「ハロゲン」とは、フッ素、塩素、臭素またはヨウ素、好ましくはフッ素または塩素を意味する。
AAD(例えば、式(I))は、有機合成法の一般的な知識を使用して合成することができる化合物の1種である。例えば、AADは、フェノールをクロロアセトンで誘導体化し、ストレッカー反応を行い、得られたアミンをアロイルクロリドでアシル化することによって合成することができる(スキーム1に示した通り)。必要に応じて、その後、特定のエナンチオマーを、例えばキラル分割によって得ることができる(スキーム2に示した通り)。
・mTORシグナル伝達は、アルツハイマー病(AD)の病理といくつかの面で共通点があることから、疾患の進行の誘因として役割を有する可能性が示唆される。全般的に、所見は、ADの脳ではmTORシグナル伝達が機能亢進していることを示している。例えば、ヒトADの脳の死後研究によって、PTEN、Akt、S6KおよびmTORの調節異常が明らかである。
・ハンチントン病のマウスモデルを使用した研究は、ラパマイシンによる治療が、ハンチントン凝集体のクリアランスを促進することを示す。MPLは、同様に、このような凝集体を除去することができ、この状態の新たな治療法となる。

・パーキンソン病:mTORC1阻害剤ラパマイシンは、L-DOPAの治療有効性に影響を及ぼすことなく、運動異常の発症を防止した。したがって、mTORC1シグナル伝達カスケードは、抗パーキンソン病療法を計画するための標的となる。
・線維症(肝臓、心臓および肺の線維症など)。線維芽細胞のMPL対する感受性、PI3K/mTORとTGFベータの間の関係およびリジル酸化酵素(LOX)発現/活性に対する効果のため、MPLは線維症の治療で使用することができる。心臓LOX発現の増加が、心不全/線維症の患者で認められ、同様に、いくつかの肺および腎疾患/LOX/線維芽細胞で認められる。さらに、線維症は、乳癌および膵臓癌などの数種類の癌において、重要な要因である。
本発明は、少なくとも1種の式(I)の化合物、または前記化合物の代謝物、薬学的に許容される塩、溶媒和化合物もしくはプロドラッグおよび少なくとも1種の薬学的に許容される担体を含む医薬組成物、医薬品およびキットを提供する。本発明によって記載された化合物から医薬組成物を調製するために、不活性な、薬学的に許容される担体は、固体または液体のいずれかであってもよい。固形調製物には、散剤、錠剤、分散可能な顆粒剤、カプセル剤、カシェ剤および座剤が含まれる。散剤および錠剤の約5から約95パーセントが、活性成分から構成されていてもよい。適切な固形担体は、当業界では公知であり、例えば、炭酸マグネシウム、ステアリン酸マグネシウム、タルク、糖または乳糖である。錠剤、散剤、カシェ剤およびカプセル剤は、経口投与に適した固形剤形として使用することができる。薬学的に許容される担体の例および様々な組成物の製造方法は、A. Gennaro(編)、Remington's Pharmaceutical Sciences、18版、(1990)、Mack Publishing Co.、Easton、Pennsylvaniaに見いだすことができる。
薬剤、組成物および医薬品は、これらに限定されないが、非経口(例えば、静脈内、脊髄内、皮下または筋肉内)、経口、局所または粘膜経路(例えば、鼻腔内)を含む標準的経路によって、レシピエントに投与することができる。いくつかの実施形態では、それらは、単独で、またはその他の追加的治療剤と組み合わせてレシピエントに投与してもよい。このような実施形態では、投与は、同時または逐次的であってもよい。
本発明の予防的および治療的方法は、任意の適切な対象に適用することができる。いくつかの実施形態では、対象は哺乳類対象である。例えば、対象は、マウス、ラット、イヌ、ネコ、ウシ、ヒツジ、ウマまたは任意のその他の社会的、経済的もしくは研究的に重要な哺乳類であってもよい。したがって、対象は、例えば、ヒトまたは非ヒト哺乳類などの哺乳類であってもよい。
細胞系
ヒト卵巣癌細胞系OVCAR-3、SKOV-3およびA2780ならびに初代細胞ヒト臍帯静脈内皮細胞(HUVEC)ならびにその他の全細胞系は、アメリカ培養細胞系統保存機関(ATCC)から入手し、それらの指示に従って維持した。星状膠細胞および神経膠腫細胞系は、Lowy Cancer Research Centre、University of New South Wales、AustraliaのDr. Kerry McDonaldから恵与された。星状膠細胞および神経膠腫細胞系は、Lowy Cancer Research Centre、University of New South Wales、AustraliaのDr. Kerry McDonaldから恵与された。
細胞増殖は、スルホローダミンB(SRB)アッセイを使用して評価した。96ウェルプレートに接種した細胞(2,000〜3,000細胞/ウェル)をMPL(0、1、5、10、25、50および100μmol/L)で72h処理した。次に、細胞を固定し、洗浄し、1%酢酸に溶解した0.4%(w/v)SRB100μlで染色した。1%酢酸で5回洗浄することによって、結合していない色素を除去してから、空気乾燥した。結合したSRBは、10mMトリス塩基(pH10.5)100μlで可溶化し、570nmでの吸光度を読み取った。全く同じ手法を使用してMPL-SO2を評価した。両薬剤をエタノールに溶解し、培地で希釈して、細胞培養アッセイに必要な最終濃度を得た。
生存能実験のために、6ウェルプレートに接種した細胞をモネパンテル(MPL)に0、1、10、50および100μMの濃度で24、48または72h曝露した。モネパンテル(Novartis社、Basel、Switzerlandから恵与された)を100%エタノールに溶解し、その後細胞培養培地で希釈した。処理期間の最後に、細胞をPBSで洗浄し、トリプシン処理し、トリパンブルーおよび血球計数計を使用して計数した。実験点はすべて4連で設定して、各実験を少なくとも2回行った。
コロニー形成アッセイのために、OVCAR-3またはA2780細胞などの5×106個の細胞を100mmのペトリ皿に入れ、一晩付着させた。培地を吸引除去し、対数増殖期の細胞を様々な濃度のMPLで72hインキュベートした。この時点で培地を吸引し、皿をPBSで洗浄し、薬物を含まない培地を各皿に添加した。培地を週に2回、3週間交換した。この後、プレートをPBSで丁寧に洗浄し、細胞を100%エタノールで固定し、濾過したクリスタルバイオレットの0.5%溶液で染色した。50個を上回る細胞からなるコロニーを倒立顕微鏡下で計数した。
MPLの細胞周期に対する効果は、標準的なフローサイトメトリー分析のプロトコールおよび手法を使用して測定した。簡単に説明すると、0.7×106個の細胞を25cm3フラスコに接種し、一晩付着させ、MPLで24または48h処理した。細胞をトリプシン処理して収集し、培地中に浮遊する細胞と共にプールした。細胞懸濁液を遠心して、PBSで洗浄し、メタノールで固定した。その後、細胞を洗浄し、PBS中のヨウ化プロピジウムおよびリボヌクレアーゼA中に30分間室温で再懸濁し、フローサイトメトリー(Becton Dickinson FACSort)によって分析した。
細胞におけるタンパク質発現は、ウェスタンブロット分析を使用して測定した。指示した濃度のMPLで処理した後、細胞溶解物を調製し、抗体でcdk2、cdk4、サイクリンA、サイクリンE、PARP-1(1:1000希釈;Cell Signalling Technology社)およびp53(1:200希釈;Santa Cruz Biotechnology社)について探索した。ゲル上にタンパク質が比較用にローディングされていることを、GAPDH抗体(1:30000希釈;Sigma-Aldrich社)でブロットを再度探索することによって確認した。
雌ヌードマウス(6週齢)は、生物資源部門(Faculty of Medicine、University of New South Wales)から購入した。マウスに実施された全手法は動物実験倫理会によって承認された。簡単に説明すると、2.5×106個の対数増殖期のOVCAR-3細胞を各マウスの左側腹部にs.c.注射した。動物の体重は週に1回測定し、一方、腫瘍体積は週に2回測定した。腫瘍増殖は、ノギスによる直交直径の測定によってモニターし、推定した腫瘍体積は、式1/2(長さ×幅2)(式中、幅は2つの直交測定値の短い方である)に基づいて計算した。動物実験倫理会の承認に基づいて、腫瘍体積が500mm3に達する前にマウスを安楽死させた。マウスを無作為化し、処理群または対照群(群当たり6匹)に割り当てて、処理は腫瘍細胞を注射して8日後に開始した。モネパンテルは、滅菌した0.5%(w/v)ヒドロペルオキシメチルセルロース(HPMC)に懸濁した。薬物は、週3回25または50mg/kgでi.p.投与した。対照群は、滅菌した媒体(0.5%HPMC)で処理した。マウスは、3週間の間処理した。最後の薬物投与の24時間後、マウスを安楽死させ、腫瘍を切除し、分析するまで-80℃で凍結した。
データをすべて、少なくとも2回の独立した実験による平均±標準誤差(S.E.M.)として報告する。MPL処理群対対照群の間の腫瘍体積の差は、ポストホックダネット検定による一元配置ANOVAを使用して解析した。定量的変数は、スチューデントt検定を使用して比較した。統計学的有意差は、P<0.05で規定された。
MPLは細胞増殖を阻害する
MPLの効果は、OVCAR-3、A2780およびSKOV-3の卵巣癌細胞系の増殖に対して調べた。SRBアッセイを使用することによって、MPLの細胞増殖に対する効果を調べた。MPLは、OVCAR-3、A2780およびSKOV-3細胞の増殖を濃度依存的に阻害し、Table 1(表1)によるとIC50値はそれぞれ、6.3、10.0および29.3であった。これらの結果から、卵巣癌細胞系は、MPLの抗増殖効果に対して感受性であることは明らかである。SKOV-3細胞は、感受性が最も低かった。SRB増殖アッセイを使用して、MPL-SO2も同様の方法で試験した。MPL-SO2は、MPLと同等の効力を有していることが見いだされた。MPL-SO2は、培養で増殖する癌細胞系の生存能を低下させ、細胞増殖を阻害した。MPL-SO2のIC50値をTable 1(表1)に示す。
MPLが増殖恒常性およびコロニーを確立する細胞系の能力も妨げるかどうかを調査するために、MPLに曝露した細胞のクローン形成活性を調査した。様々な濃度のMPLに72h曝露した後、細胞を洗浄し、次いで薬物を含まない培地で2週間インキュベートした。MPLは、これらの細胞によるコロニー形成を著しく妨げることが見いだされた。高い濃度のMPLでは、クローン形成能力のほとんど完全な喪失が引き起こされた(図2)。
MPLが細胞増殖およびコロニー形成を阻害する機構を調査するために、MPLの細胞周期に対する効果をフローサイトメトリーを用いて調べた。MPLが細胞周期進行を妨害することが見いだされた(図3)。MPLに曝露した細胞の進行は、濃度および時間に依存してG1期で停止した。G1期の細胞の蓄積には、SおよびG2-M期の細胞のパーセントの急低下が付随した。MPL誘導性細胞周期停止に関与する分子機構を研究するために、細胞周期調節タンパク質cdk2、cdk4、サイクリンAおよびEの発現を調べた。MPL処理細胞が発現するcdk2、cdk4、サイクリンAおよびEは低レベルであった(図4)。
MPLが細胞増殖およびコロニー形成を阻害する機構を見いだすために、MPLの細胞周期に対する効果をフローサイトメトリー(FACS)を用いて調べた。MPLが細胞周期進行を妨害することが見いだされた(図22)。MPLに曝露した細胞では、細胞周期は、濃度および時間に依存してG1期で停止した。G1期の細胞の蓄積には、SおよびG2-M期の細胞のパーセントの急低下が付随した。MPL誘導性細胞周期停止に関与する分子機構を研究するために、細胞周期調節タンパク質cdk2、cdk4、サイクリンAおよびEの発現を調べた。MPL処理細胞が発現するcdk2、cdk4、サイクリンEおよびサイクリンAは低レベルであった(図4および図16)。
MPL誘導性細胞死がPARPの切断に関与するかどうかを調査するために、MPL処理細胞の溶解物のPARP-1および切断PARP-1のウェスタンブロット分析を実施した。PARP-1の切断は、DNAの修正によって誘導される生存を防止することによって、アポトーシスを促進する。PARPは、細胞が生存能を維持するのを助け、したがって、PARPの切断は、細胞の解体を容易にし、アポトーシスを受ける細胞のマーカーとして役立つ。図5は、PARPは、MPL処理細胞において切断されたことを示している。
PARP切断の強い誘導が、細胞死を表すことを示すMPL処理OVCAR-3およびA2780細胞から調製した細胞溶解物のウェスタンブロット分析(図18)。
図19Aおよび図19Bに示したように、OVCAR-3またはA2780細胞のMPLによる処理は、細胞において見いだされるATPレベルの低下の原因となる。
図6は、細胞のMPLによる処理が、液胞の形成を引き起こすことを示しており、MPLがこれらの細胞において自己貪食を誘導し得ることを示唆している。図7は、MPL処理が細胞自己貪食のもう1つの指標であるADP/ATPの細胞比を低下させることを示している。
図20〜図22は、ヌードマウスにおける、MPLのインビボ試験を示している。OVCAR-3腫瘍を有するマウスを最初にi.p.で、または最後の実験のように経口的に処理した。得られた結果から、投与した用量、特に50mg/kg用量(i.p.および経口の両方)では、これらの動物の腫瘍成長を顕著に遅らせる活性が明らかである。腫瘍の組織学から、腫瘍細胞死の広範な領域が明らかになった(図23)。

6- 細胞のATPレベルを低下させる;
7- LC3B-IのLC3B-IIへの変換によって確認されるように、進行性の自己貪食を引き起こす;
8- 自己貪食は、卵巣癌細胞系および神経膠腫癌細胞系の両方において顕微鏡的に明らかであった;
9- MPLはまた、PARP-1の切断、したがって細胞死を誘導する;
10- これは、s.c.腫瘍を有するヌードマウスにおいて、腫瘍の用量依存的抑制を示すインビボにおけるデータによって確認される;
11- i.p.および経口の両方の投与経路が効果的であったことを示す。
さらに、MPLは、いくつかの標準的化学療法に対して抵抗性の細胞の増殖を阻害する。
本出願の別の態様は、癌および神経変性疾患における自己貪食の役割である。神経変性の原因となる異常に迅速なアポトーシスを起こす神経細胞において、自己貪食は、加速した細胞死から神経細胞を防御するための機構として役立つ。
神経変性疾患におけるmTOR/自己貪食
アルツハイマー病
Abetaおよびtauの蓄積は、アルツハイマー病に特徴的な進行性の認知障害の直接的な原因となるか、または寄与すると考えられている。mTORは、Abetaおよびtau誘導性神経変性において役割を担う可能性があることが示された。
ハンチントン病は、ポリグルタミン鎖伸長によって引き起こされる9種類の遺伝性神経変性障害の1つである。伸長したポリグルタミンタンパク質は、細胞内の凝集体中に異常に蓄積する。mTORは、細胞モデル、トランスジェニックマウスおよびヒト脳において、ポリグルタミン凝集体に隔離されることが示されている。mTORの隔離は、そのキナーゼ活性を弱め、突然変異ハンチンチン断片の重要なクリアランス経路である自己貪食を誘導する。これによってポリグルタミン毒性から防御し、特異的mTOR阻害剤ラパマイシンはハンチントン病の細胞モデルにおいてハンチンチン蓄積および細胞死を減衰させるので、自己貪食の阻害は逆の効果を有する。

哺乳類ラパマイシン標的であるmTORは、ラパマイシンの標的としてFKBP12、Tor1およびTor2を同定したサッカロマイセス・セレビシエ(saccharomyces cerevisiae)のラパマイシン抵抗性突然変異体の遺伝的および分子的研究によってTORが最初に発見され、FKBP12-ラパマイシン複合体がTor1およびTor2に結合し、それらの細胞機能を阻害することを強く支持する先例に基づいて命名された。

Xはヘテロ原子、N−アルキルまたはNHであり、
nは1から20である)
の化合物、またはその薬学的に許容される塩もしくは溶媒和化合物を含む組成物であって、
1種または複数のmTOR経路関連疾患が、神経変性疾患、加齢性疾患、移植拒絶反応に関連する疾患、慢性炎症性疾患、グリコーゲン蓄積に関連する疾患、全身性エリテマトーデス、炎症および免疫活性化に関連する疾患、貧血、白血球減少症、血小板減少症、腎不全、肥満、糖尿病/インスリン抵抗性、非アルコール性脂肪肝に関連する疾患、多発性嚢胞腎疾患、および線維症から選択される、組成物。
R3が-CF3またはハロゲンであり、
R4が-SCF3、-SOCF3、-SO2CF3、-OCF3または-CF3であり、
R5がHであり、
R6がアルキルであり、
XがOであり、
nが1から5である、請求項1に記載の組成物。
