Claims (11)
- 片側熱入力を用いた、回転に基づく分析方法のためのカートリッジ(1)であって、
マイクロ流体チャネル及びチャンバ構造(4)が形成され、複数の処理チャンバがチャネルによって相互接続されている、面状に拡がる基体(2)と、
前記分析方法を実行するための分析装置の支持プレート(34)に前記基体(2)を位置決め及び/又は固定するための、前記基体(2)に形成された複数の位置決め及び/又は固定要素(32)と、
熱入力側(8)と反対側の前記基体(2)の上側(16)に片側配置され、少なくとも1つのチャンバ(56)を覆う、前記基体(2)に固定されたカバー本体(14)と、を備えたカートリッジ(1)。 - 前記カバー本体(14)によって覆われた前記チャンバは、遺伝物質の複製のための増幅チャンバ(56)である、請求項1に記載のカートリッジ(1)。
- 前記カバー本体(14)は、前記基体(2)の前記上側(16)を少なくともほぼ完全に覆っている、請求項1又は2に記載のカートリッジ(1)。
- 前記カバー本体(14)は、前記基体(2)の前記上側(16)に向かって突出し、前記覆われたチャンバ(56)を囲むフレームウェブ(86)を有する、請求項1~3のいずれか一項に記載のカートリッジ(1)。
- 前記フレームウェブ(86)は、意図した使用状態において、前記基体(2)上に載っている、又は、前記基体(2)からわずかな距離をおいて、特に0.5ミリメートル未満、好ましくは0.1ミリメートル以下の距離をおいて、配置されている、請求項4に記載のカートリッジ(1)。
- 前記覆われたチャンバ(56)は、前記基体(2)の前記上側(16)において盛り上がって突出しており、前記フレームウェブ(86)と部分的に重なり合っている、請求項5に記載のカートリッジ(1)。
- 前記カバー本体(14)は、特に熱可塑性のプラスチックから形成されている、請求項1~6のいずれか一項に記載のカートリッジ(1)。
- 前記基体(2)は、その中に挿入されたチャネル及びチャンバ構造(4)を有する特に熱可塑性の基板と、前記チャネル及びチャンバ構造(4)をシールするシール被膜(6)とを含む、請求項1~7のいずれか一項に記載のカートリッジ(1)。
- 回転に基づく分析方法であって、
請求項1~8のいずれか一項に記載のカートリッジ(1)を分析装置の支持プレート(34)に固定し、
前記カートリッジ(1)を保持した状態において前記支持プレート(34)を回転させ、
それぞれの方法ステップに応じて、前記支持プレート(34)に配置された所定数の加熱要素(42)によって、片側において前記基体(2)に熱を導入し、
前記それぞれの方法ステップに応じて、前記回転の回転数を、20Hzまでの低回転数範囲、20~40Hzの中回転数範囲、及び40Hzからの高回転数範囲の間において変化させる、分析方法。 - 前記カバー本体(14)によって覆われた前記チャンバ(56)内においてポリメラーゼ連鎖反応を実行し、
サイクル的な熱入力によって、前記チャンバ(56)内の液体温度を、一方においては50~75℃に、他方においては80~100℃に設定し、且つ、中回転数から高回転数までを使用する、請求項9に記載の分析方法。 - カートリッジ(1)の使用方法であって、
請求項9又は10に記載の回転に基づく分析方法において請求項1~8のいずれか一項に記載のカートリッジ(1)を使用する使用方法。
