各搾出液10mLに、供試芽胞菌液0.1mLを添加し、25℃にて15分間接触させた後、滅菌中和溶液を加えよく攪拌した。
芽胞菌としてBacillus subtilis NBRC3134(108CFU/mLレベル)及びClostridium difficile ATCC9689(108CFU/mLレベル)を用いた。
この1mLをSCD寒天培地で混釈培養し生菌数を確認し、下記の基準で評価した。
評価基準:
1点:供試菌のlog reductionが3以上の菌数減少
2点:供試菌のlog reductionが2以上、3未満の菌数減少
3点:供試菌のlog reductionが2未満の菌数減少
として上記各菌種について菌数減少を点数で評価し、菌数減少の点数の平均値を求め、以下の基準で除菌性を評価した。
◎:平均値が1.0点以上、1.5点未満。
○:平均値が1.5点以上、2.0点未満。
△:平均値が2.0点以上、2.5点未満。
×:平均値が2.5点以上。
とし、△、○、◎を実用性のあるものとして判定した。

試験方法:
試験管に0.9mLの搾出液と、108TCID50/mLとなるように調整したウイルス液0.1mLを加え、ミキサーで混合し、25℃で10分間作用させた。作用後0.1mLをサンプリングし、9.9mLの作用停止液(2%チオ硫酸ナトリウム、0.2%レシチン及び1.5%ポリソルベート80を含む細胞増殖液)に添加して100倍希釈後、ウイルスに対する水性除菌剤(搾出液)の作用を停止させた。これをウイルス感染価測定用資料原液として感染価の測定に用いた。同様に、搾出液の代わりに、リン酸緩衝生理食塩水(phosphate bufferred saline、以下、PBSと略すことがある)を用いた試験を実施し、コントロールとした。リン酸緩衝生理食塩水は、塩化ナトリウム8g、塩化カリウム0.2g、リン酸水素二ナトリウム1.44g、リン酸二水素カリウム0.24gの4種類の塩を混合し、超純水で900mLに溶解後、塩酸でpHを7.4に調整する。1Lにメスアップ後、オートクレーブにより滅菌する。ウイルスとして、ネコカリシウイルス、マウスノロウイルス、B型肝炎ウイルス、C型肝炎ウイルス、アデノウイルス、AIDSウイルス、インフルエンザウイルスを用いた。
ウイルス感染価測定用試料原液をPBSで10倍段階希釈した後、感染価測定用試料原液及び希釈ウイルス液50μLとウシ胎仔血清(FBS)を5%添加したDulbecco‘s modified Eagle’s Medium(DMEM)に懸濁した細胞を96ウェルプレートに植え込んだ。その後、炭酸ガスふ卵器において37℃、5%CO2の条件で4日間培養した。

細菌については、抗菌持続性の5種の細菌に対する平均値を求め、以下の基準で抗菌持続性効果を評価した。

○:泡立ち・べたつき・水っぽさ・ごわごわ感がなく、適度な湿り気があり使用感が良い。
△:やや泡立ち・べたつき・水っぽさ・ごわごわ感がある、もしくは適度な湿り気がない。
×:泡立ち・べたつき・水っぽさ・ごわごわ感が非常に強い、もしくはほとんど湿り気がなく使用感が悪い。
