Claims (28)
- 染色体のテロメア反復配列及びサブテロメア配列を増幅する方法であって、前記方法は:
a)以下によって核酸伸長産物を作製すること:
i)二本鎖染色体DNAの3’オーバーハングにおけるテロメア反復配列に伸長プライマーをハイブリダイズすることであって:
(1)二本鎖染色体DNAは、テロメア反復配列を含むテロメア領域及びサブテロメア配列を含むサブテロメア領域を有し;及び
(2)伸長プライマーは:
(A)アニーリング条件下で3’オーバーハングにおけるテロメア反復配列にハイブリダイズする3’部分及び
(B)アニーリング条件下で3’オーバーハングにおけるテロメア反復配列にハイブリダイズしないアンカー配列を有する5’部分
を含む、前記ハイブリダイズすること;及び
ii)時間制御された鎖−置換伸長反応を行って二本鎖染色体DNAのサブテロメア領域の方へ伸長プライマーを伸張することであって、伸長反応は、予め定められた長さの範囲内でテロメア領域を有する二本鎖染色体DNAのみからのテロメア反復配列及びサブテロメア配列の両方を含む伸長産物を産生するように調節される、前記伸長すること;及び
b)その後のPCR反応によって伸長産物の配列を増幅すること、
を含み;これによりテロメア反復配列及びサブテロメア配列を有する核酸を含む長さの限られた増幅産物を産生し、
時間制御された伸長反応は、予め定められた長さの伸長産物を産生するように調節され、
前記時間制御された伸長反応は、0.1〜30分、0.1〜10分、0.1〜5分、0.1〜4分、0.1〜3分、0.1〜2分、0.1〜1分、または0.1〜0.5分であるように調節される、前記方法。 - 配列は:
(1)アニーリング条件下で伸長産物におけるサブテロメア領域に独特の配列にハイブリダイズする第1の増幅プライマー;及び
(2)アニーリング条件下でアンカー配列にハイブリダイズする第2の増幅プライマー、
を使用して増幅される、請求項1に記載の方法。 - 第1の増幅プライマーは、配列5’−GATGGATCCTGAGGGTGAGGGTGAGGG−3’[配列番号:2]、5’−CGGGCCGGCTGAGGGTACCGCGA−3’[配列番号:10](染色体1)、5’−GCTAATGCACTCCCTCAATAC−3’[配列番号:11](染色体5)または5’−CATTCCTAATGCACACATGATACC−3’[配列番号:12](染色体9)を含む、請求項2に記載の方法。
- 第1の増幅プライマーは、配列5’−GATGGATCCTGAGGGTGAGGGTGAGGG−3’[配列番号:2]を含み、及び第2のプライマーは、配列5’−TGCTCGGCCGATCTGGCATC−3’[配列番号:8]を含む、請求項2に記載の方法。
- 増幅されたテロメア産物の長さ範囲は、PCR反応において時間制御された伸長時間を使用して決定することができる、請求項1に記載の方法。
- 対照配列を同時増幅することをさらに含む、請求項1に記載の方法。
- 対照配列は、複数の非テロメア反復配列を含む、請求項6に記載の方法。
- 短いテロメアの存在量を決定するための方法であって:
a)被験体から3’オーバーハングを含む二本鎖染色体DNAを含む試料を得ること;
b)請求項1の方法を使用して二本鎖染色体DNAから長さの限られた増幅産物を産生すること;及び
c)長さの限られた増幅された産物から短いテロメアの存在量を決定すること、
を含む前記方法。 - d)試料からの総テロメアの存在量の測定と短いテロメアの存在量を比較することをさらに含む、請求項8に記載の方法。
- 工程d)は、総テロメアの存在量の関数として短いテロメアの存在量を決定することを含む、請求項9に記載の方法。
- 短いテロメアの存在量を決定することは、qPCRを使用して行われる、請求項8に記載の方法。
- qPCRは、第1及び第2のプライマーを使用して行われ、
i)前記第1のプライマーは、第1の鎖の少なくとも1つの反復単位にハイブリダイズし、及び前記第2のプライマーは、第2の鎖の少なくとも1つの反復単位にハイブリダイズし、
ii)前記ハイブリダイズしたプライマーは、これらのそれぞれの鎖にハイブリダイズしたときに、プライマー伸長ができる、及び前記第1のプライマーの少なくとも1つのヌクレオチドは、前記第1のプライマーが前記第1の鎖の少なくとも1つの反復単位にハイブリダイズしたときに、前記第1のプライマーと前記反復単位のヌクレオチドとの間に内部塩基対ミスマッチを生じ、
iii)前記第1のプライマーは、また第1及び第2のプライマーが互いにハイブリダイズするときに、前記第2のプライマーの3’末端ヌクレオチドとミスマッチを生じ、
iv)前記第2のプライマーの少なくとも1つのヌクレオチドは、前記第2のプライマーが前記第2の鎖の少なくとも1つの反復単位にハイブリダイズしたときに、前記第2のプライマーと前記反復単位のヌクレオチドとの間の内部塩基対ミスマッチを生じる、請求項11に記載の方法。 - 短いテロメアの存在量を決定することは、サザンブロット、ドットブロット法、スロットブロット、免疫化学、核酸シーケンシング、またはデジタルPCRによって試料における平均テロメア長を測定することを含む、請求項8に記載の方法。
- 短いテロメアの存在量は、存在量比の測定である、請求項8に記載の方法。
- 総テロメアの存在量は、ゲノム参照配列の存在量に相対的に測定される、請求項9に記載の方法。
- ゲノム参照配列は、単一コピー参照核酸配列または非テロメア反復DNA配列を含む、請求項15に記載の方法。
