温速度5℃/分で行った。
結果を、以下の表5で実証し、表5は、室温(RT)から120℃までの範囲(主として結合水)及び120℃から1000℃までの範囲(主として結合ヒドロキシル)にわたって、TGAにより測定した、シリカ試料の重量パーセント減少を示す。
表面積及び多孔度の分析
シリカ試料の表面積を、BET N2吸着により測定し、等温プロットもまた測定して、細孔の径及び容積を決定した。
ガス収着(N2、77K)からの算出Brunauer Emmett Teller(BET)比表面積を、Micromeritics 3-Flexガス収着分析器で測定した。全ての粉末試料(400~700mg)を、オーブン中で、真空(10-3mbar)下、200℃で8時間、予脱気し、次に分析を行う前に、3-Flex analyser中で、超高真空(10-6mbar)下、200℃で16時間、脱気した。
表面積
表面積の測定値を表6に示す。Zeodent 103試料及びSpheron P-1500は、中程度の表面積80~90m2/gを、Zeodent 103は45m2/gの表面積を有し、残りの試料は低い表面積を有していた。
77KでのN2吸脱着プロファイルの等温プロット及び細孔径分布を測定し、最新の非局所密度汎関数理論(NLDFT)によって算出した。1測定した表面積及び等温測定値から算出した多孔度を表6に示す。

Zeodent 及びSpheron P-1500の試料は、中程度に高い表面積及びメソ多孔度を有することが見出され、残りの試料は、本質的に表面積が少なく非孔質であった。
比表面積及び細孔容積
装置
BELSORP-mini(MicrotracBEL、日本)
比表面積、細孔容積、及び粒径を、窒素吸着法を用いて、決定した。分析に先立ち、不活性ガスを連続的に流すのと連動して昇温することによって達成される脱気プロセスにおいて、シリカ試料を予調整し、物理的に結合している不純物を除去した。シリカ表面に吸着されたガスの体積を窒素の沸点(-196℃)で測定した。吸着されたガスの量をシリカ粒子の比表面積と相関させた。計算は、BET(Brunauer, Emmett and Teller)理論に基づいた。窒素吸着法はまた、細孔容積及び細孔径の決定をも可能にする。測定は、以下のプロトコールを用いて行った:

操作手順
1)0.2gの試料をサンプルチューブに充填し、圧を測定条件範囲まで低下させる。
2)サンプルチューブの弁を閉じ、秤量する(A)。
3)Aとサンプルチューブ単独(ブランク)との重量差を概算する。
4)多孔度の測定を開始する。
5)比表面積を、次の2つの式を用いて算出する。

Claims (12)
- 非溶融で球状で無水の非晶質シリカゲル粒子を含む歯磨き剤組成物であって、シリカゲル粒子が、
a. 0.03ml/gから0.1ml/g未満の細孔容積;
b. 1μから10μまでの平均粒径;
c. 50m2/g以下のBET表面積;
d. 20 mL/100gから50mL/100gの油吸収容量;及び
e. 0.2重量%未満の水含有量
を含む、上記歯磨き剤組成物。 - シリカゲル粒子が、1800MPaから2000MPaまでの範囲の圧縮強度を含む、請求項1に記載の組成物。

- 減感剤を含む、請求項1から6のいずれか一項に記載の組成物。
- フッ化物イオン源を含む、請求項1から7のいずれか一項に記載の組成物。
- 請求項1から8のいずれか一項に記載の歯磨き剤組成物であって、シリカゲル粒子が、
a. 0.05ml/gの細孔容積;
b. 4μの平均粒径;
c. 40m2/gのBET表面積;及び
d. 30mL/100gの油吸収容量;
を含む、上記歯磨き剤組成物。 - 請求項1から9のいずれか一項に記載の歯磨き剤組成物であって、シリカゲル粒子が、組成物の0.1重量%から5重量%までの量で存在する、上記歯磨き剤組成物。
- 歯又は義歯の表面の清浄化、歯又は義歯の表面の研磨、歯又は義歯の表面からのステインの除去に用いるための、請求項1から10のいずれか一項に記載の歯磨き剤組成物。
- 非溶融で球状で無水の非晶質シリカゲル粒子を含む歯磨き剤組成物を製造する方法であって、
沈降及びそれに続いて、水を除去し、粒状シリカ材料を産生するためのか焼の工程を含み、
シリカゲル粒子が、
a. 0.03ml/gから0.1ml/g未満の細孔容積;
b. 1μから10μまでの平均粒径;
c. 50m2/g以下のBET表面積;
d. 20 mL/100gから50mL/100gの油吸収容量;及び
e. 0.2重量%未満の水含有量
を含む、上記方法。
